Clinical Significance of SH2B1 Adaptor Protein Expression in Non-small Cell Lung Cancer

The SH2B1 adaptor protein is recruited to multiple ligand-activated receptor tyrosine kinases that play important role in the physiologic and pathologic features of many cancers. The purpose of this study was to assess SH2B1 expression and to explore its contribution to the non-small cell lung cancer (NSCLC). Methods: SH2B1 expression in 114 primary NSCLC tissue specimens was analyzed by immunohistochemistry and correlated with clinicopathological parameters and patients' outcome. Additionally, 15 paired NSCLC background tissues, 5 NSCLC cell lines and a normal HBE cell line were evaluated for SH2B1 expression by RT-PCR and immunoblotting, immunofluorescence being applied for the cell lines. Results: SH2B1 was found to be overexpressed in NSCLC tissues and NSCLC cell lines. More importantly, high SH2B1 expression was significantly associated with tumor grade, tumor size, clinical stage, lymph node metastasis, and recurrence respectively. Survival analysis demonstrated that patients with high SH2B1 expression had both poorer disease-free survival and overall survival than other patients. Multivariate Cox regression analysis revealed that SH2B1 overexpression was an independent prognostic factor for patients with NSCLC. Conclusions: Our findings suggest that the SH2B1 protein may contribute to the malignant progression of NSCLC and could offer a novel prognostic indicator for patients with NSCLC.

The generalized discrete Fourier transform (GDFT) realizes the rapid extraction of specific order harmonics, and shortens the extraction time to 1/3 of a fundamental cycle (6.7 ms at 50 Hz grid) for 6r ± 1 order harmonics. However, the GDFT becomes less accurate when the system presents background harmonics other than specific-order harmonics in the inputs. Therefore, this paper analyzes the influence introduced by these background harmonics on the GDFT. First, the frequency feature of the GDFT is derived by a geometric method based on the z-domain unit circle. Then, the extraction of the 6r ± 1 order harmonics is taken as an example to show the extraction accuracy of the GDFT. The Accuracy of Harmonic Extraction (AHE) index, is also given to evaluate the accuracy of the extraction. Simulation and experimental results confirmed the correctness of the theoretical analysis by showing that the GDFT has the same accuracy as the RDFT when only 6r ± 1 order harmonics are involved. However, if there are other background harmonics in the inputs, the extraction accuracy is affected greatly. Therefore, when the background harmonics cannot be ignored, it is necessary to analyze the AHE of the GDFT. When AHE > 95%, the GDFT has good accuracy and is applicable to real applications.
A Hierarchical Model for Mobile Ad Hoc Network Performability Assessment

Dynamic topology is one of the main influence factors on network performability. However, it was always ignored by the traditional network performability assessment methods when analyzing large-scale mobile ad hoc networks (MANETs) because of the state explosion problem. In this paper, we address this problem from the perspective of complex network. A two-layer hierarchical modeling approach is proposed for MANETs performability assessment, which can take both the dynamic topology and multi-state nodes into consideration. The lower level is described by Markov reward chains (MRC) to capture the multiple states of the nodes. The upper level is modeled as a small-world network to capture the characteristic path length based on different mobility and propagation models. The hierarchical model can promote the MRC of nodes into a state matrix of the whole network, which can avoid the state explosion in large-scale networks assessment from the perspective of complex network. Through the contrast experiments with OPNET simulation based on specific cases, the method proposed in this paper shows satisfactory performance on accuracy and efficiency.
Development of monocular video deflectometer based on inclination sensors

The video deflectometer based on digital image correlation is a non-contacting optical measurement method which has become a useful tool for characterization of the vertical deflections of large structures. In this study, a novel imaging model has been established which considers the variations of pitch angles in the full image. The new model allows deflection measurement at a wide working distance with high accuracy. A monocular video deflectometer has been accordingly developed with an inclination sensor, which facilitates dynamic determination of the orientations and rotation of the optical axis of the camera. This layout has advantages over the video deflectometers based on theodolites with respect to convenience. Experiments have been presented to show the accuracy of the new imaging model and the performance of the monocular video deflectometer in outdoor applications. Finally, this equipment has been applied to the measurement of the vertical deflection of Yingwuzhou Yangtze River Bridge in real time at a distance of hundreds of meters. The results show good agreement with the embedded GPS outputs.

Objective: To investigate the effects of secondary left-sided portal hypertension (LSPH) on the radical operation rate of patients with pancreatic cancer and systemically evaluate the prognosis of patients with LSPH secondary to pancreatic cancer after radical surgery. Materials and Methods: The data of patients with pancreatic cancer who underwent laparotomy over a 15-year period in Department of Hepatobiliary Surgery of Chinese PLA Air Force General Hospital from Jan. 1, 1997, to Jun. 30, 2012 was retrospectively reviewed. Results: A total of 362 patients with pancreatic cancer after laparotomy were selected, including 73 with LSPH and 289 without LSPH. Thirty-five patients with LSPH (47.9%) and 147 without non-LSPH (50.9%) respectively underwent radical operations. No significant difference was found between these two groups regarding the total resection rate and stratified radical resection rate according to different pathological types and cancer locations. The mean and median survival time of patients after radical operation in LSPH group were $13.9{\pm}1.3$ months and 14.8 months, respectively, while those in non-LSPH group were $22.6{\pm}1.4$ months and 18.4 months, respectively(P<0.05). Conclusions: Radical operations for pancreatic cancer and secondary LSPH are safe and effective. Because high-grade malignancy and poor prognosis are closely associated, the decision for radical surgery should be made more meticulously for the patients with pancreatic cancer.

In this study, we report the cDNA cloning and sequence determination of Hh‐JHAMT from the seabuckthorn carpenterworm, Holcocerus hippophaecolus, by using rapid amplification of cDNA ends. The full‐length cDNA of putative Hh‐JHAMT was 1659 bp and contained a highly conserved Motif I, SAM motif I, which showed that Hh‐JHAMT like enzyme was a member of SAM‐dependent MTases. Moreover, putative Hh‐JHAMT had high homology to the other members of the JHAMT peptide family: 59% with Spodoptera litura, 54% with Bombyx mori and 54% with Helicoverpa armigera. Multiple alignments and phylogenetic analysis revealed that Hh‐JHAMT was closely related to JHAMT from Lepidoptera. Real‐time quantitative PCR experiments showed that Hh‐JHAMT mRNA expression was highest in the corpora allata (CA) complex, and was also detected at high levels during earlier larval and adult stages. The JHAMT mRNA level gradually declined during larval development, and the lowest amount of expression was observed in the pupal stage, while it increased to a higher level during adult stages. The pattern of Hh‐JHAMT expression was similar to the mode of JH biosynthesis. These results provided information concerning molecular characteristics of Hh‐JHAMT, whose expression profile suggests that the Hh‐JHAMT gene might be changed with larval development, metamorphosis and adult reproduction of the H. hippophaecolus.
