        Analysis of genetic diversity among indigenous landraces from sesame (Sesamum indicum L.) core collection in China as revealed by SRAP and SSR markers

        The molecular genetic diversity of 404 indigenous landraces from sesame core collection in China were evaluated by 11SRAP and 3 SSR markers, 175 fragments were generated, of which 126 were polymorphic with an average polymorphism rate of 72%. Jaccard’s genetic similarity coefficients (GS=0.7130), Nei's gene diversity (h=0.2418) and Shannon's Information index (I=0.3847) were calculated, a dendrogram of the 404 landraces was made, landraces from various zones were distributed throughout the dendrogram, accessions from different agro‐ecological zones were indistinguishable by cluster analysis, geographical separation did not generally result in greater genetic distance, a similar pattern was obtained using principal coordinates (PCO) analysis. As to seven agro‐ecological zones, the maximum Nei’s gene diversity (h = 0.2613)and Shannon index (I = 0.3980) values in zone VII indicated that they were genetically more diverse than those in other zones, while the least genetically diverse region was zone III (h = 0.1772, I = 0.2858). Nei's genetic identity and genetic distance among landraces from seven agro‐ecological zones were also analyzed, the genetic relationship of seven zones was inferred using the UPGMA method. This study demonstrated that SRAP and SSR markers were appropriate for evaluation of sesame genetic diversities. There existed extensive genetic diverse among indigenous landraces and the abundance of genetic diversity of landraces in different agro‐ecological zones was various. Understanding of these characteristics of indigenous landraces in China can provide theoretical foundation for further collection, effective protection and reasonable utilization of these sesame landraces in breeding.

        Effects on Range of motion of Neck joint through Five-Animals Micro-Exercises

        This research was aiming to provide new-styles sport exercises for white and blue-collar workers and prevented people from suffering occupational neck joint illnesses, released the pains from joints of neck and improving the ranges of motions of neck joints throughout the observations that was investigated among staffs who were from a research institute. This research carried out the suggested intervention called Five-Animals Micro-Exercises and then observe its effects that indexes which reflected the flexion, extension, lateral flexion, rotation and pain degree of joint neck would change. There were 320 staffs were chosen from all working staffs who employed by the institute as individual subjects. These 320 staffs did the Five-Animal Micro-Exercise and lasted for 20 weeks during the intervention period. In the progress of intervention, all subjects were divided into groups according to their working locations and organized to do the exercise which lasted for 10 to 15 minutes then exercise frequency was required at least 2 times a day and no more less than 5 days a week. Groups leaders would be elected among the groups and needed to record sport situations of group members. All group members could decide to join the grouped exercise or did the sport alone when they are convenient. The results of intervention would be collected when beginning and ending of this experiment, which is in order to determine the indexes flexion, extension, lateroflexion, rotation, pain degree of joint neck and body attitude assessment. Moreover, 177 subjects who showed higher compliance and reached the required standard of exercising were selected from weekly sport situations subjects’ feedback. The neck joint range of motion had significant changes after intervention, include flexion, extension, lateroflexion, and rotation(p<.05).Besides,, the pain degree and points of body attitude assessment of subjects stated dramatically variation (p<.05).The Five-Animal Micro-Exercise contributes on enlarging the degrees of flexion, extension, lateroflexion, rotation, and pain degree of joint neck are is helpful on improving joint neck degree. It can also relief joint pain, relax muscle, upraises endurance of muscles and increases the coordination of body, which are good for preventing cervical spondylopathy, and help people to maintain their body cures.

        A Rapidly New-typed Detection of Norovirus Based on F0F1-ATPase Molecular Motor Biosensor

        In order to adapt port rapid detection of food borne norovirus, presently we developed a new typed detection method based on F0F1-ATPase molecular motor biosensor. A specific probe was encompassed the conservative region of norovirus and F0F1-ATPase within chromatophore was constructed as a molecular motor biosensor through the “ε-subunit antibody-streptomycinbiotin- probe” system. Norovirus was captured based on probe-RNA specific binding. Our results demonstrated that the Limit of Quantification (LOQ) is 0.005 ng/mL for NV RNA and also demonstrated that this method possesses specificity and none cross-reaction for food borne virus. What’s more, the experiment used this method could be accomplished in 1 h. We detected 10 samples by using this method and the results were consistent with RT-PCR results. Overall, based on F0F1-ATPase molecular motors biosensor system we firstly established a new typed detection method for norovirus detection and demonstrated that this method is sensitive and specific and can be used in the rapid detection for food borne virus.

        수상레저스포츠 참여자의 안전관리의식이 안전추구행동에 미치는 영향 분석

        This study investigated the relationship between the safety management awareness and safety behavior for the safe and continuous participation of water sports participants. The conclusions based on the derived results are as follows. First, among safety management consciousness, safety management education was confirmed to have a significant effect on safety behavior, and safety education through continuous and various channels is considered an essential element for safety behavior. Second, among the sub-factors of safety management awareness, safety compliance have a statistically significant influence on safety behavior, requiring continuous management and update of related rules and regulations. Third, it was confirmed that the risk treatment method among safety management consciousness influenced safety behavior. It is necessary to review an integrated reporting system for convenient reporting and prompt processing. Fourth, it was confirmed that the safety understanding of safety management consciousness did not affect safety behavior, but it was judged that the measurement items consisted of negative perceptions, so a closer review is needed in subsequent studies.

        Protective Effects of Verapamil against H2O2-Induced Apoptosis in Human Lens Epithelial Cells

        Verapamil is used in the treatment of hypertension, angina pectoris, and atrial fibrillation. Recently, several studies have demonstrated that verapamil increased the optic nerve head blood flow and improved the retrobulbar circulation. All these show that verapamil is potentially useful for ophthalmic treatment. Thus, the aim of this study is to investigate whether verapamil could protect human lens epithelial cell (HLEC) from oxidative stress induced by H2O2 and the cellular mechanism underlying this protective function. The viability of HLEC was determined by the MTT assay and apoptotic cell death was analyzed by Hoechst 33258 staining. Moreover, Caspase-3 expression was detected by immunocytochemistry and flow cytometry analysis. We also detected Caspase-3 mRNA expression by reverse-transcription-polymerase chain reaction and the GSH content in cell culture. The results showed that oxidative stress produced significant cell apoptotic death and it was reduced by previous treatment with the verapamil. Verapamil was effective in reducing HLEC death mainly through reducing the expression level of apoptosis-related proteins, caspase-3, and increasing glutathione content. Therefore, it was suggested that verapamil was effective in reducing HLEC apoptosis induced by H2O2.

      • A Novel Modulation Strategy for Diode-assisted Buck-boost VSI with Maximum Voltage Gain and Minimum Switching Frequency

        Diode-assisted buck-boost voltage source inverter (VSI) boosts the dc source voltage by introducing a diode-capacitor network. With capacitive charging in parallel and discharging in series, it extends voltage regulation range and avoids extreme boost duty ratio. As for the unique structure, various pulse width modulation (PWM) strategies are provided with regarding to the chopped intermediate dc-link voltage. In order to maximize voltage gain, as well as to achieve the increased efficiency, this paper proposed a novel modulation strategy by reducing the switching frequency of power devices in the inverter bridge. The operation principle and relationship of switching device voltage stress versus voltage gain are analyzed in detail. Simulation and experiments are implemented to verify the theoretical analysis. Compared with existing modulation strategies, diode-assisted buck? boost VSI with new modulation scheme demonstrates good performances: less power device requirement and higher efficiency in high voltage gain application. Owing to improved performance, it is a more competitive topology for wide range dc/ac voltage regulation in renewable energy application, especially for ac output with relatively high fundamental frequency. Furthermore, the idea of proposed modulation scheme can also be extended to the control of other quasi two-stage inverter.

        Functional roles of Tryptophan residues in diketoreductase from Acinetobacter baylyi

        Diketoreductase (DKR) from Acinetobacter baylyi contains two tryptophan residues at positions 149 and 222. Trp-149 and Trp-222 are located along the entry path of substrate into active site and at the dimer interface of DKR, respectively. Single and double substitutions of these positions were generated to probe the roles of tryptophan residues. After replacing Trp with Ala and Phe, biochemical and biophysical characteristics of the mutants were thoroughly investigated. Enzyme activity and substrate binding affinity of W149A and W149F were remarkably decreased, suggesting that Trp-149 regulates the position of substrate at the binding site. Meanwhile, enzyme activity of W222F was increased by 1.7-fold while W222A was completely inactive. In addition to lower thermostability of Trp-222 mutants, molecular modeling of the mutants revealed that Trp-222 is vital to protein folding and dimerization of the enzyme.       • An Improved Control Strategy for Z-source Inverter with Maximum Boost Capability and Minimum Switching Frequency

        Z-source inverter introduces a unique Xshaped impedance network and boosts the input voltage by utilizing shoot-through (ST) states. It provides a competitive single-stage approach for DC to AC power conversion in relatively low voltage boost ratio range (1?2). This paper proposes a novel control strategy for Z-source inverter. With this strategy, only one phase leg operates at high switching frequency to achieve both ST duty ratio control and output voltage regulation, while the other two phase legs maintain the fixed switching states during one sextant. Therefore, it reduces the equivalent switching frequency of power devices in the inverter bridge to about 1/3. However, it introduces the low frequency current and voltage ripple in the inductor and capacitor which related to the fundamental frequency of output voltage. It has the potential to achieve higher efficiency especially in high-switching-frequency and medium-frequency DC-AC power application. Then, the paper provides a detailed close-loop controller design approach for the novel modulation strategy. Furthermore, the detailed comparisons among the existing strategies and proposed control strategy are conducted in theory. Finally, all the theoretical analysis and calculation are verified by simulation results.

        Analysis of Cracking Development and Mechanical Characteristics of High-Filled Cut-and-Cover Tunnel

        The cut-and-cover tunnel (CCT) construction scheme has led to the popularity of high-filled cut-and-cover tunnel (HFCCT). HFCCT possesses a backfilling height of 30 − 50 m. However, such a high soil column produces a large force on the structure and the structure is prone to crack, causing structural damage and difficulty in normal use. In order to provide a reference for the structural safety assessment of HFCCT, we conducted a similarity model test to study the variation law of the development of crack, displacement, and internal force of HFCCT with a coefficient of λ (the ratio of the backfilling height to the height of the cut-and-cover tunnel). In the test, a 1:20 similar scale CCT model with slope angle 70° and groove width ratio 1 was built using data from the HFCCT on the Lanyu Line. The backfilling process on site was simulated in a simulation box (360 cm long, 120 cm wide and 209 cm high). A numerical model was also built using PFC2D to verify the accuracy of the model test and further explain the crack formation mechanism of the HFCCT from a microscopic perspective. The results show that with the increase of the coefficient of λ, the bearing stage of the CCT can be roughly divided into three stages: steady growth stage, rapid growth stage, and accelerated growth stage. Furthermore, the development of cracks is closely related to the displacement and internal force of the CCT. The larger the displacement of the CCT, the greater the instability of the structure and the larger the ratio of the bending moment to the internal force, which causes the crack to develop faster.
