        Concentrations of Progesterone and Estradiol in Peripheral Plasma during the Estrous Cycle and after Ovariectomy in Huanghuai Goats of High or Poor Prolificacy

        The objective of this study was to characterize the litter sizes in Huanghuai goats with high prolificacy (HP, five or more kids born per litter on at least one occasion), and to compare their peripheral blood concentrations of progesterone and estradiol with those of goats with poor prolificacy (PP, up to three kids born per litter on any occasion). The circulating concentrations of progesterone and estradiol were measured by radioimmunoassay from daily blood samples taken during natural estrus cycles and at 1-5 days after ovariectomy. Estrus was synchronized using two doses of a prostaglandin analog. Litter size for the HP goats increased up to a parity of five and decreased thereafter. The percentage of goats with litter sizes of $\geq$4 from parities 3 to 6 ranged from 44.5% to 58.3%. Although small differences in litter size were obtained for goats of parities three, four and six relative to five, parity five does had the highest mean litter size. Progesterone concentrations began to rise earlier and were higher in the HP than in the PP goats on most days of the luteal phase, but not during the follicular phase of the cycle or after ovariectomy. There was a significant difference between the two groups (p<0.05) in the magnitude of the progesterone plateau. Mean estradiol concentrations in the HP group remained significantly higher than in the PP group (p<0.05) during the estrus cycle. There were two estradiol peaks in the HP goats during the early luteal phase, but only one in the PP group. Measurements of individual corpora lutea (CL) in vitro showed that there was a greater prevalence of small CL (<6 mm in diameter) in the HP group than in the PP group (p<0.05). After ovariectomy, the estradiol level on day 1 was significantly higher than at the nadir during the estrus cycle in both the HP (p<0.01) and PP (p<0.05) goats, while levels decreased by 12.3% and 26.2% respectively compared with the mid-luteal period in HP and PP goats (p<0.05). The overall mean estradiol concentrations in HP goats were lower than in the PP group, but no significant differences were found between groups at 1-5 days after ovariectomy.

        Distinct regulation patterns of the two prophenoloxidase activating enzymes corresponding to bacteria challenge and their compensatory over expression feature in white shrimp (Litopenaeus vannamei)

        Prophenoloxidase activating enzyme 2 (PPAE2), which belongs to the second PPAE family of prawns, was isolated from white shrimp Litopenaeus vannamei. The currently identified lvPPAE2 and lvPPAE1 from our former report were taken as model candidates to analyze the relationship of the two shrimp PPAE families as well as the regulation mechanism of shrimp PPAEs. The tissue expression of lvPPAE2 was more ubiquitous than lvPPAE1. The mRNA abundance of lvPPAE2 was about 10 percent of lvPPAE1 in co-existed tissues. When challenged with Vibrio harveyi. LvPPAE2 showed a distinct transcriptional regulation pattern compared to lvPPAE1. Silence of lvPPAE2 significantly increased shrimp's susceptibility to V. harveyi, suggesting the lvPPAE2 plays essential role in shrimp host defense. A novel PPAE specific compensatory over expression feature was found in the two lvPPAEs. Single gene specific silence of lvPPAE1 and lvPPAE2 resulted in a significant difference in reduction of hemolymph PO activity. Double silence of the two lvPPAEs failed to cause a further reduction on PO activity or shrimp mortality to bacteria, despite that double silence sufficiently suppressed both of the two lvPPAEs. Our findings suggest both lvPPAEs contribute to shrimp melanization cascade and host defense against bacteria. Distinct regulation pattern corresponding to the same pathogen invasion suggests the two lvPPAEs are actually under different regulation ways. A novel PPAE specific compensatory over expression mechanism found in our study offered us a clue in understanding the robustness of shrimp innate immunity and network of crustacean proPO activating system.

        Since the global warming, the hot and humid climate becomes a great challenge for the production and health of sow during summer. The conventional cooling systems consumes a lot of energy by cooling the whole building, so a water-cooled overhead sow cover system that sims to enhance the heat loss of the sow without elevating the indoor relative humidity(RH) was designed and optimized using CFD to simulate the cooling effect and evaluate the impact parameters. A 3D CFD model within enclosed space have been developed and analyzed under unsteady state condition comparing the k-ε turbulence models such as the standard, RNG, and realizable models to validate the field experimental data. In addition, the structure of the cooling system by changing the tube spacing(10cm, 15cm, 20cm) and the curtain(W or W/O) in front of the sow occupied zone, the thermal condition of the ambient air and inlet water temperature combined with water flow was optimized during the simulation. The RNG k-ε model was found to be more suitable for this model with the lowest error of 1% at 10cm tube spacing by the validating experimental data of vertical temperature gradient within the cooled cover. The more dense tube spacing could increase the cooling effect by larger heat transfer area. Adding the curtain could not only reduce the air velocity across the sow occupied zone, but also the heat transfer with ambient air. The ambient air and inlet water temperature and water flow rate also affects the cooling efficiency. The optimized model according to the production system will be considered in the future study.

      • Self-Shielding Magnetized vs. Shaped Parallel-Magnetized PM Brushless AC Motors

        The performance of two designs of permanent magnet brushless motor, by having self-shielding magnetized magnets or sinusoidally shaped parallel-magnetized magnets with essentially sinusoidal airgap flux distributions, are compared. It is shown that the parallel-magnetized motor with shaped sintered NdFeB magnets can result in a higher airgap flux density and torque density than that of a self-shielding magnetized motor equipped with an anisotropic injection moulded NdFeB ring magnet.

        TEM investigation of hydrogen-implanted and annealed single-crystal SrTiO3

        In this study, the structural properties of single-crystal SrTiO3 implanted with H+ have been investigated by transmission electronmicrocopy (TEM). The investigation was carried out on an as-implanted sample and samples after annealing at 500.C and 700 .Cfrom the implanted energy similar to the process in the case of silicon. The as-implanted sample shows large out-of-plane strain alsosimilar to the report in silicon. The microcavities are lled with amorphous material as the result of complicated interaction of the defectsduring the annealing.

      • Coronary Atherosclerosis T<sub>1</sub>-Weighed Characterization With Integrated Anatomical Reference

        Objectives: The aim of this work is the development of coronary atherosclerosis T<SUB>1</SUB>-weighted characterization with integrated anatomical reference (CATCH) technique and the validation by comparison with high-risk plaque features (HRPF) observed on intracoronary optical coherence tomography (OCT) and invasive coronary angiography. Background: T<SUB>1</SUB>-weighted cardiac magnetic resonance with or without contrast media has been used for characterizing coronary atherosclerosis showing promising prognostic value. Several limitations include: 1) coverage is limited to proximal coronary segments; 2) spatial resolution is low and often anisotropic; and 3) a separate magnetic resonance angiography acquisition is needed to localize lesions. Methods: CATCH acquired dark-blood T<SUB>1</SUB>-weighted images and bright-blood anatomical reference images in an interleaved fashion. Retrospective motion correction with 100% respiratory gating efficiency was achieved. Reference control subjects (n = 13) completed both pre- and post-contrast scans. Stable angina patients (n = 30) completed pre-contrast scans, among whom 26 eligible patients also completed post-contrast scans. After cardiac magnetic resonance, eligible patients (n = 22) underwent invasive coronary angiography and OCT for the interrogation of coronary atherosclerosis. OCT images were assessed and scored for HRPF (lipid-richness, macrophages, cholesterol crystals, and microvessels) by 2 experienced analysts blinded to magnetic resonance results. Results: Per-subject analysis showed none of the 13 reference control subjects had coronary hyperintensive plaques (CHIP) in either pre-contrast or post-contrast CATCH. Five patients had CHIP on pre-contrast CATCH and 5 patients had CHIP on post-contrast CATCH. Patients with CHIP had greater lipid abnormality than those without. Per-segment analysis showed elevated pre- and post-contrast plaque to myocardium signal ratio in the lesions with HRPF versus those without. Positive correlation was observed between plaque to myocardium signal ratio and OCT HRPF scoring. CHIP on pre-contrast CATCH were associated with significantly higher stenosis level than non-CHIP on invasive coronary angiography. Conclusions: CATCH provided accelerated whole heart coronary plaque characterization with simultaneously acquired anatomical reference. CHIP detected by CATCH showed positive association with high-risk plaque features on invasive imaging studies.

        Effect of diets with different energy and lipase levels on performance, digestibility and carcass trait in broilers

        Objective: A 28-d trial was conducted to evaluate the effect of diets with different energy and lipase levels on performance, nutrient digestibility, serum profiles, gut health, and carcass quality in broilers. Methods: A total of 720 one-day-old male Ross 308 broilers ($45.4{\pm}0.5g$) were randomly assigned to one of the following four treatments: i) RET, reduced energy treatment (metabolizable energy = 2,950 and 3,100 kcal/kg for starter and finisher diet), ii) BDT, basal diet treatment (metabolizable energy = 3,050 and 3,200 kcal/kg for starter and finisher diet, iii) RET015, RET+0.15 g/kg lipase, and iv) RET03, RET+0.3 g/kg lipase. There were 10 replications (cages) per treatment with 18 birds per cage. Results: During d 1 to 14, broilers fed BDT, RET015, and RET03 diets had higher (p<0.05) body weight gain than those fed RET diet. During d 1 to 14, 15 to 28 and the overall experiment, feed conversion ratio in RET03 treatment was lower (p<0.05) compared with RET treatment. On d 14, the apparent total tract digestibility (ATTD) of dry matter (DM), ether extract (EE), and gross energy in RET03 treatment was higher (p<0.05) than those in RET treatment, while the ATTD of N was increased (p<0.05) by RET03 treatment. On d 28, broilers fed RET03 diet had higher (p<0.05) ATTD of DM than those fed RET and RET015 diets, while the ATTD of EE in BDT and RET03 treatments was increased (p<0.05) compared with RET and RET015 treatments. Broilers fed RET03 diet had higher villus height (VH) and VH:crypt depth (CD) ratio than those fed RET and BDT diets. The activity of pancreatic lipase in BDT and RET03 treatments was higher (p<0.05) than that in RET treatment. Conclusion: Taken together, lipase supplementation (3,000 U/kg feed) increased growth performance, nutrient digestibility, VH, VH:CD ratio and lipase activity, but decreased triglyceride, low-density lipoprotein cholesterol and the abdominal fat percentage in broilers fed reduced energy diet.

        Location determining method of critical sliding surface of fillings in a karst cave of tunnel

        A location determining method is proposed for critical sliding surface in the stability analysis of the filling materials in karst caves. First, a preliminary location of the sliding surface is determined based on simulation results which includes displacement contour and plastic zone. The sliding surface will locate on the bottom contact interface when the friction angle is relative small. However, a weakened contact interface always becomes the critical sliding surface no matter what the friction angle is. Then when the friction angle becomes larger, the critical sliding surface inside fillings can be determined by a parabola, the coefficient of which increases linearly with the friction angle under the same cohesion. Finally, the critical sliding surface approximately remains unchanged with friction angle. The influence of cohesion is similar to that of friction angle. Although affected by shape, size or position of the karst cave, the critical sliding surface mainly depends on both friction angle and cohesion. Thus, this method is always useful in determining the critical sliding surface.
