      • From single-molecule detection to next-generation sequencing: microfluidic droplets for high-throughput nucleic acid analysis

        <P>Droplet-based microfluidic technologies have proved themselves to be of significant utility in the performance of high-throughput chemical and biological experiments. By encapsulating and isolating reagents within femtoliter-nanoliter droplet, millions of (bio) chemical reactions can be processed in a parallel fashion and on ultra-short timescales. Recent applications of such technologies to genetic analysis have suggested significant utility in low-cost, efficient and rapid workflows for DNA amplification, rare mutation detection, antibody screening and next-generation sequencing. To this end, we describe and highlight some of the most interesting recent developments and applications of droplet-based microfluidics in the broad area of nucleic acid analysis. In addition, we also present a cursory description of some of the most essential functional components, which allow the creation of integrated and complex workflows based on flowing streams of droplets.</P>

        “V-junction”: a novel structure for high-speed generation of bespoke droplet flows

        <P>We present the use of microfluidic “V-junctions” as a droplet generation strategy that incorporates enhanced performance characteristics when compared to more traditional “T-junction” formats. This includes the ability to generate target-sized droplets from the very first one, efficient switching between multiple input samples, the production of a wide range of droplet sizes (and size gradients) and the facile generation of droplets with residence time gradients. Additionally, the use of V-junction droplet generators enables the suspension and subsequent resumption of droplet flows at times defined by the user. The high degree of operational flexibility allows a wide range of droplet sizes, payloads, spacings and generation frequencies to be obtained, which in turn provides for an enhanced design space for droplet-based experimentation.         Polymorphism, Expression of Natural Resistance-associated Macrophage Protein 1 Encoding Gene (NRAMP1) and Its Association with Immune Traits in Pigs

        Natural resistance-associated macrophage protein 1 encoding gene (NRAMP1) plays an important role in immune response against intracellular pathogens. To evaluate the effects of NRAMP1 gene on immune capacity in pigs, tissue expression of NRAMP1 mRNA was observed by real time quantitative polymerase chain reaction (PCR), and the results revealed NRAMP1 expressed widely in nine tissues. One single nucleotide polymorphism (SNP) (ENSSSCG00000025058: g.130 C>T) in exon1 and one SNP (ENSSSCG00000025058: g.657 A>G) in intron1 region of porcine NRAMP1 gene were demonstrated by DNA sequencing and PCR-RFLP analysis. A further analysis of SNP genotypes associated with immune traits including contain of white blood cell (WBC), granulocyte, lymphocyte, monocyte (MO), rate of cytotoxin in monocyte (MC) and $CD4^-CD8^+$ T lymphocyte subpopulations in blood was carried out in four pig populations including Large White and three Chinese indigenous breeds (Wannan Black, Huai pig and Wei pig). The results showed that the SNP (ENSSSCG00000025058: g.130 C>T) was significantly associated with level of WBC % (p = 0.031), MO% (p = 0.024), MC% (p = 0.013) and $CD4^-CD8^+$ T lymphocyte (p = 0.023). The other SNP (ENSSSCG00000025058: g.657 A>G) was significantly associated with the level of MO% (p = 0.012), MC% (p = 0.019) and $CD4^-CD8^+$ T lymphocyte (p = 0.037). These results indicate that the NRAMP1 gene can be regarded as a molecular marker for genetic selection of disease susceptibility in pig breeding.

        공동체 미학의 애니메이션 캐릭터 창조에 대한 응용 연구 : 중국 애니메이션 영화 <강자아> 사례를 중심으로

        본 연구의 목적은 공동체 미학이 애니메이션 캐릭터 창조에 어떻게 적용되는지를 탐구하는 것으로, 중국 애니메이션 영화 <강자아>를 사례로 삼았다. 본 논문은 텍스트 연구 방법을 사용하여 <강자아> 내의 캐릭터와 줄거리를 심층 분석함으로써 공동체 미학의 구체적인 적용을 탐구한다. 이에는 공동체 미학의 기본 개념과 이론 틀 소개는 물론 <강자아> 내 주요 캐릭터의 감정 공명과 연결에 대한 심층 분석을 포함한다. 공동체 미학이 문화 전달 분야에서의 중요성을 설명하고, <강자아>의 사례분석을 통해 공동체 요소가 캐릭터 형성과 스토리 전개에 미치는 영향을 보여준다. 연구 결과는 공동체 미학이 애니메이션 캐릭터 창조에 중요한 적용 가치를 가지며 <강자아>는 공동체 미학의 개념을 성공적으로 활용했음을 보여준다. 이는 중국 애니메이션 영화의 창작을 풍부하게 하고 관객에게 깊은 감정 체험과 문화적 성찰을 제공한다. 그것은 중국 애니메이션 영화의 발전에 유익한 시사점과 참고를 제공하며, 캐릭터의 감정적 깊이와 복잡성을 통해 관객이 감정 공명을 느끼고 영화의 줄거리에서 공동체의 힘을 체감할 수 있도록 한다. 공동체 미학의 적용은 관객의 감정적 체험과 문화적 이해를 깊게 하는 데 도움이 된다. The purpose of this study is to explore how communitarian aesthetics are applied to the creation of animation characters, using the Chinese animated film < Jiang Ziya > as a case study. This thesis employs textual research methods to conduct an in-depth analysis of the characters and plot within < Jiang Ziya > in order to investigate the specific application of communitarian aesthetics. This includes an introduction to the basic concepts and theoretical framework of communitarian aesthetics as well as a detailed analysis of the emotional resonance and connections of the main characters within < Jiang Ziya >. It elucidates the importance of communitarian aesthetics in the field of cultural transmission and demonstrates, how community elements affect character formation and story development. The findings indicate that communitarian aesthetics have significant applicative value in the creation of animated characters and that < Jiang Ziya > has successfully utilized the concepts of communitarian aesthetics. It offers valuable insights and references for the development of Chinese animated films, suggesting that through the emotional depth and complexity of the characters, the audience can experience emotional resonance and feel the power of the community within the film's narrative.

        Bombyx mori transferrin: Genomic structure, expression and antimicrobial activity of recombinant protein

        Transferrin (Tf) is a multifunctional, iron binding protein found in both vertebrates and invertebrates. Although transferrin has been suggested to play a role in innate immunity, its immunological function during infection has not been characterized. In this study, we identified and characterized Bombyx mori transferrin (BmTf). The promoter region of BmTf has numerous putative NF-κB binding sites, suggesting its possible function in innate immunity. Analysis of BmTf gene expression shows that it is highly inducible in response to a wide variety of pathogens including bacteria, fungus, and viruses. Recombinant BmTf protein produced in a baculovirus system exhibits iron binding capacity and antibacterial activity against various Gram-positive and -negative bacteria. Taken together, our results indicate that BmTf is an inducible immune effector molecule that may play an important role in pathogen clearance of insect innate immunity.

      • Prognostic Value of MAC30 Expression in Human Pure Squamous Cell Carcinomas of the Lung

        Recent evidence haas indicated that meningioma-associate protein (MAC30) exhibits different expression patterns in various tumors. However, little is known about the value of MAC30 in human squamous cell carcinoma of lung (SQCLC). The purpose of our study was to investigate the expression of MAC30 and to explore its clinical significance in SQCLC patients. A total of 156 Chinese patients diagnosed with SQCLC were selected for this study. The expression of MAC30 in all tissues was confirmed by immunohistochemical staining. Quantitative real-time PCR was performed to analyze MAC30 mRNA expression in 32 cases of SQCLC patients with corresponding non-tumor lung tissues. We observed enhanced mRNA expression of MAC30 in SQCLC as compared to control samples. Further, elevated MAC30 protein expression was strongly associated with poor tumor differentiation, TNM stage, and lymph node metastasis. In addition, we observed that patients with increased MAC30 expression demonstrated poor overall survival. Multivariate analysis explicated that increased MAC30 expression was a valuable independent predictable factor for poor tumor differentiation and short survival in SQCLC patients. Our present study suggests that MAC30 may serve as a biomarker for poor tumor differentiation and outcomes of patients with SQCLC.
