Object Detection Based on Two Level Fast Matching

Shape template matching is an important approach in object detection and recognition. In this paper, we propose a fast and novel object detection method, which represents edge map contours with salient points and retrieves the target object by using a backtracking method with two stages from coarse matching to fine matching. Our method has two main contributions. One is we propose the way to represent contour structure with salient points. The other is that the object detection method proposed can directly operate on the real images, which improves its practicability. According to the experimental results, the proposed approach improves the speed than previous works.
A Fast and Practical Contour Completion Approach with Chord-to-Point Distance

As a kind of critical information in images and computer vision tasks, contour has a general application due to the robustness to illumination. However, it is far from meeting the current requirement because of expensive computation and noise in the real-world application. This paper proposes a novel approach of extracting contours in images. Firstly, we can obtain the salient points on the target contour by use of the accuracy property of Chord-to-point distance under combination of an ellipse model method. Then, a hyperbolic curve segment is used to fit the salient points, which can represent the target contour. The extensive experiments show that our method has better robustness and gives more exact approximation to the original target contour. Our work provides more selections for the practical application of contour completion.
Object Detection by Matching Salient Line Segments

Shape template matching is an important approach in object detection and recognition. In this paper, we propose a fast and novel method to represent edge maps by using salient line segments, which is used to detect objects based on shape template matching. Firstly, image edges are computed and, on these image edge fragments, corner points are extracted. Then, a parabola model is proposed to represent image edges. Secondly, based on salient points, a directional chamfer matching framework is used to compute the similarity between template image and corresponding locations in target image. Our method has two main contributions. One is we apply the way to represent contours with salient points to the framework, which is used to detect the target rapidly and accurately. The other is that our method can directly operate on real images, which improves its practicability. A series of experiments are performed on two benchmark data sets. Comparing with previous work, our method is much more time-saving and memory-saving.
Unconstrained Face Verification by Optimally Organizing Multiple Classifiers

Face verification is still a difficult problem in badly uncontrolled conditions. Motivated by the observation that failure of facial image pair-matching is always caused by inconsistent visual con-ditions of both total face and face parts, we propose to train multiple classifiers on total face and face parts with respect to different visual consistency conditions. For training these classifiers, multiple similarity measures of multiple features on the matched and mismatched face image pairs are consid-ered as the positive and negative training samples respectively. For a new face image pair, we compute its similarity measures and visual consistency measures (VCM). Relying on these VCMs, we adap-tively select VCM-specific classifiers and optimally organize them to form an ensemble by the reliabil-ity estimation and adaptively weighting combination. Our contributions are the visual consistency measure and the optimally organized VCM-specific classifier ensemble which can better overcome the influence of uncontrolled factors such as pose, illumination and expression etc. Experiments on the challenging unconstrained face dataset LFW (Labeled Face in the Wild) confirm the good performance of our algorithm with the record 0.8927 compared with state-of-the-art approaches in the same experiment settings.

Double stranded targets on the cDNA microarray contain representatives of both the coding and noncoding strands, which will introduce hybridization competition with probes. Here, the effect of double and single strands of targets on the signal intensity and the ratios of Cy5/Cy3 within the same slide were compared. The results show that single stranded targets can increase the hybridization efficiency without changing the Cy5/Cy3 ratio. Based on these results, a new strategy was established by generating cDNA targets with asymmetric PCR, instead of conventional PCR, to increase the sensitivity of the cDNA microarray. Furthermore, the feasibility of this approach was validated. The results indicate that the cDNA microarray system based on asymmetric PCR is more sensitive, with no decrease in the reliability and reproducibility as compared with that based on conventional symmetric PCR.
Gene expression profile of NFκB repressing factor(NKRF) knockdown cells by microarray analysis

Human NFκB repressing factor(NKRF) is a negative regulation transcription factor, which is able to repress transcription by binding to the negative regulatory element(NRE) near the NFκB binding site in certain genes’ promoters. Current researches reveals that NKRF represses the activation of IFN-β, IL-8,hiNOS and HIV-1 by NFκB. We used optical fiber beadchip to analysis the different gene expression patterns of RNAi mediated NKRF knockdown HEK293 cells and found that several genes showed significant change of expression levels. Real-time PCR was performed to verify the changes of expression of candidate genes. We analyzed the function of candidate genes by searching the gene ontology databases and publications and revealed that these genes functioned in cell cycle, cell proliferation, apoptosis, cell migration, DNA repair, transcription, metabolism, response to stimulus and signal transduction. This study provides new perspectives on NKRF’s potential multiple functions.

In microarray data mining, one of the key problems is how to handle weak signals. Based on a bent piecewise linear accumulated distribution generally found in the microarray data, a new detectable threshold finding method is proposed to filter genes with unreliable information in this paper. More reliable and reproducible data is produced for the subsequent data mining.

The oligonucleotide microarray, a high-throughput polymorphism detection technology, holds great promise for the characterization of complex genetic variance. To achieve greater sensitivity and specificity for it to be an effective platform technology we present results and discuss some of the factors influencing signal intensities and single-mismatch discrimination in array-based mutation/SNP detection. Probes with a series of concentrations were spotted onto the slide in order to find the optimal concentration with the identifiable satisfying signals and the stable ratios between matched and mismatched probes. It was found that under our experimental conditions, when the initial probe concentration is higher than the maximum immobilization capability of the slide (7.5 µM), the hybridization signal will be saturated and the ratio between matched and mismatched probes will be more stable than at a lower probe concen-tration. Considering the cost of probes and the systematic stability, a constant spotting cone n-tration of 10 µM was selected. The stability of different types of mismatched oligo-DNA du-plexes on the glass surface was also confirmed. The results show that the order of stability of mismatched oligo-DNA duplexes on a glass surface is in general agreement with previous reports conducted using liquid and polyacryl-amide gel pads. This suggests that the influence of the mismatched base pair on the stability of the duplex in a solid hybridization system is similar to that in the solution hybridization environment.
Macrophage Migration Inhibitory Factor (MIF) Interacts with Bim and Inhibits Bim-mediated Apoptosis

The pro-apoptotic Bcl-2 family member Bim acts as a sensor for apoptotic stimuli and initiates apoptosis through the mitochondrial pathway. To identify novel regulators of Bim, we employed the yeast two-hybrid system and isolated the human gene encoding macrophage migration inhibitory factor (MIF), a ubiquitously expressed proinflammatory mediator that has also been implicated in cell proliferation, the cell cycle and carcinogenesis. The interaction between MIF and Bim was confirmed by both in vitro and in vivo protein interaction assays. Intriguingly, protein complexes between MIF and the three major Bim isoforms (BimEL/BimL/BimS) could be detected in HEK293 and K562 cells, especially in cells undergoing apoptosis. Moreover, exogenous expression of MIF partially inhibited Bim-induced apoptosis in HEK293 cells. SiRNA-mediated knockdown of MIF increased apoptosis in K562 cells exposed to the chemical oxidant diamide. Endogenous MIF may regulate the pro-apoptotic activity of Bim and inhibit the release of cytochrome c from mitochondria.
