Human α-galactosidase A (GLA) has been used in enzyme replacement therapy for patients with Fabry disease. We expressed recombinant GLA from Chinese hamster ovary cells with very high productivity. When compared to an approved GLA (agalsidase beta), its size and charge were found to be smaller and more neutral. These differences resulted from the lack of terminal sialic acids playing essential roles in the serum half-life and proper tissue targeting. Because a simple sialylation reaction was not enough to increase the sialic acid content, a combined reaction using galactosyltransferase, sialyltransferase, and their sugar substrates at the same time was developed and optimized to reduce the incubation time. The product generated by this reaction had nearly the same size, isoelectric points, and sialic acid content as agalsidase beta. Furthermore, it had better in vivo efficacy to degrade the accumulated globotriaosylceramide in target organs of Fabry mice compared to an unmodified version.

Purpose: Attention deficit hyperactivity disorder (ADHD) is a common disorder in school-aged children. Patients with restless legs syndrome (RLS) often present with ADHD symptoms and vice versa. Thisstudy was the first to attempt to identify the prevalence of RLS and sleep problems in children withADHD in Korea. Methods: Patients diagnosed with ADHD were asked to complete a sleep questionnaire. The sleepquestionnaire included items to help identify the presence of four typical symptoms that are used asdiagnostic criteria for RLS. Results: A total of 56 patients, including 51 boys and 5 girls (mean age, 10.7 years old) participated. Of these, 24 complained of pain, discomfort, or an unpleasant sensation in the legs. Based on the RLSdiagnostic criteria, 2 patients were diagnosed with definite RLS and 4 with probable RLS. There wereno significant differences in age, medication dosage, or neuropsychological test scores between thepatients with and without RLS symptoms. Conclusion: Approximately 42.9% of patients with ADHD presented with RLS symptoms and 7.1% ofthese were diagnosed with RLS. Patients with ADHD also experienced various other sleep disorders. Thus, appropriate assessment and treatment for sleep disorders in patients with ADHD is essential.

<P><B>BACKGROUND:</B></P><P><I>Lens culinaris</I> agglutinin-reactive fraction of α-fetoprotein (AFP-L3) is a serum biomarker for hepatocellular carcinoma (HCC). AFP-L3 is typically measured by liquid-phase binding assay (LiBA). However, LiBA does not always reflect AFP-L3 concentrations because of its low analytical sensitivity. Thus, we aimed to develop an analytically sensitive multiple reaction monitoring–mass spectrometry (MRM-MS) assay to quantify AFP-L3 in serum.</P><P><B>METHODS:</B></P><P>The assay entailed the addition of a stable isotope-labeled internal standard protein analog, the enrichment of AFP using a monoclonal antibody, the fractionation of AFP-L3 using <I>L. culinaris</I> agglutinin lectin, deglycosylation, trypsin digestion, online desalting, and MRM-MS analysis. The performance of the MRM-MS assay was compared with that of LiBA in 400 human serum samples (100 chronic hepatitis, 100 liver cirrhosis, and 200 HCC). Integrated multinational guidelines were followed to validate the assay for clinical implementation.</P><P><B>RESULTS:</B></P><P>The lower limit of quantification of the MRM-MS assay (0.051 ng/mL) for AFP-L3 was less than that of LiBA (0.300 ng/mL). Thus, AFP-L3, which was not observed by LiBA in HCC samples (n = 39), was detected by the MRM-MS assay, improving the clinical value of AFP-L3 as a biomarker by switching to a more analytical sensitive platform. The method was validated, meeting all the criteria in integrated multinational guidelines.</P><P><B>CONCLUSIONS:</B></P><P>Because of the lower incidence of false-negative findings, the MRM-MS assay is more suitable than LiBA for early detection of HCC.</P>

오늘날 다양한 종류의 지식을 창조해내는 창의성이야말로 디지털 경제시대에 기업 경쟁력의 핵심원천이라고 할 수 있다. 본 연구는 창의성 및 창의성 경영에 대한 이론고찰과 실증연구의 결과를 토대로 창의성 경영의 필요성 및 유용성을 밝히고자 하였다. 연구 결과 문화적 창의성, 전략적 창의성, 관리적 창의성이라는 구성차원이 주요한 창의성 경영의 과정특성을 구성하는 것으로 나타났다. 또한 창의성 경영의 투입-과정 변수와 결과로서의 경영성과간에 높은 상호관련성이 존재하는 것으로 나타났으며, 특히 제도 투입요인과 전략적 창의성이라는 과정특성이 창의성 성과를 설명함에 있어 가장 중요한 요인임이 밝혀졌다. This study investigated the relationships between creativity management factors and corporate performance. To do this, we collected data from 250 Korean firms (response rate 32.7%) and tested 6 hypothesis derived from I-P-O(system) model of creativity management (hereafter, CM). Major findings are as follows: 1) CM process was actually grouped into cultural factor, strategic & top management factor, and organization & operation factor, instead of 5 factors that we expected, 2) there exists strong correlations among CM input variables, process variables, and corporate performance, especially, managerial factors and strategic creativity are most important factors explaining corporate performances, 3) KOSDAQ firms revealed higher levels of CM input, process, and performance than KOSE firms. However, industry characteristics did not make any significant differences.

<P>Reproducible sample preparation remains a significant challenge in large-scale clinical research using selected reaction monitoring-mass spectrometry (SRM-MS), which enables a highly sensitive multiplexed assay. Although automated liquid-handling platforms have tremendous potential for addressing this issue, the high cost of their consumables is a drawback that renders routine operation expensive. Here we evaluated the performance of a liquid-handling platform in preparing serum samples compared with a standard experiment while reducing the outlay for consumables, such as tips, wasted reagents, and reagent stock plates. A total of 26 multiplex assays were quantified by SRM-MS using four sets of 24 pooled human serum aliquots; the four sets used a fixed number (1, 4, 8, or 24) of tips to dispense digestion reagents. This study demonstrated that the use of 4 or 8 tips is comparable to 24 tips (standard experiment), as evidenced by their coefficients of variation: 13.5% (for 4 and 8 tips) versus 12.0% (24 tips). Thus we can save 37% of the total experimental cost compared with the standard experiment, maintaining nearly equivalent reproducibility. The routine operation of cost-effective liquid-handling platforms can enable researchers to process large-scale samples with high throughput, adding credibility to their findings by minimizing human error.</P> [FIG OMISSION]</BR>

<P>Recently, the recombinant 30Kc19 protein, originating from silkworm hemolymph of <I>Bombyx mori</I> has attracted attention due to its cell-penetrating property and potential application as a protein delivery system. However, this observation of penetration across cell membrane has raised questions concerning the interaction of the protein-lipid bilayer. Here, we report a dimerization propensity of the 30Kc19 protein in the presence of amphiphilic moieties; sodium dodecyl sulfate (SDS) or phospholipid. Native PAGE showed that the 30Kc19 monomer formed a dimer when SDS or phospholipid was present. In the glutathione-<I>S</I>-transferase (GST) pull-down assay, supplementation of the 30Kc19 protein to mammalian cell culture medium showed dimerization and penetration; due to phospholipids at the cell membrane, the main components of the lipid bilayer. Mutagenesis was performed, and penetration was observed by 30Kc19 C76A and not 30Kc19 C57A, which meant that the presence of cysteine at position 57 (Cys-57) is involved in dimerization of the 30Kc19 at the cell membrane during penetration. We anticipate application of the native 30Kc19 protein with high cell-penetrating efficiency for delivery of cargos to various cell types. The intracellular cargo delivery using the 30Kc19 protein is a non-virus derived (e.g. TAT) delivery method, which can open up new approaches for the delivery of therapeutics in bioindustries, such as pharma- and cosmeceuticals.</P>
