Inducible cyclooxygenase (COX-2) has been implicated in the processes of inflammation and carcinogenesis. Thus, the potential COX-2 inhibitors have been considered as anti-inflammatory or cancer chemopreventive agents. In this study, the methanolic extract of the cortex of Eugenia caryophyllata Thunberg (Myrtaceae) was found to potently inhibit the prostaglandin E₂ production in lipopolysaccharide (LPS)-activated mouse macrophage RAW264.7 cells (98.3% inhibition at the test concentration of 10 ㎍/㎖). Further, hexane-soluble layer was the most active partition compared to ethyl acetate, n-butanol, and water-soluble parts. By bioassay-guided fractionation of hexane-soluble partition, eugenol was isolated and exhibited a significant inhibition of PGE₂ production (IC_(50) = 0.37 μM). In addition, eugenol suppressed the cyclooxygenase-2 (COX-2) gene expression in LPS-stimulated mouse macrophage cells. On the line of COX-2 playing an important role in colon carcinogenesis further study was designed to investigate the effect of eugenol on the growth and COX-2 expression in HT-29 human colon cancer cells. Eugenol inhibited the proliferation of HT-29 cells and the mRNA expression of COX-2, but not COX-1. This result suggests that eugenol might be a plausible lead candidate for further developing the COX-2 inhibitor as an anti-inflammatory or cancer chemopreventive agent.

Since reactive oxygen radicals play an important role in carcinogenesis and other human diseases including neurodegenerative states, antioxidants present in natural products have received considerable attention for alleviation of these disease states. Therefore, in order to identify antioxidants in plant extracts, fifty-seven methanolic extracts derived from indigenous Korean plants were primarily assessed for potential to scavenge stable l,l-diphenyl-2-picrylhydrazyl (DPPH) free radicals. As a result, nine plant extracts were found to exhibit the DPPH free radical scavenging activity in the criteria of IC_(50)) <40㎍/㎖. In particular, the extracts of Melioma oldhami (IC_(50)= 0.1㎍/㎖), Myrica rubra (IC_(50)=16.2㎍/㎖), Sympolocos paniculata (IC_(50)=23.0㎍/㎖), Carpinus laxiflora (IC_(50) = 25.1㎍/㎖), and Cleyera japonica (IC_(50) = 26.2㎍/㎖) showed a potent radical scavenging activity. Further study for the identification of active compounds from these lead extracts might be warranted.
Sesquiterpenoids from the Rhizome of Curcuma zedoaria

In the course of searching for biologically active sesquiterpenoids from Curcuma genus, twoo sesquiterpenoids were isolated from the rhizome of Curcuma zedoaria(Zingiberaceae). Their structures were identified as ar-turmerone(1) and β-turmerone(2). The structure elucidation of compounds 1 and 2 was carried out by comparison of their physical and spectral data with previously reported values.
