        Efficient Development of Stable Recombinant Chinese Hamster Ovary (rCHO) Cell Lines to Produce Antibodies by Using Dimethyl Sulfoxide (DMSO) in Electroporation

        Development of stable rCHO cell lines is still time consuming and labor intensive, although it is a critical step in the commercial development of recombinant antibodies. The current work demonstrates, for the first time, that electroporation of CHO cells with DMSO can enhance stable expression of recombinant antibodies in rCHO cells. Electroporation with DMSO resulted in an average 3.7-fold and 2.8-fold increases in expression levels of aflibercept and pembrolizumab, respectively, in pools of stable rCHO cells. It also resulted in an average of 2.2-fold and 2.6-fold increases in the expression of aflibercept and pembrolizumab, respectively, in single-cell derived rCHO clones. Simple batch cultures of rCHO cell clones with the highest expression produced 1.0 g/l for aflibercept and 1.4 g/l for pembrolizumab without a time-consuming gene amplification process. Electroporation with DMSO also shortened the development of rCHO cell lines to 2-3 months, allowing rapid establishment of stable rCHO cell lines with a desirable expression level antibodies.

      • Complexation and Decomplexation study in Less Symmetric Hemicarcerands

        Hemicarcerands are defined as host system which can incarcerate and liberate guests at elevated temperatures, but form stable hemicarceplexes at lower working temperatures. Decomplexation rates were examined for DMSO complexes of four different hemicarcerands which have a different fourth bridge such as CH₂, (CH₂)₂, (CH₂)₃, and (CH₂)₄groups, respectively. When unique bridge was anthracene moiety, two sets of host were observed in 1H NMR at 25℃ and the signals due to chloroform complex disappeared at 55℃. The effect of host structures on ¹H and ?? NMR spectra of trifluoromethylbenzene was studied by the chemical shift change of this incarcerated guest. At 100℃, the incarcerated veratrole showed sharp peaks in its ¹H NMR spectrum, which enabled the assignments of all guest aromatic peaks. Finally, a hemicarcerand containing sulfur group in its fourth bridge has been synthesized and modification of this sulfur group to sulfone was done in a moderate yield.

        In the case of a suspension part, since it is located in the lower part of the vehicle, it has a great advantage of improving fuel economy due to weight reduction. In order to reduce the weight of vehicles, there are increasing cases of applying aluminum to suspension parts. This paper describes the stages of development of a rear cross member with a welded aluminum low pressure hollow casting method and extrusion method. The result of the aluminum cross member study showed that the weight was reduced by 6.3㎏ and natural frequency was improved by 46.7㎐ compared with the steel cross member.

        <P>The design and development of new pyrene-based fluorescent probes, <B>P-Hcy-1</B> and <B>P-Hcy-2</B>, which display selective fluorescence enhancements in response to homocysteine (Hcy), are described. The distinctly different fluorescence responses of <B>P-Hcy-1</B> and <B>P-Hcy-2</B> to Hcy <I>vs.</I> Cys are explained by theoretical calculations.       • Synthesis of Hierarchical Silica/Titania Hollow Nanoparticles and Their Enhanced Electroresponsive Activity

        <P>Wrinkled silica nanoparticle (WSN)-based hollow SiO2/TiO2 nanoparticles (W-HNPs) with hierarchically arrayed internal surfaces were prepared via the combination of sol-gel, TiO2 coating, and etching of core template techniques. The hierarchical internal surface of W-HNPs was attained using WSNs as a core template. Compared with SiO2 sphere-templated hollow SiO2/TiO2 nanoparticles (S-HNPs) with flat inner surfaces, W-HNPs displayed distinctive surface areas, TiO2 loading amounts, and dielectric properties arising from the hierarchical internal surface. The unique properties of W-HNPs were further investigated as an electrorheological (ER) material. W-HNP-based ER fluids exhibited ca. 1.9-fold enhancement in the ER efficiency compared to that of S-HNP-based ER fluids. Such enhancement was attributed to the unique inner surface of W-HNPs, which effectively enhanced the polarizability by increasing the number of charge accumulation sites, and to the presence of the high-dielectric TiO2. This study demonstrated the advantages, in terms of practical ER applications, of hollow nanomaterials having uniquely arrayed internal spaces.</P>

        Preparation of pore size controllable macroporous polymer beads

        Macroporous polymer beads have been used in various fields, for example as solid supports in solid-phase organic synthesis and heterogeneous catalysts and as adsorbents in column chromatography. Their broad applications have stimulated many researchers to develop various kinds of macroporous polymer beads. However, despite all the efforts, it is still hard to control the porosity and pore size distributions within the polymer beads. In this study, we prepared pore size controllable macroporous poly(methyl methacrylateco-methyl methacrylic acid) (PMMA-co-PMAA) beads. First, we prepared silica submicron particles embedded PMMA beads by the polymerization of methyl methacrylate monomer with mono-dispersed silica particles. Then, the silica NP embedded PMMA beads were hydrolyzed with sodium hydroxide to obtain a macroporous structure by removing the embedded silica particles. At the same time, part of the methyl methacrylate turned into methacrylic acid, which caused the PMMA beads to become PMAA-co-PMMA beads. Through this method, two types of beads having different pore sizes were prepared as a model study. The pore size of the beads could be easily controlled by adjusting the size of the silica particles.

      • Influence of Information on Emotion for Consumer Test of 6 Novel Korean Traditional Beverages to US Consumers

        The objectives of this study were to determine how US consumers perceive 6 Korean traditional beverages (Bokbunja, Hongsaam, Korean date, Omija, Sansuyu, and Yuja) and to find out the influence of extrinsic information (name, nutritional information, picture of raw material, and stories related to each sample) on emotion using EsSense Profile™ when consuming novel beverages. Bokbunja, Yuja, and Korean date were samples received high acceptability and evoked mostly positive emotions. When the extrinsic information of the samples was provided, acceptability of all six samples tended to increase. Information also influenced the perceived emotion of consumers while evaluating samples. With the difference between samples, there were emotion terms showed significant differences of intensity between the blind and informed conditions. Also principal component analysis biplots of emotions revealed the influence of information during evaluation. When tested in the informed condition, samples evoked relatively positive emotion compare to sample tested in the blind condition.

      • Determination of migrated heavy metals from poly (cyclohexane-1,4-dimethylene terephthalate (PCT) Food contact materials

        Poly(cyclohexane-1,4-dimethylene terephthalate (PCT) has excellent chemical resistance and color safety and is used as a substitute for Bisphenol A in various food containers. Food contact materials may contain trace amounts of heavy metals from residual chemical, additives and contaminants during the manufacturing process. That could be migrated into food when packaging materials are used for handling, cooking and storing. The purpose of this study was to determine heavy metals such as Pb, Cd, As and Sb from PCT food packaging materials. Total 100 samples such as food containers, baby bottle and blender were purchased from domestic markets. Migration tests with a food simulant (4% acetic acid) were examined at 100°C for 30 min. The heavy metal levels in the food simulant were quantified by ICP-MS. The analytical methods were validated by measuring the limit of detection (LOD), limit of quantification (LOQ) and recovery. The amounts of migration of Pb, Cd, As and Sb ranged from N.D.∼0.490 μgL<SUP>-1</SUP> at 100°C. It would be applicable to risk assessment of heavy metals for the safety management of PCT food packaging materials.

        Endocrinology-metabolism ; Relationship between metabolic syndrome and thyroid nodules in healthy Koreans

        Background/Aims: This study evaluated the relationship between thyroid nodules and metabolic syndrome (MS) and its components in apparently healthy Koreans. Methods: We reviewed the records of 3,298 subjects with no noticeable symptoms who underwent thyroid ultrasound imaging as part of a routine check-up between July 2009 and June 2010; of these, 1,308 were excluded based upon predefined criteria. Among the remaining 1,990 patients, we examined the association between MS and its components and the incidence of thyroid nodules. Results: Of the 1,990 subjects included in this study, 38.4% (n = 764) had thyroid nodules and 12.7% (n = 253) had MS. Female sex, older age, higher body mass index, larger waist circumference, higher glycated hemoglobin level, lower thyroid stimulating hormone level, and presence of MS were all closely related with the presence of thyroid nodules (all p < 0.05). Furthermore, the relevant number of MS components showed a positive linear correlation with the occurrence of thyroid nodules (p < 0.001). Evidence of MS alone was not independently associated with thyroid nodules after adjusting for sex and age in a multivariate binary logistic regression analysis; however, glycated hemoglobin for females and waist circumference for males, as well as both age and thyroid stimulating hormone for all patients, were identified as independent predictors for the existence of thyroid nodules (all p < 0.05).Conclusions: This study suggests a positive relationship between the components of MS and thyroid nodules in an ostensibly healthy Korean population. Our data support the idea that the recent increase in thyroid nodules is partly due to increases in both MS and obesity.
