Proteomic Approach Analysis of Mammary Membrane Proteins Expression Profiles in Holstein Cows

To investigate host defense mechanisms for protecting the mammary gland from mastitis infection, the membrane fraction of mammary tissues from Holstein cows was purified by differential velocity centrifugation, and then the sodium dodecyl sulfate-polyacrylamid gel electrophoresis (SDS-PAGE) separated proteins were identified by ion trap mass spectrometer equipped with a Surveyor high performance liquid chromatography (HPLC) system. A total of 183 proteins were identified. Bioinformatics software was applied to analyse physicochemical characteristics of the identified proteins and to predict biochemical function. These data may provide valuable information to investigate the mechanisms of mammary gland milk secretion and infectious disease, and enable a clear identification of proteins and potential protein targets for therapies.
Temperature-Dependent Structural and Transport Properties of Liquid Transition Metals

The evolution of the atomic structures and diffusivity of liquid transition metals with increasing temperature has been studied here using molecular dynamics. An analysis of Honeycutt and Andersen (HA) Indices indicates that relatively low order atomic clusters of rhombohedra-related structures increase with an increase in temperature. The results illustrate that the distortion in the local structural order with a predominant rhombohedra character enhances the diffusivity in liquid metals. The excess entropy approximated by the two-body contribution increases with the distortion of the local structural order traced by the HA Indices. The relationship between the excess entropy and the reduced diffusion coefficient supports the universal scaling law proposed by M. Dzugutov. The calculated diffusivities were compared with predictions of four diffusion models. The temperature dependence of the diffusivity cannot be described by the Arrhenius Law, the moving oscillator model or the free volume model but rather by the density fluctuation model with the square proportionality of temperature.
Risk analysis for the compressed natural gas (CNG) station explosion accident

With the shortage of the gasoline resource day by day, the adjustment of county"s energy strategy, environment engineering requirement and the continuous progress of the automobile technology, more and more autobiles were designed on the CNC as their fuel and correspondingly CNG station were increased at the same time. But the subsequent accidents such as the leakage of the gas and the fire disaster make the security fo the CNG station become a serious problem which people should pay more and more attention on it. If the gas leaked were not ignited immediately it may construct gas cloud cluster and may expolde when it comes out an open fire. Consequently people in a certain idstance may be wounded by the shockwave and the architectures in the dangerous range will be destroyed. This paper contribute to calculate the dangerous range of the CNG station explosion accident and to formulate the evacuate conditions for pedestrian after the accident.
Aggregation mechanism of fine fly ash particles in uniform magnetic field

of fly ash particles with size range of 0.023-9.314m in a uniform magnetic field was inves-tigated for removing them. A binary collision-aggregation model evaluating particle aggregation coefficient was devel-oped. Based on the model, particle removal efficiency was calculated by solving the General Dynamic Equation. Thecomparison was done between the calculated and experimental data. The modeling aggregation coefficient shows thatthe aggregation coefficient increases with particle size, and the bigger the size difference between two particles is, thehigher than those of the smaller and bigger ones. The effect of the magnetic flux density on total particle removal effi-ciency is similar to that on aggregation coefficient. Before particles are saturatedly magnetized, their total removal effi-ciency increases with an increase in the magnetic flux density. Higher removal efficiency can also be caused by pro-longing the particle residence time in the magnetic field or increasing their mass concentration. The particle numbermedian diameter decreases with an increase in the total removal efficiency. Calculation results are found to coincideessentially with the experimental data.
Molecular profiling analysis for colorectal cancer patients with Pi-Xu or Shi-Re syndrome

Background: Traditional Chinese medicine (TCM) syndromes (ZHENG in Chinese) constitute the basis of understanding the disorders of patients and guiding the use of the Chinese herbs. Colorectal cancer is divided into various subtypes mainly according to the ZHENG identification. Objective: We aimed to determine the molecular basis underlying Pi-Xu (spleen deficiency) and Shi-Re (dampness-heat) ZHENG that are commonly found in colorectal cancer patients. Methods: About 80 colorectal cancer patients, including 47 Pi-Xu ZHENG and 33 Shi-Re ZHENG were enrolled. Blood and tissue samples of these patients were available for protein and mRNA expression. The protein expression was determined by Immunohistochemistry (IHC) staining and mRNA profiling was detected by expression microarray. Furthermore, mRNA fold change was evaluated by qRT-PCR. Results: The colorectal cancer patients with Shi-Re ZHENG had a poor prognosis, compared with Pi-Xu ZHENG (95% CI: 0.05–0.33; p < 0.0001). Moreover, there was a significant difference in protein expression levels (especially for mutant TP53, PCNA, PD-L1 and Ki-67) among Pi-Xu and Shi-Re ZHENG (p < 0.01). Meanwhile, mRNA expression (especially for wild type TP53, KDM6A, PCNA, PD-L1, Ki-67, CCL-2, IL-1a and COX-2) was also remarkably different between Pi-Xu and Shi-Re groups (p < 0.01). Conclusion: Our results suggest that Shi-Re ZHENG conditions may contribute to poor overall survival in patients with colorectal cancer. Compared with Pi-Xu ZHENG, high expression of mutant TP53, PCNA, PD-L1, Ki-67, CCL-2, IL-1a and COX-2 may serve as potential biomarkers for diagnosis and prognosis of colorectal cancer patients displayed Shi-Re ZHENG.
NOx emission from incineration of organic liquid waste in a circulating fluidized bed

incineration test of a toxic chemical organic waste liquid was conducted on a circulating fluidized bed(CFB) incinerator. The flue gas was measured online with the advanced SAE-19 flue gas analyzer. The effects of severalfactors, in terms of flow rate of waste liquid, ratio of waste liquid injected into dense bed of the CFB, excess air co-efficient, the secondary air fraction and bed temperature on NOx emissions, were verified. The experimental resultsthe excess air coefficient or the bed temperature and those decrease with increase in the ratio of waste liquid injectedinto the dense bed of the CFB or the secondary air fraction. During the test runs, NOx concentration in flue gas metthe national regulation on NOx emissions due to suppresive effect of low temperature and staged combustion in CFBon NOx formation.

Purpose: The purpose of the present study was to investigate the aberrance of histone H3 lysine 4 trimethylation (H3K4me3) in patients with IgA Nephropathy (IgAN). Materials and Methods: In this study, H3K4me3 variations in peripheral blood mononuclear cells (PBMCs) from 15 IgAN patients and 15 healthy subjects were analyzed using chromatin immunoprecipitation linked to microarrays analysis (ChIP-chip). ChIP real-time PCR was used to validate the microarray results. Expression analysis by quantitative real-time PCR (qRT-PCR) revealed correlations between mRNA and H3K4me3 levels. DNA methylation status was analyzed by quantitative methylation-specific PCR. Results: We found that 321 probes displayed significant H3K4me3 differences in IgAN patients compared with healthy controls. Among these probes, 154 probes displayed increased H3K4me3and 167 probes demonstrated decreased H3K4me3. For further validation, we selected 4 key relevant genes (FCRL4, GALK2, PTPRN2 and IL1RAPL1) to study. The results of ChIP real-time PCR coincided well with the microarray data. Quantitative RT-PCR revealed the correlations between the mRNA expression and the methylation levels of H3K4me3. Different degrees of DNA methylation alterations appeared on the selected positive genes. Conclusion: Our studies indicated that there were significant alterations in H3K4me3 in IgAN patients. These findings may help to explain the disturbed immunity and abnormal glycosylation involved in IgAN patients.

The antioxidant activity of proanthocyanidins extracted from the lotus seedpods (LSPC) in vitro and ameliorative effects on memory impairment induced using scopolamine in mice were studied. 2,2-diphenyl-1- picrylhydrazyl (DPPH) radical scavenging ability, oxygen radical absorbance capacity, and cellular antioxidant activity assays revealed a potent LSPC antioxidant activity. Y-maze and step-down avoidance testing showed that LSPC administration at 30, 60, and 90 mg/kg ameliorated memory impairment. LSPC improved glutathione peroxidase and superoxide dismutase activities, inhibited activities of monoamine oxidase-B, total nitric oxide synthase, neuronal nitric oxide synthase, and acetylcholinesterase, and had no influence on inducible nitric oxide synthase and nitric oxide levels in the brain. LSPC ameliorated scopolamineinduced memory impairment based on improvement of the antioxidant system and cholinergic activity, which may be associated with potent antioxidant ability.
