        Mitochondrial genome of Euchirus longimanus (Coleoptera: Scarabaeidae) and phylogenetic relationship of Euchirini

        The long-armed scarab beetles, a charismatic group within the small tribe Euchirini, are characterized by their large body sizes and distinctly elongated forelegs in males. While mitochondrial genomes have been sequenced for two of the included genera, Cheirotonus and Propomacrus, the type genus of the tribe, Euchirus, remains unexplored. It will provide valuable insights into the phylogenetic position and internal relationships of the Euchirini to investigate the mitochondrial genome of Euchirus and conduct phylogenetic analyses. In this study, it presents the first mitochondrial genome of Euchirus longimanus. The length of the mitochondrial genome of Euchirus longimanus is 16,591 bp. We analyzed its genes arrangement, base composition, codon usage frequency, and constructed phylogenetic trees based on the nucleic acid sequences of 13 protein-coding genes. The phylogenetic results are consistent with previous studies, confirming the phylogenetic position of Euchirini within the paraphyletic Melolonthinae. Notably, we firstly elucidate the phylogenetic relationship among all three genera: Propomacrus + (Cheirotonus + Euchirus), supported by robust values in Bayesian inference (BI) analysis. However, the maximum likelihood (ML) tree shows weaker support for the internal node of Euchirini and exhibits distinct topological variations compared to the BI tree, suggesting the potential paraphyly of Cheirotonus with the inclusion of Euchirus longimanus. These findings highlight the importance of including additional species of Euchirus and Cheirotonus in the future phylogenetic investigations to achieve a compre hensive understanding of their evolutionary relationships.

      • A New Bias Field Estimation Method based on Adapted PSO Method

        It is hard to segmentation brain MR images for the bias fields. In this paper, a new fuzzy anisotropic diffusion function is presented to reduce the effect of the noise. We use Legendre polynomial functions to reconstruct the bias field, which make the entropy of the recovered image be smallest. But it needs to compute a lot of parameters to reconstruct the bias. The traditional method uses the gradient descending method to compute the parameters. The method plunges into local best easily. In order to deal with this problem, Particle swarm optimization (PSO) method is analyzed. A new particle swarm technique is proposed that incorporates initial location information and use mutate operation make the particles away from local maxima. The experiments show that the new method can get accurate result robustly.

      • Photochemical and magnetic activities of FeTiO<sub>3</sub> nanoparticles by electro-spinning synthesis

        <P><B>Abstract</B></P> <P>Ilmenite FeTiO<SUB>3</SUB> nanoparticles were prepared by electrospinning synthesis using the sol–gel precursors, followed by the sintering at 650°C. The pure crystalline phase was verified by X-ray powder diffraction (XRD) measurements together with Rietveld refinements. The surface properties of the nanoparticles were studied by TEM, SEM, BET and XPS measurements. The optical absorption of FeTiO<SUB>3</SUB> nanoparticles is in the visible wavelength region. The indirect allowed band gap (2.6eV) of FeTiO<SUB>3</SUB> shows the characteristic charge-transfer (CT) transitions of O-2p→(Ti/Fe)-3d in visible light wavelength region. The photochemical properties were investigated by the methylene blue (MB) degradation under the irradiation of visible-light. FeTiO<SUB>3</SUB> nanoparticles were efficient in the photo-degradation of MB dye solutions. The photocatalysis activities and mechanisms were discussed on the crystal structure characteristics such as multivalent elements of Ti<SUP>3+</SUP>, Fe<SUP>3+</SUP>/Fe<SUP>2+</SUP> redox couples.         YAP1 inhibits the senescence of alveolar epithelial cells by targeting Prdx3 to alleviate pulmonary fibrosis

        The senescence of alveolar type II (AT2) cells impedes self-repair of the lung epithelium and contributes to lung injury in the setting of idiopathic pulmonary fibrosis (IPF). Yes-associated protein 1 (YAP1) is essential for cell growth and organ development; however, the role of YAP1 in AT2 cells during pulmonary fibrosis is still unclear. YAP1 expression was found to be downregulated in the AT2 cells of PF patients. Deletion of YAP1 in AT2 cells resulted in lung injury, exacerbated extracellular matrix (ECM) deposition, and worsened lung function. In contrast, overexpression of YAP1 in AT2 cells promoted alveolar regeneration, mitigated pulmonary fibrosis, and improved lung function. In addition, overexpression of YAP1 alleviated bleomycin (BLM) -induced senescence of alveolar epithelial cells both in vivo and in vitro. Moreover, YAP1 promoted the expression of peroxiredoxin 3 (Prdx3) by directly interacting with TEAD1. Forced expression of Prdx3 inhibited senescence and improved mitochondrial dysfunction in BLM-treated MLE-12 cells, whereas depletion of Prdx3 partially abrogated the protective effect of YAP1. Furthermore, overexpression of Prdx3 facilitated self-repair of the injured lung and reduced ECM deposition, while silencing Prdx3 attenuated the antifibrotic effect of YAP1. In conclusion, this study demonstrated that YAP1 alleviates lung injury and pulmonary fibrosis by regulating Prdx3 expression to improve mitochondrial dysfunction and block senescence in AT2 cells, revealing a potential novel therapeutic strategy for pulmonary fibrosis.

        Electronic structure and optical properties of V-doped Bi<sub>4</sub>Ti<sub>3</sub>O<sub>12</sub> nanoparticles

        <P><B>Abstract</B></P> <P>Pure and V<SUP>5+</SUP>-doped Bi<SUB>4</SUB>Ti<SUB>3</SUB>O<SUB>12</SUB> (BTO) photocatalysts were prepared by sol–gel routine and subsequent hydrothermal synthesis. The samples were developed into the platelike microparticles with the exposed (001) facets. The crystalline phases were verified via the XRD measurement. The surface characteristics of the photocatalysts were studied by TEM, SEM, BET, EDX and XPS measurements. The UV–vis optical absorption spectra showed that the V-doped photocatalysts exhibit an obvious red shift in the visible light absorption band compared with the pure BTO. Rhodamine B (RhB) dye solution was used as the model organic pollutants to verify the influence of V-doping on the photocatalytic activity. The 10 mol % V-doped BTO showed efficient photocatalytic ability.         Peripheral nerve defects repaired with autogenous vein grafts flled with platelet-rich plasma and active nerve microtissues and evaluated by novel multimodal ultrasound techniques

        Background: Developing biocompatible nerve conduits that accelerate peripheral nerve regeneration, lengthening and functional recovery remains a challenge. The combined application of nerve microtissues and platelet-rich plasma (PRP) provides abundant Schwann cells (SCs) and various natural growth factors and can compensate for the deficiency of SCs in the nerve bridge, as well as the limitations of applying a single type of growth factor. Multimodal ultrasound evaluation can provide additional information on the stiffness and microvascular flow perfusion of the tissue. This study was designed to investigate the effectiveness of a novel tissue-engineered nerve graft composed of an autogenous vein, nerve microtissues and PRP in reconstructing a 12-mm tibial nerve defect and to explore the value of multimodal ultrasound techniques in evaluating the prognosis of nerve repair. Methods: In vitro, nerve microtissue activity was first investigated, and the effects on SC proliferation, migration, factor secretion, and axonal regeneration of dorsal root ganglia (DRG) were evaluated by coculture with nerve microtissues and PRP. In vivo, seventy-five rabbits were equally and randomly divided into Hollow, PRP, Micro-T (Microtissues), Micro-T + PRP and Autograft groups. By analysing the neurological function, electrophysiological recovery, and the comparative results of multimodal ultrasound and histological evaluation, we investigated the effect of these new nerve grafts in repairing tibial nerve defects. Results: Our results showed that the combined application of nerve microtissues and PRP could significantly promote the proliferation, secretion and migration of SCs and the regeneration of axons in the early stage. The Micro-T + PRP group and Autograft groups exhibited the best nerve repair 12 weeks postoperatively. In addition, the changes in target tissue stiffness and microvascular perfusion on multimodal ultrasound (shear wave elastography; contrast-enhanced ultrasonography; Angio PlaneWave UltrasenSitive, AngioPLUS) were significantly correlated with the histological results, such as collagen area percentage and VEGF expression, respectively. Conclusion: Our novel tissue-engineered nerve graft shows excellent efficacy in repairing 12-mm defects of the tibial nerve in rabbits. Moreover, multimodal ultrasound may provide a clinical reference for prognosis by quantitatively evaluating the stiffness and microvescular flow of nerve grafts and targeted muscles

        Melatonin prevents lung injury by regulating apelin 13 to improve mitochondrial dysfunction

        Pulmonary fibrosis is a progressive disease characterized by epithelial cell damage, fibroblast proliferation, excessive extracellular matrix (ECM) deposition, and lung tissue scarring. Melatonin, a hormone produced by the pineal gland, plays an important role in multiple physiological and pathological responses in organisms. However, the function of melatonin in the development of bleomycin-induced pulmonary injury is poorly understood. In the present study, we found that melatonin significantly decreased mortality and restored the function of the alveolar epithelium in bleomycin-treated mice. However, pulmonary function mainly depends on type II alveolar epithelial cells (AECIIs) and is linked to mitochondrial integrity. We also found that melatonin reduced the production of reactive oxygen species (ROS) and prevented apoptosis and senescence in AECIIs. Luzindole, a nonselective melatonin receptor antagonist, blocked the protective action of melatonin. Interestingly, we found that the expression of apelin 13 was significantly downregulated in vitro and in vivo and that this downregulation was reversed by melatonin. Furthermore, ML221, an apelin inhibitor, disrupted the beneficial effects of melatonin on alveolar epithelial cells. Taken together, these results suggest that melatonin alleviates lung injury through regulating apelin 13 to improve mitochondrial dysfunction in the process of bleomycin-induced pulmonary injury.

        Comparison of Direct and Extraction Immunoassay Methods With Liquid Chromatography-Tandem Mass Spectrometry Measurement of Urinary Free Cortisol for the Diagnosis of Cushing’s Syndrome

        Background: Twenty-four-hour urinary free cortisol (UFC) measurement is the initial diagnostic test for Cushing’s syndrome (CS). We compared UFC determination by both direct and extraction immunoassays using Abbott Architect, Siemens Atellica Solution, and Beckman DxI800 with liquid chromatography-tandem mass spectrometry (LC-MS/MS). In addition, we evaluated the value of 24-hr UFC measured by six methods for diagnosing CS. Methods: Residual 24-hr urine samples of 94 CS and 246 non-CS patients were collected. A laboratory-developed LC-MS/MS method was used as reference. UFC was measured by direct assays (D) using Abbott, Siemens, and Beckman platforms and by extraction assays (E) using Siemens and Beckman platforms. Method was compared using Passing–Bablok regression and Bland–Altman plot analyses. Cut-off values for the six assays and corresponding sensitivities and specificities were calculated by ROC analysis. Results: Abbott-D, Beckman-E, Siemens-E, and Siemens-D showed strong correlations with LC-MS/MS (Spearman coefficient r=0.965, 0.922, 0.922, and 0.897, respectively), while Beckman-D showed weaker correlation (r=0.755). All immunoassays showed proportionally positive bias. The areas under the curve were 0.975 for Abbott-D, 0.972 for LC-MS/MS, 0.966 for Siemens-E, 0.948 for Siemens-D, 0.955 for Beckman-E, and 0.877 for Beckman-D. The cut-off values varied significantly (154.8–1,321.5 nmol/24 hrs). Assay sensitivity and specificity ranged from 76.1% to 93.2% and from 93.0% to 97.1%, respectively. Conclusions: Commercially available immunoassays for measuring UFC show different levels of analytical consistency compared to LC-MS/MS. Abbott-D, Siemens-E, and Beckman-E have high diagnostic accuracy for CS.
