        효소면역측정법에 의한 우유중의 Aflatoxin M1 분석

        국내산 우유중 aflatoxin M_1 (AFM_1)의 오염현황을 조사하기 위하여, 효소면역측정법(ELISA)을 개발하고 이를 이용한 독소의 정량을 행하였다. 항체의 생산을 위하여 AFM_1 bovine serum albumin 결합체를 Freund's adjuvant와 함께 토끼에 피하주사하여 면역하였다. 가장 높은 항체역가를 보인 항혈청과 AFB_1-horseradish peroxidase 결합체를 이용하여 직접법에 의한 경합적 효소면역측정법(competitive direct ELISA ; cdELISA)의 조건을 확립하였으며, 그 검출한계는 0.003ppb로 나타났다. 또한 유사독소에 대한 교차반응율은 aflatoxin M_1, M_2, B_1, _2, G_1, G_2, B_2a, and G_2a 에 대하여 각각 100, 29.9, 25.0, 2.7, 13.0, 0.65, 0, 및 0%이었다. 다음으로, AFM_1을 인위적으로 오염시킨 우유를 C_18 cartridge로 세척후 cdELISA로 분석하였을 때, 0.01~1 ppb(10~1,000 ppt)의 범위에서 그 회수율의 평균은 104%(CV의 평균, 6.4%)로 나타났다. 시유 및 목장우유를 대상으로 AFM_1의 오염현황을 cdELISA로 조사하였을 때, 평균 오염도는 80.4±55.0 ppt (n=64; range, 5,6~280 ppt)이었다. For a survey of the occurrence of aflatoxin M_1 (AFM_1) in domestic cow's milk, we developed an enzyme-linked immunosorbent assay (ELISA) system, and quantitated the toxin in cow's milk. In order to produce specific antibodies AFM_1 conjugated to bovine serum albumin (AFM_1-BSA) and Freund's adjuvant were immunized subcutaneously to rabbits. By use of the antiserum showing the highest titer and AFB_1-HRP conjugate, we established a competitive direct ELISA (cdELISA) for AFM_1 whose detection limit was 0.003 ppb. The cross-reactivities of the antiserum against aflatoxin M_1, M_2, B_1, _2, G_1, G_2, B_2a, and G_2a were 100, 29.9, 25.0, 2.7, 13.0, 0.65, 0, and 0%, respectively. When the cdELISA was applied to the cow's milk spiked with AFM_1 and followed by cleanup with C_18 cartridge, the mean recovery of the assay was 104% (mean of CV, 6.4%) in the final concentration of 0.01~1 ppb (10~1,000 ppt). When cow's milk samples gathered from markets and farms were assayed by the cdELISA, the mean concentration and SD of AFM_1 was 80.4±55.0 ppt (n=64; range, 5,6~280 ppt).

        효소면역측정법을 이용한 Fumonisin의 검출법 개발

        Fumonisin에 대한 효소면역측정법(ELISA)을 개발하기 위하여 특이항체를 생산하고, 분석조건을 확립하여, 인위적으로 오염시킨 옥수수 시료의 분석을 행하였다. Fumonisin B_1 (FB_1)을 cholera toxin (CT)에 결합시킨 FB_1-CT conjugate를 면역원으로 하여 Freund's adjuvant와 함께, 또는 단독으로 두 군의 토끼에 면역하고 특이항체를 생산하였다. 항혈청을 ELISA로 분석한 결과 그 adjuvant와 함께 면역한 경우 항체역가(titer)가 높게 나타났다. 가장 높은 항체역가(1 : 16,000)를 나타낸 항혈청 및 그로부터 정제한 항체를 이용하여 간접법 및 직접법에 의한 경합적 ELISA(ciELISA 및 cdELISA)를 각각 확립하였다. 이 항체의 유사독소와의 교차반응을 ciELISA로 조사하였을 때, FB_3에 대한 교차반응율은 2%로 매우 낮았으나, FB_2에 대한 것은 179%로 다소 높았다. FB_1의 검출한계는 ciELISA에서 0.03ppb, cdELISA에서 0.3 ppb로 각각 나타났다. 실제 곡물시료의 분석에 ELISA 활용가능성을 조사하기 위하여 인위적으로 FB_1을 오염시킨 옥수수를 75% methanol로 추출하고 그 회수율을 측정한 결과, 분석이 불안정하여 회수율이 일정하게 나타나지 않았다. 그러나 방해물질의 제거를 위하여 시료추출액을 strong anion exchange(SAX) cartridge를 거쳐 세척한 다음 분석하였을 때, 3~10 ppm 범위의 FB_1 오염 옥수수에서 평균 34%(CV의 평균, 8.2%)의 안정된 회수율을 보였다. In order to develop enzyme-linked immunosorbent assay (ELISA) for fumonisins, production of specific antibodies, establishement of ELISA conditions, and quantitation of the toxin from spiked corns by ELISA were performed. Fumonisin B_1 (FB_1) conjugated to cholera toxin (CT) with or without Freund's adjuvant was subcutaneously injected into 2 groups of rabbits. When the titer of the antisera produced by each rabbit tested, higher titer was observed in case of the immunization with the adjuvant. By use of the antiserum showing the highest titer (1 : 16,000) and its purified antibodies, competitive indirect and direct ELISA's (ciELISA and cdELISA) were established, respectively. When the cross-reactivity of the antibody against fumonisin analogs was investigate by the ciELISA is was very low against B_3 (2%) but high against fumonisin B_2 (179%). The sensitivity of the ELISAs was also very high, because the detection limit for FB_1 was 0.03 ppb in ciELISA and 0.3 ppb in cdELISA. When the ELISA's were applied to the spiked corns after extraction with 75% methanol, the assay recovery of FB_1 was too unstable to assay. However, when cleanup by strong anion exchange (SAX) cartridge was introduced to remove interfering materials, the mean ELISA recovery of FB_1 from corns spiked to 3~10 ppm was found to be 34.0% and stable (mean of CV, 8.2%).

        Sampling for Data Freshness Optimization: Non-linear Age Functions

        In this paper, we study how to take samples at a datasource for improving the freshness of received data samples at aremote receiver. We use non-linear functions of the age of informationto measure data freshness, and provide a survey of non-linearage functions and their applications. The sampler design problemis studied to optimize these data freshness metrics, even when thereis a sampling rate constraint. This sampling problem is formulatedas a constrained Markov decision process (MDP) with a possiblyuncountable state space. We present a complete characterizationof the optimal solution to this MDP: The optimal sampling policyis a deterministic or randomized threshold policy, where thethreshold and the randomization probabilities are characterizedbased on the optimal objective value of the MDP and the samplingrate constraint. The optimal sampling policy can be computed bybisection search, and the curse of dimensionality is circumvented. These age optimality results hold for (i) general data freshness metricsrepresented by monotonic functions of the age of information,(ii) general service time distributions of the queueing server, (iii)both continuous-time and discrete-time sampling problems, and(iv) sampling problems both with and without the sampling rateconstraint. Numerical results suggest that the optimal samplingpolicies can be much better than zero-wait sampling and the classicuniform sampling.

        Inhibition of BMP signaling overcomes acquired resistance to cetuximab in oral squamous cell carcinomas

        <P><B>Abstract</B></P> <P>Despite expressing high levels of the epidermal growth factor receptor (EGFR), a majority of oral squamous cell carcinoma (OSCC) patients show limited response to cetuximab and ultimately develop drug resistance. However, mechanism underlying cetuximab resistance in OSCC is not clearly understood. Here, using a mouse orthotopic xenograft model of OSCC, we show that bone morphogenic protein-7-phosphorylated Smad-1, -5, -8 (BMP7-p-Smad1/5/8) signaling contributes to cetuximab resistance. Tumor cells isolated from the recurrent cetuximab-resistant xenograft models exhibited low EGFR expression but extremely high levels of p-Smad1/5/8. Treatment with the bone morphogenic protein receptor type 1 (BMPRI) inhibitor, DMH1 significantly reduced cetuximab-resistant OSCC tumor growth, and combined treatment of DMH1 and cetuximab remarkably reduced relapsed tumor growth <I>in vivo</I>. Importantly, p-Smad1/5/8 level was elevated in cetuximab-resistant patients and this correlated with poor prognosis. Collectively, our results indicate that the BMP7-p-Smad1/5/8 signaling is a key pathway to acquired cetuximab resistance, and demonstrate that combination therapy of cetuximab and a BMP signaling inhibitor as potentially a new therapeutic strategy for overcoming acquired resistance to cetuximab in OSCC.</P> <P><B>Highlights</B></P> <P> <UL> <LI> BMP7-p-Smad1/5/8 signaling contributes to cetuximab resistance in OSCC. </LI> <LI> DMHI, a BMP signaling inhibitor, reduced cetuximab-resistant OSCC tumor growth. </LI> <LI> Combined treatment of DMH1 and cetuximab significantly reduced relapsed tumor growth <I>in vivo.</I> </LI> <LI> p-Smad1/5/8 overexpression in OSCC patients was associated with poor disease free survival. </LI> </UL> </P>

        Triterpenoid saponins from Clinopodium chinense (Benth.) O. Kuntze and their biological activity

        Four new ursane-type triterpenoid saponins, clinopoursaponins A–D (1–4), six new oleanane-type triterpenoid saponins, clinopodiside VII–XII (5–10), as well as eight known triterpene analogues (11–18), were isolated from the aerial parts of Clinopodium chinense (Benth.) O. Kuntze. The structures of the new compounds were determined based on extensive spectral analyses, including 1D (1H and 13C) and 2D NMR experiments (COSY, NOESY, HSQC, 2D TOCSY, HSQC-TOCSY and HMBC), HR-ESI-MS and chemical methods. Compounds 1–18 were evaluated for their protective effects against anoxia/reoxygenation-induced apoptosis in H9c2 cells and cytotoxicities against murine mammary carcinoma cell line 4T1. Compounds 8, 9 and 18 exhibited significant protective effects, while compound 1 exhibited cytotoxic activity with IC50 value of 7.4 μm compared to 7.6 μm for the positive control 10-hydroxycamptothecin.

      • Parthenolide-Induced Apoptosis, Autophagy and Suppression of Proliferation in HepG2 Cells

        Purpose: To investigate the anticancer effects and underlying mechanisms of parthenolide on HepG2 human hepatocellular carcinoma cells. Materials and Methods: Cell viability was assessed by MTT assay and cell apoptosis through DAPI, TUNEL staining and Western blotting. Monodansylcadaverin(MDC) and AO staining were used to detect cell autophagy. Cell proliferation was assessed by Ki67 immunofluorescence staining. Results: Parthenolide induced growth inhibition in HepG2 cells. DAPI and TUNEL staining showed that parthenolide could increase the number of apoptotic nuclei, while reducing the expression of the anti-apoptotic protein Bcl-2 and elevating the expression of related proteins, like p53, Bax, cleaved caspase9 and cleaved caspase3. Parthenolide could induce autophagy in HepG2 cells and inhibited the expression of proliferation-related gene, Ki-67. Conclusions: Parthenolide can exert anti-cancer effects by inducing cell apoptosis, activating autophagy and inhibiting cell proliferation.

        Age-Optimal Multi-Flow Status Updating with Errors: A Sample-Path Approach

        In this paper, we study an age of information minimization problem in continuous-time and discrete-time status updating systems that involve multiple packet flows, multiple servers, and transmission errors. Four online scheduling policies are proposed. We develop a unifying sample-path approach and use it to show that, when the packet generation and arrival times are synchronized across the flows, the proposed policies are (near) optimal for minimizing any time-dependent, symmetric, and non-decreasing penalty function of the ages of the flows over time in a stochastic ordering sense.

      • A Study of Smartphone Based Disaster Information Reporting System under Disaster Environment

        With the development of technology and communication network, smartphones are increasingly used in emergency platforms. Under disaster environment, however, the information reporting system may confronted with poor communication conditions or low phone battery. In this study we first defined two parameters: the communication signal and the battery status. Based on the definitions, we proposed an algorithm for smartphone based reporting system under disaster environment. With the guarantee that disasters can be reported promptly and accurately and based on the analysis of smartphone power consumption, the algorithm provides power saving mechanisms to obviously increase the system working time. At last, a smartphone based disaster information reporting system (MDIRS) is designed and applied in disaster environment.

        과시소비성향, 판매촉진과 제조국 이미지가 중국여성소비자의 패션 명품 구매태도에 미치는 영향

        The purposes of this study were study intended to investigate effects of conspicuous consumption, sales promotions and images of the country of manufacture on Chinese consumers` purchasing attitudes to fashion luxury brands. The study was conducted by experimental conspicuous consumption(2)X[types of sales promotions(2) X images of the country of manufacture(2)] design by combining the 3 variables mentioned above. 312 women living in Chengdu were collected for this study. For data analysis, factor analysis, T-test, frequency analysis and ANOVA were used. The results were as follows: First, Chinese consumers` favor to fashion luxury brands: 2-way interactions, in sales promotions and images of the country of manufacture, was found for favor to fashion luxury brands. Second, Chinese consumers` purchase intent to fashion luxury brands: only the main-effects of conspicuous consumption and images of the country of manufacture was meaningfully positive. It demonstrates that only conspicuous consumption and images of the country of manufacture effected Chinese consumers` purchase intent to fashion luxury brands.
