CD70 is expressed in normal activated immune cells as well as in several types of tumors. It has been established that anti-CD70 mAb induces complement-dependent death of CD70^+ tumor cells, but how anti-CD70 mAb affects the intrinsic signaling is poorly defined. In this report, we show that ligation of CD70 expressed on EBV-transformed B cells using anti-CD70 mAb induced production of reactive oxygen species (ROS) and subsequent apoptosis. We observed an early expression of endoplasmic reticulum (ER) stress response genes that preceded the release of apoptotic molecules from the mitochondria and the cleavage of caspases. CD70-induced apoptosis was inhibited by pretreatment with the ER stress inhibitor salubrinal, ROS quencher N-acetylcysteine, and Ca^(2+) chelator BAPTA. We supposed that ROS generation might be the first event of CD70-induced apoptosis because N- acetylcysteine blocked increases of ROS and Ca^(2+), but BAPTA did not block ROS generation. We also round that CD70 stimu¬lation activated JNK and p38 MAPK. JNK inhibitor SP600125 and p38 inhibitor SB203580 effectively blocked upregulation of ER stress-related genes and cleavage of caspases. Inhibition of ROS generation completely blocked phosphorylation of JNK and p38 MAPK and induction of ER stress-related genes. Taken together, we concluded that cross-linking of CD70 on EBV-transformed B cells triggered ER stress-mediated apoptosis via ROS generation and JNK and p38 MAPK pathway activation. Our report reveals alternate mechanisms of direct apoptosis through CD70 signaling and provides data supporting CD70 as a viable target for an Ab-based therapy against EBV-related tumors.

Background: Epstein Barr virus (EBV) infected B cells are transformed into lymphoblastoid cell lines. Some researchers suggested some a few similarities between this process and carcinogenesis. We observed the expression of CD80 and CD86, co-stimulatory molecules on EBV-transformed B cells and changes of CD54 expression after stimulation of CD80 and CD86. Methods: CD80 and CD86 were stimulated using anti-CD80 and anti-CD86 monoclonal antibodies. To assess apoptosis and surface protein expression, flow cytometric analysis was performed. Intracellular signal molecules were evaluated by RT-PCR and immunoblot. Morphology and localization of proteins were examined using inverted or confocal microscope. Results: Cross-linking of CD80 and CD86 induced apoptosis and interfered with proliferation of EBV-transformed B cells, and dispersion of clumped cells. We also examined that their stimulation induced ROS accumulation and reduced CD54 expression. Interestingly, we observed that CD80 and CD86 diminished the expression of CD54 in different methods. Both CD80 and CD86 downregulated activation of focal adhesion kinase. CD80 stimulus inhibited CD54 expression through mainly RhoA inactivation, while CD86 down-regulated Ras and JNK phosphorylation. Conclusion: These results suggest that co-stimulatory CD80 and CD86 molecules, expressed EBV-transformed B cells, may play a role in apoptosis and cell adhesion.

<P>Hepatitis C virus (HCV) E2 protein binds to CD81, which is a component of the B cell co-stimulatory complex. The E2-CD81 interaction leads to B cell proliferation, protein tyrosine phosphorylation and to the hypermutation of immunoglobulin genes. Epidemiological studies have reported a high prevalence of B cell non-Hodgkin lymphoma (NHL) in HCV-positive patients, suggesting a potential association between HCV and Epstein-Barr virus (EBV) in the genesis of B lymphocyte proliferative disorders. In the present study, in order to investigate the association between EBV and HCV in B cells, we created an <I>in vitro</I> EBV-induced B cell transformation model. CD81 was gradually overexpressed during transformation by EBV. B cells isolated from HCV-positive patients grew more rapidly and clumped together earlier than B cells isolated from healthy donors following EBV infection. Pre-stimulation of CD81 expressed by resting B cells with anti-CD81 monoclonal antibody (mAb) or HCV E2 accelerated the generation of lymphoblastoid cell lines (LCLs) by EBV infection. These cells proliferated prominently through the early expression of interleukin-10 and intracellular latent membrane protein (LMP)-l. By contrast, the overexpression of CD81 on EBV-transformed B cells by anti-CD81 mAb or HCV E2 protein induced apoptosis through reactive oxygen species (ROS)-mediated mitochondrial dysfunction. These results suggest that the engagement of CD81 expressed by B cells has differential effects on B cell fate (proliferation or apoptosis) according to EBV infection and the expression level of CD81.</P>
Standardized Industrial Soy Sauce Process: Preparation of Starter Culture for Koji Production

Koji production is difficult to manage because of using spore. In this study, liquid starters were prepared for Koji production. As a liquid starter, mold isolated from spore was inoculated to submerged medium of 10% soybean and wheat (55:45). Then, this liquid starter was inoculated with 1, 5, 10, and 20% of the raw materials and spore with 2x10<SUP>8</SUP> CFU/g. The Koji process was conducted at 99% humidity for 3 days. Aspergillus oryzae produces enzymes to degrade the raw materials of Koji. The protease activities increased after 18 hours; however, in 20% inoculation, it increased immediately but decreased after 28 hours. The α-amylase activity also showed the same tendency. The protease and α-amylase activities were the highest in the 10%. However, glutaminase activity was the highest in spore inoculation. Free amino acids showed similar composition in spore and liquid starters. Except for the 1%, the others had the maximum amount of free amino acids at 28 hours and then decreased. It reached a maximum after 44 hours due to the slow growth. These results suggest liquid culture may be suitable for use as a starter in Koji production to develop industrial soy sauce processes.

The purpose of this study was to present the analysis method for each sector of POE, lighting, energy, and ventilation and the remodeling approach accordingly, including the process from analyzing the current status of the library to establishing a remodeling plan. The lighting analysis was analyzed using Climate Studio based on sunlight indicators sDA (Solar Daylight Autonomy) and ASE (Anual Sunlight Exposure) and average illumination presented by LEED v4 (2013) indoor environmental quality to analyze the light environment in various ways. The energy used ECO2-OD and Climate Studio, and the goal was to analyze basic energy requirements using ECO2-OD and to analyze energy requirements when applying natural and mechanical ventilation based on climate data by building a multi-zone air flow network energy model through Climate Studio. Ventilation was carried out through Simscale CFDs heat convective simulation to understand the air flow rate, air residence time, and air temperature inside the building, and to establish a ventilation plan applicable to remodeling alternatives based on previous lighting and energy analysis results. The project was conducted in the order of POE survey, environmental performance analysis, remodeling plan establishment, and plan analysis, but the thesis was written in the order of POE survey, remodeling plan establishment, and environmental performance analysis.

Bioabsorbable plates and screws for bone fixation system have been used owing to many advantages over conventional metallic devices. This study investigated the effects and safety of recently developed modifiable bioabsorbable plate and screw made of 100% poly (L-lactic acid) in the healing process of mandible fracture in arabbit model. In vitro extract test for cytotoxicity and bacterial reverse mutation test for genotoxicity were carried out. Any cytotoxicity caused by extract from experiment sample was not observed. In genotoxicity test, increase in the number of revertants was not observed. As an in vivo test, plates and screws were used for rigid fixation of exper-imental mandible fracture in twenty rabbits. Tissue specimens with plate were taken from the rabbits after 4, 6, 8,10 weeks and histological analysis was studied. After 4 weeks, the plate was covered by connective tissue and severe chronic active inflammation in soft tissue was observed. After 6 weeks, absorbed part in plate and new bone forma-tion around periostium and decrease in inflammation were detected. After 10 weeks, new bone formation was observed in all samples and the thickness of bone increased. The remodeling was not complete yet. This study dem-onstrates that recently developed biodegradable plate and screw system made of poly(L-lactic acid) was effective in mandible fracture regions known as high load-bearing areas. This indicates that bioabsorbable poly (L-lactic acid) plates and screws are available for most of bone fractures, although the adjustment process and long-term follow-up study are necessary for clinical application.
