We have now constructed a novel recombinant baculovirus Autographa californica nuclear polyhedrosis virus (AcNPV) producing polyhedra with Bacillus thuringiensis (Bt) CryIAc crystal protein. The recombinant polyhedra produced by the recombinant baculovirus, Btrus, in insect cells was characterized. The recombinant baculovirus has two independent transcription units in opposite orientations with two promoters, p10 or polyhedrin gene promoter each initiating transcription of either native polyhedrin or fusion protein with polyhedrin and Bt CrylAc crystal protein. Surprisingly, this recombinant baculovirus stably produced recombinant polyhedra which were nearly similar to those of wild-type AcNPV. The immunogold staining experiment showed that the recombinant polyhedra were assembled with polyhedrin and Bt Cry1Ac crystal protein, and contained virus particles. Insecticidal toxicity of recombinant polyhedra of Btrus to the faU webworm, Hyphantria cunea, was strikingly improved in comparison with the wild-type AcNPV.
폴리드나바이러스(CpBV) 유래 면역억제 유전자를 이용한 베큘로바이러스 병원력 제고 기술

Baculoviruses have been used to control some serious lepidopteran pests. However, their narrow target insect spectrum and slow efficacy are main limitations to be used in various applications. This study introduces a technique to overcome these Limitations by inhibiting insect immune defence to enhance the viral pathogenicity. Polydnaviruses are an insect DNA virus group and symbiotic to some ichneumonid and braconid endoparasitoids. Cotesia plutellae bracovirus (CpBV) is a braconid polydnavirus and encodes several immunosuppressive genes. We selected seven CpBV genes and recombined them to wild type Autographa calif arnica multiple nucleopolyhedrovirus (AcNPV). A bioassay of these seven recombinants indicated that most recombinants had similar or superior efficacy to wild type AcNPV against beet armyworm, Spodoptera exigua, and diamondback moth, Plutella xylostella. Recombinant AcNPV with CpBV-ELP was the most potent in terms of lethal time by shortening more than 2 days compared to wild type AcNPV. This recombinant was further proved in its dose-dependent pathogenicity and its efficacy by spray application on S. exigua infesting cabbage cultivated in pots. We discussed the efficacy of CpBV-ELP recombinant AcNPV in terms of suppressing antiviral activity of target insects.

The discovery of novel skeletons with a new mode of action as an insecticide is important in overcoming resistance and environmental problems associated with conventional insecticides. Recently, Insect growth regulators (IGRs) have become an effective target for mosquito control with insect selectivity and relatively low toxicity. Juvenile hormone(JH)-based IGRs including JH agonists (JHAs) and antagonists (JHANs) fatally affect the physiological regulations in insects and are effective for control the target insects. High-throughput screening is a common strategy used to identify biological active compounds with new skeletons by the screening of large compound libraries. In this study, we introduce the Lorecrezole and Penfluridol as novel JH-based insect growth regulators by the two-hybrid yeast ß-galactosidase assay method in a yeast cell using KRICT chemical library. We developed synthetic methods for Loreclezole and synthesized Loreclezole derivatives. Evaluation of their activities in-vitro tests, unlike the expectation that compounds with high JHAN activity could be obtained. This suggests that compounds with JHAN activity may also be used as novel insecticides of the IGR family

Expression of insecticidal protein by fusion product of truncated HD-1 [CryIA(a)] N-terminal and HD-73 [CryIA(c)] C-terminal fragment of Bacillus thuringiensis subsp. Kurstaki was investigated. Immunological analysis of transformants by using polyclonal antisera raised against the whole-crystal protein of HD-1 revealed that SK4 and SK5 were observed cross-reaction with polypeptides of 77-kDa and 105-kDa, respectively. Bioassay of the transformant pSK5 to Plutella maculipennis and Heliothis assulta were 96% and 97%, respectively.
