<P><B>Abstract</B></P> <P><B>Background</B></P> <P>Non-small-cell lung cancer (NSCLC) accounts for approximately 85% of all lung cancer cases and it is intrinsically resistant to anticancer drugs. Nootkatone (NKT), which is the main fragrant component of grapefruit, has been identified as a bioactive compound with a wide range of beneficial applications. NKT can activate AMP-activated protein kinase (AMPK) in liver and muscle cells, however, little is known about the role of NKT in cancer, particularly its role in NSCLC with high rates of liver kinase B1 (LKB1) and KRAS mutations.</P> <P><B>Purpose</B></P> <P>The anti-cancer activities of NKT in NSCLC A549 cells and ADR-resistant A549/ADR cells were investigated and compared to those of metformin, an AMPK activator that is used clinically as an AMPK activator.</P> <P><B>Methods</B></P> <P>Cell viability, proliferation and NKT sensitization were determined by the MTT assay. Mechanisms of NKT against anti-cancer activities including AMPK activation, cell cycle arrest, and synergistic cytotoxic effect were evaluated by Western blot analysis, and flow cytometry. In <I>in vivo</I> experiments, athymic BALB/c male nude mice were used for experiments. After the successful generation of tumor models through subcutaneous injection of A549/ADR cells, NKT and/or ADR were administered and mice were kept for weekly measurements for up to 7 weeks. The animals were then sacrificed, and the tumors were removed from all animals and weighed.</P> <P><B>Results</B></P> <P>NKT activated AMPK via LKB1-independent and CAMKK2-dependent pathways, leading to inhibition of cell growth and induction of G1 cell arrest. The effect of NKT is comparable but superior to that of metformin, an AMPK activator in clinical use. Importantly, NKT inhibited the activation of oncogenic AKT and ERK proteins, while metformin inhibited AKT but failed to impact ERK, the major oncogenic protein of NSCLC cells with KRAS mutation. The synergistic activity of NKT and ADR was more effective than that of metformin and ADR. <I>In vivo</I> data confirmed synergistic effects of NKT and ADR without systemic side effects.</P> <P><B>Conclusion</B></P> <P>We demonstrate for the first time that NKT can sensitize ADR-resistant A549/ADR cells to ADR <I>in vitro</I> and <I>in vivo.</I> Metformin, on the other hand, failed to show any synergistic effect with ADR in A549/ADR cells.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

<P><I>Background and Methods:</I> Numerous genetic studies have mapped asthma susceptibility genes to a region on chromosome 5q31-33 in several populations. This region contains a cluster of cytokines and other immune-related genes important in immune response. In the present study, to determine the genetic variations and patterns of linkage disequilibrium (LD), we resequenced all the exons and promoter regions of the 29 asthma candidate genes in the chromosome 5q31-33 region. <I>Results:</I> We identified a total of 314 genetic variants, including 289 single nucleotide polymorphisms (SNPs), 22 insertion/deletion polymorphisms and 3 microsatellites. Standardized variance data for allele frequency revealed substantial differences in SNP allele frequencies among different ethnic groups. Interestingly, significant ethnic differences were observed mainly in intron SNPs. LD block analysis using 174 common SNPs with a frequency of >10% disclosed strong LD within most candidate genes. No significant LD was observed across genes, except for one LD block (CD14-IK block). Gene-based haplotype analyses showed that 1-5 haplotype-tagging SNPs may be used to define the six or fewer common haplotypes with a frequency of >5%, regardless of the number of SNPs. <I>Conclusion:</I> Overall, our results provide useful information for the identification of immune-mediated disease genes in the chromosome 5q31-33 region, as well as valuable evidence for gene-based haplotype analysis in disease association studies.</P><P>Copyright © 2006 S. Human body has a complete left and right symmetry structure, and the left and right balance by Yin and Yang. When the balance is broken, the left and right Meridian becomes abnormal condition. Acupuncture is a kind of therapy to recover from energy unbalance of the left and right Meridian to a new balance condition. In the study, we observed the electric potential along the stomach meridian (ST) in order to verify the energy consensus phenomenon by transportation of bio-energy between operator and subject during acupuncture. The acupuncture effects on opposite meridian site were investigated by comparing the electric potentials between the right and left ST sites. Meridian electrical potentials (MEPs) between operator and subject were simultaneously generated during the acupuncture and the polarity of MEPs was opposite. The results might imply the bio-energy transportation between operator and subject. In addition, we observed three different patterns of MEPs on both ST sites which might represent the condition of the related meridians because meridians in the body are organically interconnected.

Background/Aims: Since cisplatin was widely used for the treatment of advanced gastric cancer, prevention and management of its side effects have become an important issue. Among the various side effects, neurotoxicity still remains as the major dose-limiting side effect of cisplatin. To investigate the frequency and the prognostic factors of neurotoxicity associated with cisplatin- containing chemotherapy, we performed this retrospective analysis. Methods: We analyzed clinical data of 317 patients with advanced gastric cancer treated with cisplatin-containing (20 mg/m2/day, D1-5 per cycle) chemotherapy in Korea Cancer Center Hospital from 1992 to 1996. Results: Peripheral neuropathy was developed in 44 patients(13.9%; 95% C.I, 10-17.8%): 28 patients in who grade I and 16 patients in who grade II. The incidence of peripheral neuropathy was increased as the cumulative dose of cisplatin increased. Age(p=0.0458) and cumulative dose of cisplatin(p 0.0001) were significant prognostic factors for the development of peripheral neuropathy in multivariate analysis. Conclusions: Peripheral neuropathy was a side effect of cisplatin-containing chemotherapy observed frequently in patients with advanced gastric cancer. Its development was closely related with cumulative cisplatin dose and patient's age. Once peripheral neuropathy developed clinically, it did not improve in most patients even after the administration cisplatin was discontinued.

Acupuncture in Oriental medicine has been widely used as a core therapeutic method due to its minimal side-effects and therapeutic efficacy. However, the electrical response to acupuncture stimulation (ERAS) has not been clearly studied under acupuncture conditions that might affect the efficacy of acupuncture therapy. In this study, the ERAS was objectively investigated by measuring meridian electric potentials (MEPs) when the electrical grounding conditions of the operator and subject were varied, and when the insulation conditions of acupuncture needle were varied. MEPs between Sang-geoheo (ST37) and Ha-geoheo (ST39) of the Stomach Meridian (ST) were measured by stimulating Jok-samni (ST36) with an acupuncture needle. For non-insulated acupuncture stimulation (NIAS), the average MEP peak was 148.6 ± 20.6 when neither the operator nor the subject were electrically grounded, 23.1 ± 8.8 when the subject only was electrically grounded, 348 ± 76.8 when the operator only was electrically grounded, and 19.9 ± 4.7 when both the operator and the subject were electrically grounded. The MEPs presented various magnitudes and patterns depending on the electrical grounding conditions. The MEP pattern was very similar to that of the charge and discharge of a capacitor. For insulated acupuncture stimulation (IAS), the average MEP peak was 20 ± 4 in all electrical grounding conditions, which is not a significant electric response for acupuncture stimulation. In terms of electricity, this study verified that acupuncture therapy might be affected by acupuncture conditions such as (1) the electrical grounding condition of the operator and the subject and (2) the insulation condition of the acupuncture needle.
