        Multivariate Analysis of Laser-Induced Breakdown Spectroscopy Spectra of Soil Samples

        ABSTRACT: Laser-induced breakdown spectroscopy (LIBS) is a rapid quantitative analytical technique that can be used to determine the elemental composition of numerous sample matrices, and it has been successfully applied in many types of samples. However, for chemically and physically complex soil samples, its quantitative analytical ability is controversial. Multivariate analytical techniques have great potential for analyzing the complex LIBS spectra. To demonstrate the feasibility of LIBS as an alternative technique to quantitatively analyze soil samples, the univariate and the partial least square (PLS) techniques are used to analyze the LIBS spectra of 12 soil samples and to build calibration models predicting Cu and Zn concentrations. The results show that PLS can significantly improve the analytical results compared with the univariate technique. The normalized root mean square error (NRMSE) and r of the univariate models are 16.60% and 0.71 in calibration and 18.80% and 0.62 in prediction for Cu and 18.97% and 0.62 in calibration and 22.81% and 0.45 in prediction for Zn. For the PLS models using the spectral range 300 to 350 nm, the NRMSE and r are 1.94% and 0.99 for both Cu and Zn in calibration and 7.90% and 0.94 for Cu and 8.14% and 0.94 for Zn in prediction, respectively. Compared with the univariate technique, PLS improves the NRMSE 87.53% and 87.78% in calibration and 44.47% and 53.44% in prediction for Cu and Zn, respectively. The results indicate that PLS can improve the quantitative analytical ability of LIBS for soil sample analysis.

        Evaluation of fracture toughness of welded structures has a significant influence on the structural design. However the residual stresses is redistributed while the welded structures is cut for preparing specimens. This study investigated an effect of the welding residual stress redistribution on the impact absorption energy of Charpy specimen. SA516Gr70 steel plate welded by at the flux cored arc(FCA) was cut. Specimens for Charpy impact testing were taken from the welded plate. Two material removal mechanisms (wire cutting and water jet) were used to make the specimens. Welding residual stress and redistribution residual stress were measured using the XRD (X-Ray Diffraction) method. The amount of redistribution of residual stress depends on the different material removal mechanism. Redistribution of residual stress of reduced the impact absorption energy by 15%.

        Effects of Feeding Purified Zearalenone Contaminated Diets with or without Clay Enterosorbent on Growth, Nutrient Availability, and Genital Organs in Post-weaning Female Pigs

        The effects of different levels of natural clay enterosorbent on the growth, nutrient availability, and genital organs of post-weaning female pigs fed with an addition of zearalenone (ZEA) were investigated in the study. A total of thirty-five post-weaning gilts ($L{\times}Y{\times}D$) with an average body weight of 12.36${\pm}$1.46 kg were used in the test. The gilts were raised individually in metabolism cages and fed a corn-soybean meal-whey basal diet with an addition of 0 or 1 mg/kg of ZEA for 24 d with four levels of natural clay enterosorbent added in the feed. The treatments were: i) control; ii) control+2.5 g/kg clay; iii) control+1 mg/kg ZEA; iv) control+1 mg/kg ZEA+1.25 g/kg clay; v) control+1 mg/kg ZEA+2.5 g/kg clay; vi) control+1 mg/kg ZEA+5.0 g/kg clay; vii) control+1 mg/kg ZEA +10 g/kg clay. Pigs fed diets contaminated with additional purified ZEA had significantly reduced apparent digestibility of crude protein (CP), gross energy (GE) and apparent metabolic rate of GE (ME/GE, p<0.05) without changes of net protein utilization (NPU, p>0.05). Final body weight, average daily gain (ADG), average daily feed intake (ADFI), vulva length, vulva width, vulva area, relative weights of genital organ and proliferative changes of the ovary tissues in gilts fed ZEA-contaminated diet were increased (p<0.05) compared to the gilts fed the control diet. Addition of natural clay enterosorbent in the ZEA-contaminated diet showed a positive protection effect on ZEA feeding, and the protection was increased linearly or quadratically as clay content increased. However, in pigs fed a diet with clay alone at 2.5 g/kg level there was no significant impact (p>0.05) on all the parameters as compared to the control. It is suggested that feeding ZEA at about 1.0 mg/kg for 24 days might result in a deleterious effect in pigs, and addition of 5 or 10 g clay enterosorbent per kg diet can effectively neutralize the detrimental effects of the ZEA feeding.

        Correlations among Shearing Force, Morphological Characteristic, Chemical Composition, and In situ Digestibility of Alfalfa (Medicago sativa L) Stem

        Alfalfa (Medicago sativa L) is a high-quality forage for ruminants and the main stem is the dominant morphological component contributing to the forage nutritive value in mature alfalfa forage. Shearing force, a fracturing property of plant stem, is an important indictor of forage value. The objectives of this study were to investigate the effects of morphological characteristic on shearing force, the relationship between shearing force and chemical composition, and the relationship between shearing force and in situ digestibility of alfalfa stem. The results showed that linear density (weight per unit length of stem) was more important than chemical composition in affecting shearing force. There was a positive relationship between lignin content and shearing force (r = 0.78). Correlations were not found between shearing force and other chemical components such as neutral detergent fiber (NDF), acid detergent fiber (ADF), cellulose and hemicelluloses. In situ digestibility (of dry matter and NDF) was related to shearing force. A negative correlation was found between shearing force and dry matter (DM) digestibility (r = -0.70), and there was also a negative correlation between shearing force and NDF digestibility (r = -0.87). When shearing force was standardized for stem diameter or stem linear density, the relationship between shearing force and digestibility was consistent regardless of stem diameter and stem linear density. Shearing force was significantly correlated with lignin content and in situ digestibility (of DM and NDF), and was a more direct indicator for estimating forage nutritive value related to animal performance, so it can be used to predict the forage value of alfalfa.

        Intramuscular Administration of Zinc Metallothionein to Preslaughter Stressed Pigs Improves Anti-oxidative Status and Pork Quality

        This study was conducted to determine the effects of exogenous zinc-metallothionein (Zn-MT) on anti-oxidative function and pork quality. After feeding a corn-soybean meal-based diet for two weeks, 48 pigs ($Duroc{\times}Landrace{\times}Chinese\;Black Pig$) were assigned randomly to four groups. Pigs in Group 1 were maintained under non-stress conditions, whereas pigs in Groups 2, 3 and 4 were aggressively handled for 25 min to produce stress. Pigs in Groups 1, 2, 3, and 4 received intramuscular administration of saline (control group; CON), 0 (negative control group; NCON), 0.8 (low dose group; LOW), and 1.6 (high dose group; HIGH) mg rabbit liver Zn-MT per kg body weight, respectively. Pigs were slaughtered at 3 and 6 h post-injection. Zn-MT treatment increased (p<0.05) the activities of superoxide dismutase (SOD) and glutathione-peroxidase (GSH-PX) while decreasing the concentration of malondialdehyde (MDA) in liver. These responses were greater (p<0.05) at 6 h than at 3 h post Zn-MT injection. Zn-MT treatment increased (p<0.05) hepatic SOD mRNA levels in a time and dose-dependent manner and decreased (p<0.05) serum glutamate-pyruvate transaminase and lactate dehydrogenase activities (indicators of tissue integrity). Zn-MT administration decreased (p<0.05) lactate concentration and increased (p<0.05) pH and water-holding capacity in the longissimus thorasis meat. Collectively, our results indicate that intramuscular administration of Zn-MT to pre-slaughter stressed pigs improved tissue anti-oxidative ability and meat quality.

        Variation and Correlation of Shearing Force with Feed Nutritional Characteristics of Wheat Straw

        This experiment was conducted to study the variation laws and correlations of shearing force and feed characteristics including morphological characteristic, chemical composition and in situ degradability of wheat straw. Feasibility of evaluating the nutritional value of wheat straws with shearing force values was analyzed in this study. Six hundred wheat straw plants (Jimai 22) were randomly selected and placed in a cool and ventilated place. Samples were collected in the 1st, 15th, 30th, 45th, 60th d after harvest to measure shearing force, morphological characteristic, nutritional composition. Rumen degradation of dry matter (DM), neutral detergent fiber (NDF) and acid detergent fiber (ADF) of wheat straws were determined by the nylon bags method. The results demonstrated that linear and quadratic effects of storage time on all the tested morphological characteristics were significantly correlative (p<0.01). As storage time goes on, all the tested nutrients and their rumen degradations of wheat straw was linearly (p<0.01) and quadratic (p<0.01) correlative except ADF content and rumen degradation of ADF. Significant correlations were determined in linear effect of shearing force on morphological characteristics (p<0.01), and linear density and diameter were a more sensitive predictor than stem thickness for shearing force. There were strong correlations between storage time and all the measured physical characteristics (shearing force, morphological characteristics and shearing force standardized by morphological characteristics) (p<0.01). Nutrition compositions were linearly correlative with shearing force and standardized shearing force (p<0.01). The linear correlation between rumen degradation of DM and NDF and shearing force and standardized shearing force were evident (p<0.01). In conclusion, shearing force, nutrition compositions and their rumen degradation of wheat straw were still dynamic with storage time after harvest. Correlation could be found between shearing force and nutritional characteristics of wheat straw. Nutrient content, morphological index and rumen degradation of DM and NDF could be predicted by changes in shearing force. Shearing force should be applied according to a standardized storage time when it is used to forecast the feed value of wheat straws.

        Toxicity of Mycotoxins from Contaminated Corn with or without Yeast Cell Wall Adsorbent on Broiler Chickens

        This study investigated the effects of feeds naturally contaminated with mycotoxins on growth performance, serum biochemical parameters, carcass traits, and splenic heat shock protein 70 (Hsp70) mRNA expression levels in broiler chickens. The efficacy of yeast cell wall (YCW) adsorbent in preventing mycotoxicosis was also evaluated. Three hundred 1-d-old Arbor Acres broiler chicks were randomly allotted to 3 treatments in completely randomized design for 42 d. Each treatment group had 5 replicate pens with 20 birds. The treatments were as follows: i) basal diet (control), ii) naturally contaminated diet (NCD), and iii) NCD+0.2% YCW adsorbent (NCDD). The NCD decreased average daily gain (ADG) (p<0.01) of 0 to 21 d, 22 to 42 d, and 0 to 42 d, and increased feed conversion ratio (p<0.01) of 22 to 42 d and 0 to 42 d. Both the breast meat percentage and thigh meat percentage of the NCD group were significantly higher (p<0.01) than that of the control group on d 21. The NCD group showed significantly increased levels of triglycerides (p<0.05) and cholesterol (p<0.05) on both d 21 and d 42 compared to the control group. However, the NCD significantly reduced (p<0.01) the high-density lipoprotein (HDL) on d 42 compared to controls. Compared with the NCD, supplementation with YCW significantly improved (p<0.01) the ADG of 0 to 21 d and 0 to 42 d, and increased (p<0.01) concentrations of HDL on d 42, and on d 21, and triglycerides (p<0.05) on d 21 and d 42. Supplementation with YCW reduced (p<0.01) the breast meat percentage, the thigh meat percentage, the concentrations of cholesterol (p<0.01) and the low-density lipoprotein (p<0.05) on d 21, and improved (p<0.01) the splenic Hsp70 mRNA expression levels compared with the NCD group. The results of this study indicated that feeding NCD for 42 d had adverse effects on broiler chickens, and that YCW might be beneficial in counteracting the effects of mycotoxins.
