      • RTN4 3'-UTR Insertion/Deletion Polymorphism and Susceptibility to Non-Small Cell Lung Cancer in Chinese Han Population

        Nogo protein, encoded by gene reticulon-4 (RTN4), includes three major isoforms by different splicing, named Nogo-A Nogo-B and Nogo-C. Nogo proteins play an important role in the apoptosis of cells, especially in tumor cells. RTN4 single nucleotide polymorphisms (SNPs) can influence the efficiency of transcription and translation thus being related with an individual's predisposition to cancer. The CAA insertion/deletion polymorphism (rs34917480) within RTN4 3'-UTR has been reported to be associated with many cancer types. In order to investigate the relationship between this polymorphism and susceptibility to non-small cell lung cancer (NSCLC) in the Chinese population, we conducted the present case-control study including 411 NSCLC patients and 471 unrelated healthy controls. The genotype distributions were significantly different between cases and controls (p=0.014). We found that the del allele could significantly increase NSCLC risk (ins/ins vs ins/del: p=0.007, OR 1.46, 95%CI=1.11-1.93; dominant model: p=0.004, OR 1.47, 95%CI=1.13-1.92 and allele model: p=0.008, OR 1.35, 95%CI=1.08-1.67). This association was stronger in participants over 60 years old, males and smokers. We therefore conclude that the CAA insertion/deletion polymorphism (rs34917480) contributes to non-small cell lung cancer risk in Chinese population. Age, sex and environmental exposure are also related to carcinogenic effects of rs34917480.

        Protective effects of 2',4'-dihydroxy-6'-methoxy-3',5'-dimethylchalcone against hydrogen peroxide-induced oxidative stress in hepatic L02 cell

        2',4'-Dihydroxy-6'-methoxy-3',5'-dimethylchalcone (DMC) is a chalcone isolated from the buds of Cleistocalyx operculatus (Roxb.) Merr. et Perry, and the hepatoprotective effects of DMC on Kunming mice have been studied in previous study. However, the effects of DMC on hepatocyte toxicity and corresponding mechanism remain unclear. The aim of this study was to evaluate the hepatoprotective mechanism of DMC in human hepatocytes (L02) treated with $H_2O_2$. The results demonstrated that pretreatment with DMC effectively protected $H_2O_2$-induced cell viability loss, cell membrane damage (lactate dehydrogenase, nitric oxide production and caspase-3 accumulation. Besides, DMC pretreatment increased the amount of glutathione, decreased malondialdehyde and the percentage of apoptotic L02 cells compared with only $H_2O_2$ treated group. Taken together, these results indicated that DMC had hepatoprotective effects against $H_2O_2$-induced liver injury by alleviating oxidative stress and apoptosis process in L02 cells, and DMC might be a potential candidate for the intervention of liver diseases.

        Impact of a Glyphosate-Tolerant Soybean Line on the Rhizobacteria, Revealed by Illumina MiSeq

        The global commercial cultivation of transgenic crops, including glyphosate-tolerant soybean, has increased widely in recent decades with potential impact on the environment. The bulk of previous studies showed different results on the effects of the release of transgenic plants on the soil microbial community, especially rhizosphere bacteria. In this study, comparative analyses of the bacterial communities in the rhizosphere soils and surrounding soils were performed between the glyphosate-tolerant soybean line NZL06-698 (or simply N698), containing a glyphosate-insensitive EPSPS gene, and its control cultivar Mengdou12 (or simply MD12), by a 16S ribosomal RNA gene (16S rDNA) amplicon sequencing-based Illumina MiSeq platform. No statistically significant difference was found in the overall alpha diversity of the rhizosphere bacterial communities, although the species richness and evenness of the bacteria increased in the rhizosphere of N698 compared with that of MD12. Some influence on phylogenetic diversity of the rhizosphere bacterial communities was found between N698 and MD12 by beta diversity analysis based on weighted UniFrac distance. Furthermore, the relative abundances of part rhizosphere bacterial phyla and genera, which included some nitrogen-fixing bacteria, were significantly different between N698 and MD12. Our present results indicate some impact of the glyphosate-tolerant soybean line N698 on the phylogenetic diversity of rhizosphere bacterial communities together with a significant difference in the relative abundances of part rhizosphere bacteria at different classification levels as compared with its control cultivar MD12, when a comparative analysis of surrounding soils between N698 and MD12 was used as a systematic contrast study.

      • Enhanced hierarchical model of object recognition based on a novel patch selection method in salient regions

        <P>The biologically inspired hierarchical model for object recognition, Hierarchical Model and X (HMAX), has attracted considerable attention in recent years. HMAX is robust (i.e. shift- and scale-invariant), but its use of random-patch-selection makes it sensitive to rotational deformation, which heavily limits its performance in object recognition. The main reason is that numerous randomly chosen patches are often orientation selective, thereby leading to mismatch. To address this issue, the authors propose a novel patch selection method for HMAX called saliency and keypoint-based patch selection (SKPS), which is based on a saliency (attention) mechanism and multi-scale keypoints. In contrast to the conventional random-patch-selection-based HMAX model that involves huge amounts of redundant information in feature extraction, the SKPS-based HMAX model (S-HMAX) extracts a very few features while offering promising distinctiveness. To show the effectiveness of S-HMAX, the authors apply it to object categorisation and conduct experiments on the CalTech101, TU Darmstadt, ImageNet and GRAZ01 databases. The experimental results demonstrate that S-HMAX outperforms conventional HMAX and is very comparable with existing architectures that have a similar framework.</P>

        Thyroid Hormone Reference Intervals among Healthy Individuals In Lanzhou, China

        Background: The common reference intervals (RIs) for thyroid hormones currently used in China are provided by equipment manufacturers. This study aimed to establish thyroid hormone RIs in the population of Lanzhou, a city in the subplateau region of northwest China, and compare them with previous reports and manufacturer-provided values. Methods: In total, 3,123 individuals (1,680 men, 1,443 women) from Lanzhou, an iodine-adequate area of China, perceived ashealthy were selected. The Abbott Architect analyzer was used to determine the serum concentration of thyroid hormones. The 95%RI was estimated using the 2.5th and 97.5th percentiles as the lower and upper reference limits, respectively. Results: The serum levels of thyroid-stimulating hormone (TSH), total triiodothyronine (TT3), antithyroglobulin (ATG) antibody,and antithyroid peroxidase (ATPO) antibody levels were significantly correlated with sex (P<0.05). TSH, total thyroxine (TT4), andATPO levels were significantly correlated with age (P<0.05). The serum levels of TSH, ATG, and ATPO in men were significantlylower than in women; in contrast, the serum TT3 level was significantly higher in men than in women (P<0.05). Serum TSH, TT3,TT4, and ATG levels differed across age groups (P<0.05), but no such variation was observed for ATG levels (P>0.05). The established RIs of TSH, ATG, and ATPO in this study differed between sexes (P<0.05). The thyroid hormone RIs established herein wereinconsistent with the manufacturer-provided values. Conclusion: The RIs of thyroid hormones in the healthy population of Lanzhou were inconsistent with those in the manufacturer’smanual. Validated sex-specific values are required for diagnosing thyroid diseases.

      • Computed Tomography Manifestations of Histologic Subtypes of Retroperitoneal Liposarcoma

        Objective: Liposarcoma (LPS) is the most common soft tissue sarcoma and accounts for approximately 20% of all mesenchymal malignancies, often occurring in deep soft tissue of retroperitoneal space. Accurate preoperative diagnosis is therefore necessary. We explored whether computed tomography (CT) could be used to differentiate between the various types of retroperitoneal liposarcoma (RPLS). Method: Forty-seven cases of RPLS, diagnosed surgically and histologically, were analyzed retrospectively. CT features were correlated with postoperative pathological appearance. Results: The study radiologist identified 29, 11, 2, 2 and 3 RPLS as atypical lipomatous tumor/well-differentiated liposarcoma (ALT/WDL), dedifferentiated liposarcoma (DDL), myxoid/round cell liposarcoma (ML/RCL), pleomorphic liposarcoma (PL) and mixed-type liposarcoma. Analysis of CT scans revealed the following typical findings of the different subtypes of RPLS: ALT/WDL was mainly visible as a well-delineated fatty hypodense tumor with uniform density and integrity margin; DDL was marked by the combination of focal nodular density and hypervascularity. ML/RCL, PL and mixed liposarcoma showed malignant biological behaviour and CT findings need further studies. Conclusions: CT scanning can reveal important details including internal components, margins and surrounding tissues. Based on CT findings, tumor type can be roughly evaluated and biopsy location and therapeutic scheme guided.

        Micellization behavior of binary mixtures of amino sulfonate amphoteric surfactant with different octylphenol polyoxyethylene ethers in aqueous salt solution: Both cationic and hydrophilic effects

        The effect of different valent inorganic cations and the hydrophilic group of surfactant on themicellization behavior of binary surfactant mixtures constituted by an amphoteric surfactant, sodium3-(N-dodecyl ethylenediamino)-2-hydropropyl sulfonate (C12AS), and three nonionic surfactantsoctylphenol polyoxyethylene ether (OP-n) with different numbers of oxyethylene glycol ethers (n),namely, OP-10, OP-7, and OP-4 was investigated in aqueous solution. These inorganic cations includeNa+, K+, Mg2+, Ca2+, Ba2+, Al3+ and Fe3+. Both the tensiometry and the UV–vis spectrophotometry usingpyrene as a probe were adopted to determine the critical micelle concentrations (cmc) of individual ormixed surfactants. Interaction parameters between two surfactants and other parameters were obtainedbased on the regular solution theory, the pseudophase separation model, Rubingh’s model, etc. Thermodynamic parameters including Gibbs energy of micellization and thermodynamic stability werealso calculated by both the equation proposed by Molyneux et al. and Maeda’s treatment, respectively. The effect of different valent cations on both the mixed cmc of surfactant mixtures and the molar fractionof C12AS in mixed micelle (X1) can be explained theoretically by the salting-out effect, electrostaticinteraction and steric effect. For three binary surfactant mixtures, the chain length of hydrophilic groupof surfactant results in different micellization behaviors. On adding salts, the interaction parametersshow that there exists a synergistic effect between two surfactants, and the effect increases withincreasing the ratio of valence (Z) and atom radius (R) of cation, especially, for the C12AS/OP-4 mixture. With increasing the Z/R value of cation, a deviation of X1 from the ideal value shows a divergence forthree binary surfactant mixtures because of different hydrophilicities of surfactant. Thermodynamicparameters indicate that the addition of cations with a large Z/R value can be contributive moreeffectively to the formation of stable mixed micelle than the case in the presence of cations with a smallZ/R value.
