Characterization of Fatty Acid Digestion of Beijing Fatty and Arbor Acres Chickens

The aim of this experiment was to compare the characterization of fatty acid digestion of Beijing Fatty (BF) and Arbor Acres (AA) chickens. One-day-old male AA and BF chickens were raised in the same house, and fed with the same diet. We first evaluated utilization of dietary fatty acids in chickens by the total collection procedure, and chickens were then killed to compare the abundance of intestinal mRNA expression of liver-fatty acid binding protein (L-FABP) and intestinal-fatty acid binding protein (I-FABP) by Real-time PCR, and also the pH of intestinal mucosa at 3 and 6 weeks of age. Another group of chickens were sampled at 6 weeks of age to compare the total bile acid concentration in serum, and lipase activity in contents of the small intestine. Results showed that compared to AA chickens, BF chickens had higher lipase activity in the content of the small intestine (p<0.05), greater total bile acid content in portal vein blood (p<0.05) at 6 weeks of age, lower intestinal mucosal pH at both 3 weeks (p<0.05) and 6 weeks (p<0.05) of age, and higher abundance of liver-fatty acid binding protein (L-FABP) mRNA expression in intestine tissues at 6 weeks of age (p<0.05), and higher digestibility of fatty acids at both 3 and 6 weeks (p<0.05) of age. There was no difference in I-FABP mRNA expression between AA and BF chickens at either age. Thus, BF chickens had greater fatty acids utilization than AA chickens that was associated with L-FABP, lipase activity, bile acid content and intestinal mucosal pH.
Modification of the degree of 4-O-methylation of secondary wall glucuronoxylan

Plant secondary walls are the major constituent of plant biomass targeted for second-generation biofuel production. Therefore, a thorough understanding of how secondary walls are constructed is critical for a better utilization of plant biomass for biofuel production. One of the major components in secondary walls is xylan, which is composed of a linear chain of β-1,4-linked xylosyl residues. In Arabidopsis, about 10% of xylosyl residues in xylan are substituted with glucuronic acid (GlcA), of which 60% are methylated at O-4. By contrast, all of the GlcA substituents in Populus xylan are methylated at O-4. It is not known how the degree of GlcA methylation in xylan is controlled. In this report, we demonstrated that simultaneous T-DNA knockout mutations of the three glucuronoxylan methyltransferase (GXM) genes, GXM1, GXM2, and GXM3/GXMT1, which are specifically expressed in secondary wall-forming cells, led to a complete loss of GlcA methylation in xylan in Arabidopsis stems. Overexpression of GXM2 and GXM3 in wild-type Arabidopsis resulted in an up to 5-fold increase in glucuronoxylan methyltransferase activity and as a result, up to 90% of the GlcA side chains in xylan were methylated as opposed to 60% seen in the wild type. The increased degree of GlcA methylation in xylan had no discernable effects on cell wall sugar composition and lignin monomer composition. These results reveal that the activities of GXM1, GXM2 and GXM3 are responsible for all of the GlcA methylation in xylan in Arabidopsis stems and that the degree of GlcA methylation in xylan can be modified by altered expression of GXMs.

A new Ni-Fe-based superalloy, HT-X, has been developed for applications in 700 °C advanced ultra-supercritical (A-USC) boilers. The HT-X alloy is subjected to various heat treatments. Tensile tests are conducted at room temperature (RT), 700 °C and 750 °C. Creep tests are carried out under conditions of 700 °C/300 MPa and 750 °C/150 MPa. After aging treatment, the yield strength of the HT-X alloy at RT and 750 °C is 787 MPa and 624 MPa, respectively. When additional thermal exposure at 750 °C for 5400 h is applied, the yield strength is decreased to 656 MPa at RT and 480 MPa at 700 °C. For an aged specimen, the a/2<110> dislocation shearing process occurs when tensile testing is conducted at RT and 750 °C. As the γ' precipitate size increases in the specimen that is thermally exposed at 750 °C for 5400 h, Orowan bowing is the dominant dislocation process, and stacking faults develop in the γ' precipitates at both RT and 700 °C. Dislocation slip combined with climb is the dominant mechanism under the creep testing conditions. The factors that affect the mechanical properties and deformation mechanisms are discussed.

<B>Abstract</B><P>A new approach is proposed to analyze Bremsstrahlung X-rays that are emitted from laser-produced plasmas (LPP) and are measured by a stack type spectrometer. This new method is based on a spectral tomographic reconstruction concept with the variational principle for optimization, without referring to the electron energy distribution of a plasma. This approach is applied to the analysis of some experimental data obtained at a few major laser facilities to demonstrate the applicability of the method. Slope temperatures of X-rays from LPP are determined with a two-temperature model, showing different spectral characteristics of X-rays depending on laser properties used in the experiments.</P>

<P>UV-induced DNA damage, a major risk factor for skin cancers, is primarily repaired by nucleotide excision repair (NER). UV radiation resistance-associated gene (UVRAG) is a tumor suppressor involved in autophagy. It was initially isolated as a cDNA partially complementing UV sensitivity in xeroderma pigmentosum (XP), but this was not explored further. Here we show that UVRAG plays an integral role in UV-induced DNA damage repair. It localizes to photolesions and associates with DDB1 to promote the assembly and activity of the DDB2-DDB1-Cul4A-Roc1 (CRL4(DDB2)) ubiquitin ligase complex, leading to efficient XPC recruitment and global genomic NER. UVRAG depletion decreased substrate handover to XPC and conferred UV-damage hypersensitivity. We confirmed the importance of UVRAG for UV-damage tolerance using a Drosophila model. Furthermore, increased UV-signature mutations in melanoma correlate with reduced expression of UVRAG. Our results identify UVRAG as a regulator of CRL4(DDB2)-mediated NER and suggest that its expression levels may influence melanoma predisposition.</P>

In actual traffic accidents, a phenomenon is observed where the speedometer needle jams at a certain value after the impact. Using the residual reading of the speedometer, we may infer the approximate vehicle speed while the impact occurred. Based on the function of typical speedometer, the impact characteristic and damage mechanism of vehicles, this paper describes the research of how the speedometer needle stops during accidents and its failure modes. According to the statistics of thirty specific traffic accident cases containing residual speedometer needle readings, we obtain and summarize the characteristics and rules concerning the accidents and the related information. Furthermore, a typical accident case is analyzed by in-depth level. The results reveal that for the accident vehicles with electronic speedometers, there is a strong correlation between the residual reading of the speedometer and the real vehicle impact speed. The conclusions may provide a new effective method and reference for accident reconstruction and the estimation of vehicle speed.

Two experiments were conducted to examine the effects of dietary inclusion of xylose on growth performance, nutrient digestibility and xylose absorption in the portal-drained viscera of broiler chicks. In Exp. 1, ninety male 14 day-old broilers were used to study the effects of different inclusion levels (0, 5, 10, 20 and 40%) of D-xylose in the semi-purified diets on the growth and nutrient digestibility of broilers. In Exp. 2, One hundred and eight male broilers, fed by precision feeding at 22 day-old, were used to investigate the absorption and transportation of dietary xylose in the portal-drained viscera of broiler. The results of Exp. 1 indicated that the growth of broilers was gradually decreased as the xylose level increased (p<0.01). With the xylose supplementation increased, the moisture in broiler excreta was gradually elevated (p<0.01), AME and the digestibilities of crude protein and ether extract were significantly reduced and the digestibilities of xylose and arabinose were also decreased (p<0.01). The results of Exp. 2 showed that the concentrations of ribose, xylose and galactose in serum were significantly influenced by different dietary levels of xylose (p<0.01), but there's no apparent difference among rhamnose, glucose and arabinose (p>0.05). The xylose concentration in serum was highest in Vena Cava, middle in Portal Vein and lowest in Ulnar Vein within 6 h after precision feeding. And then the xylose concentration in Portal Vein and Ulnar Vein were higher than that of Vena Cava. The concentration of ribose, xylose and galactose in serum were also significantly changed with time prolongation (p<0.01). The concentration of xylose in serum was highest in the 40% xylose treatment, middle in the 20% xylose group and lowest in the control group. The glycogen contents in liver and muscle were linearly decreased as the level of xylose increased (p<0.01). The higher the dietary level of xylose was, the lower digestibility of dietary xylose was (p<0.10). 40% xylose markedly decreased the digestibility of dietary glucose (p<0.01). In conclusion, high levels of xylose in the diets inhibited the growth and nutrient digestibility of broiler. The outputs of xylose from the hydrolyzation of wheat-based diet by xylanase should have no adverse effects on broiler performance.
