        Up-Regulation of MiR-1915 Inhibits Proliferation, Invasion, and Migration of Helicobacter pylori-Infected Gastric Cancer Cells via Targeting RAGE

        Purpose: Helicobacter pylori (HP)-infected gastric cancer (GC) is known to be a fatal malignant tumor, but the molecular mechanismsunderlying its proliferation, invasion, and migration remain far from being completely understood. Our aim in this studywas to explore miR-1915 expression and its molecular mechanisms in regulating proliferation, invasion, and migration of HP-infectedGC cells. Materials and Methods: Quantitative real-time PCR and western blot analysis were performed to determine miR-1915 and receptorfor advanced glycation end product (RAGE) expression in HP-infected GC tissues and gastritis tissues, as well as humangastric mucosal cell line GES-1 and human GC cell lines SGC-7901 and MKN45. CCK8 assay and transwell assay were performedto detect the proliferation, invasion, and migration capabilities. MiR-1915 mimics and miR-1915 inhibitor were transfected intoGC cells to determine the target relationship between miR-1915 and RAGE. Results: MiR-1915 was under-expressed, while RAGE was over-expressed in HP-infected GC tissues and GC cells. Over-expressedmiR-1915 could attenuate cellular proliferation, invasion, and migration capacities. RAGE was confirmed to be the target gene ofmiR-1915 by bioinformatics analysis and luciferase reporter assay. Moreover, HP-infected GC cellular proliferation, invasion, andmigration were inhibited after treatment with pcDNA-RAGE. Conclusion: MiR-1915 exerted tumor-suppressive effects on cellular proliferation, invasion, and migration of HP-infected GCcells via targeting RAGE, which provided an innovative target candidate for treatment of HP-infected GC.

      • Feature-preserving, Adaptive and Anisotropic Smoothing Algorithm for Triangular Mesh Models

        Despite the great success in smoothing of triangular mesh, the classical methods mostly require human interactions. In order to reduce the parameter settings, an adaptive and anisotropic smoothing algorithm for removing noise and preserving features of triangular mesh model was proposed. First calculate the expected normal vector of the model triangles, the expected normal vector should be an accurate representation of the desired movement direction and be similar to the original surface normal vector, and then we compute the value of the offset value. We develop an adaptive coefficient scheme which can avoid the parameter settings to obtain the coefficient value for each vertex. Finally, we updated every vertex’s position of the model by formula. Experimental results show that the algorithm can be adaptive to preserve sharp features and avoid shrinkages by comparing with classical methods.

        The Antihydrolytic Effect and Properties of Crosslinked Polyurethane Containing Natural Dimer Fatty Acids Building Blocks

        Four series of cross-linked polyurethanes with polyester polyols as soft segments were prepared by selfcuring of isocyanate-terminated polyurethanes. Polyester polyols herein were derived from different dimer fatty acids weight content, such as dicarboxylic acid leading to various hydrophobicity and steric hindrance. The structure of cross-linked polyurethane was characterized by Fourier transform infrared spectroscopy (FTIR). The surface properties and mechanical and thermal properties of cross-linked polyurethane were investigated using contact meter, tensile tester, differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). The results indicated that with increasing dimer fatty acid weight content from 0% to 100%, the cross-linked polyurethanes exhibited increased hydrophobicity (water contact angle increased from 91o to 96o), improved water resistance (water uptake decreased from 1.41% to 0.34%), toluene resistance (weight loss decreased from 33.2% to 22.7%), and hydrolytic resistance (weight loss decreased from 0.60% to 0.27%), enhanced mechanical properties (tensile strength increased from 1.3 to 4.8 MPa), and thermal stability (decomposition temperature of 50% weight loss increased from 425 to 462 oC).

        Analysis, Modeling and Compensation of Dynamic Imbalance Error for a Magnetically Suspended Sensitive Gyroscope

        Magnetically suspended sensitive gyroscopes (MSSGs) provide an interesting alternative for achieving precious attitude angular measurement. To effectively reduce the measurement error caused by dynamic imbalance, this paper proposes a novel compensation method based on analysis and modeling of the error for a MSSG. Firstly, the angular velocity measurement principle of the MSSG is described. Then the analytical model of dynamic imbalance error has been established by solving the complex coefficient differential dynamic equations of the rotor. The generation mechanism and changing regularity of the dynamic imbalance error have been revealed. Next, a compensation method is designed to compensate the dynamic imbalance error and improve the measurement accuracy of the MSSG. The common issues caused by dynamic imbalance can be effectively resolved by the proposed method in gyroscopes with a levitating rotor. Comparative simulation results before and after compensation have verified the effectiveness and superiority of the proposed compensation method.

        VSV-G Viral Envelope Glycoprotein Prepared from Pichia pastoris Enhances Transfection of DNA into Animal Cells

        Vesicular stomatitis virus G glycoprotein (VSV-G) has been widely used for pseudotyping retroviral, lentiviral, and artificial viral vectors. The objective of this study was to establish a potential approach for large-scale production of VSV-G. To this end, VSV-G was cloned with an N-terminal His-tag into Pichia pastoris expression vector pPIC3.5K. Three clones (Mut<sup>s</sup>) containing the VSV-G expression cassette were identified by PCR. All clones proliferated normally in expansion medium, whereas the proliferation was reduced significantly under induction conditions. VSV-G protein was detected in cell lysates by western blot analysis, and the highest expression level was observed at 96 h post induction. VSV-G could also be obtained from the condition medium of yeast protoplasts. Furthermore, VSV-G could be incorporated into Ad293 cells and was able to induce cell fusion, leading to the transfer of cytoplasmic protein. Finally, VSV-G-mediated DNA transfection was assayed by flow cytometry and luciferase measurement. Incubation of VSV-G lysate with the pGL3-control DNA complex increased the luciferase activity in Ad293 and HeLa cells by about 3-fold. Likewise, incubation of VSV-G lysate with the pCMV-DsRed DNA complex improved the transfection efficiency into Ad293 by 10% and into HeLa cells by about 1-fold. In conclusion, these results demonstrate that VSV-G could be produced from P. pastoris with biofunctionalities, demonstrating that large-scale production of the viral glycoprotein is feasible.

      • Flavonoids of Rosa roxburghii Tratt Act as Radioprotectors

        Background: To study the radioprotective effects of flavonoids from Rosa roxburghii Tratt (FRT). Materials and Methods: The radioprotective effects of FRT were investigated by examining cell viability, 30-day survival of mice and the number of colony-forming units in spleen (CFU-S) after total-body 60Co irradiation. Results: The survival rates of irradiated cells gradually increased with increasing concentrations of FRT. The survival rate was the highest at 87% with a concentration of $30{\mu}g/mL$. Pretreatment with FRT was needed to realize its radioprotective activity in mice at the dose of 60 mg/kg. With the increasing doses of 30 mg/kg, 60 mg/kg and 120 mg/kg, the numbers of CFU-S increased, and were significantly different compared with the control group. Conclusions: Pretreatment with FRT prior to irradiation resulted in significantly higher cell survival at 24 h after 5 Gy radiation, increased 30-day survival in mice after exposure to a potentially lethal dose of 8 Gy, and resulted in a higher number of CFU-S in mice after exposure to a dose of 6 Gy. These results collectively indicate that FRT is an effective radioprotective agent.

        Design and Analysis of Lorentz Force-type Magnetic Bearing Based on High Precision and Low Power Consumption

        Magnetically suspended control & sensitive gyroscope (MSCSG) is a novel type of gyroscope with the integration of attitude control and attitude angular measurement. To improve the precision and reduce the power consumption of Lorentz Force-type Magnetic Bearing (LFMB), the air gap flux density distribution of LFMB has been studied. The uniformity of air gap flux density is defined to qualify the uniform degree of the air gap flux density distribution. Considering the consumption, the average value of flux density is defined as well. Some optimal designs and analyses of LFMB are carried out by finite element simulation. The strength of the permanent magnet is taken into consideration during the machining process. To verify the design and simulation, a high-precision instrument is employed to measure the 3-dimensional magnetic flux density of LFMB. After measurement and calculation, the uniform degree of magnetic flux density distribution reaches 0.978 and the average value of the flux density is 0.482T. Experimental results show that the optimal design is effective and some useful advice can be obtained for further research.

      • Efficacy of Taxane-Based Regimens in a First-line Setting for Recurrent and/or Metastatic Chinese Patients with Esophageal Cancer

        Objective: To compare the efficacy of taxane-based regimens in the first line setting retrospectively in Chinese patients with recurrent and/or metastatic esophageal cancer. Methods: We analyzed 102 recurrent and/or metastatic esophageal cancer patients who received taxanes-based regimens in a first-line setting from January 2009 to December 2013. Sixteen (15.7%) patients were administered Nab-PTX based chemotherapy and 86 patients (84.3%) received paclitaxel (PTX) or docetaxel (DTX) based chemotherapy. Patients in the PTX/DTX group could be further divided into TP (71 patients) and TPF (15 patients) groups. Results: The objective response rate (ORR) of all patients was 20.6%, and the disease control rate (DCR) was 67.6%. The median overall survival (OS) was 10.5 months (95% CI 10.1-16.4) and the median progression-free survival (PFS) was 6.04 months (95% CI 5.09-7.91). The DCR was higher in the TPF group than the TP group (93.3% vs. 59.1%; p = 0.015 ). There were no significant differences in ORR, OS, and PFS among Nab-PTX, TPF and TP groups. Conclusions: The three regimens of Nab-PTX based, TP and TPF proved active in a first line setting of Chinese patients with recurrent and/or metastatic esophageal cancer, and should thus be regarded as alternative treatments.

        Pesticide-induced changes in fecundity and rice stripe virus transmission ability in Laodelphax striatellus (Homoptera: Delphacidae)

        The widespread use of pesticides in rice has caused the resurgence of the small brown planthopper (SBPH), Laodelphax striatellus. In addition to inciting damage by feeding on phloem cells, SBPH also functions as a vector for rice stripe virus (RSV), which can cause serious yield losses. In general, studies focused on pesticide-induced stimulation of SBPH populations have focused on the insects; little information is available on the impact of pesticides on RSV transmission by SBPH. The present study investigated the effects of two pesticides (validamycin and triazophos) on the fecundity and RSV transmission ability of SBPH. Our results demonstrated that the fecundity of non-viruliferous (naïve) or viruliferous SBPH was enhanced by exposure to triazophos at the LC20 and validamycin at 200 mg L−1. Meanwhile, the increased number of eggs produced by viruliferous females treated with pesticides was larger than those from naïve females. Exposure to validamycin caused an increase in horizontal transmission of RSV; furthermore, vertical viral transmission rates of SBPH were significantly increased by exposure to triazophos. The present study provides valuable information for understanding the mechanisms underlying the resurgence of SBPH and subsequent outbreaks of RSV.

        Synthesis of Poly(1-oxo-2-phenyltrimethylene) Catalyzed by Palladium-Rare Earth Catalyst

        Poly(1-oxo-2-phenyltrimethylene) was successfully synthesized in the presence of palladium acetate and rare earth acetate $RE(OAc)_3$ (RE=Pr, Nd, Eu, Dy, Ho) multiplex catalyst system. The final products were characterized by elemental analysis, nuclear magnetic resonance (NMR) and Fourier transform infrared spectroscopy (FTIR). The effects of various reaction parameters including molar ratio of rare earth/$Pd^{2+}$, methanol usage, 2,2-bipyridyl, p-toluensulfonic acid, and p-benzoquinone were studied in detail, and the influence of different rare earths on the copolymerization was also discussed. It is found that the rare earths have favorable co-catalytic properties and palladium-rare earth multiplex catalyst showed higher catalytic activity than palladium catalyst in the copolymerization of carbon monoxide (CO) and styrene (ST).
