        NSM00158 Specifically Disrupts the CtBP2-p300 Interaction to Reverse CtBP2-Mediated Transrepression and Prevent the Occurrence of Nonunion

        Carboxyl-terminal binding proteins (CtBPs) are transcription regulators that control gene expression in multiple cellular processes. Our recent findings indicated that overexpression of CtBP2 caused the repression of multiple bone development and differentiation genes, resulting in atrophic nonunion. Therefore, disrupting the CtBP2-associated transcriptional complex with small molecules may be an effective strategy to prevent nonunion. In the present study, we developed an in vitro screening system in yeast cells to identify small molecules capable of disrupting the CtBP2-p300 interaction. Herein, we focus our studies on revealing the in vitro and in vivo effects of a small molecule NSM00158, which showed the strongest inhibition of the CtBP2-p300 interaction in vitro. Our results indicated that NSM00158 could specifically disrupt CtBP2 function and cause the disassociation of the CtBP2-p300-Runx2 complex. The impairment of this complex led to failed binding of Runx2 to its downstream targets, causing their upregulation. Using a mouse fracture model, we evaluated the in vivo effect of NSM00158 on preventing nonunion. Consistent with the in vitro results, the NSM00158 treatment resulted in the upregulation of Runx2 downstream targets. Importantly, we found that the administration of NSM00158 could prevent the occurrence of nonunion. Our results suggest that NSM00158 represents a new potential compound to prevent the occurrence of nonunion by disrupting CtBP2 function and impairing the assembly of the CtBP2-p300-Runx2 transcriptional complex.

        Verification of SARAX code system in the reactor core transient calculation based on the simplifi ed EBRII benchmark

        This paper shows the verification work of SARAX code system in the reactor core transient calculationbased on the simplified EBR-II Benchmark. The SARAX code system is an analysis package developed byXi'an Jiaotong University and aims at the advanced reactor R&D. In this work, a neutron-photon coupledpower calculation model and a spatial-dependent reactivity feedback model were introduced. To verifythe models used in SARAX, the EBR-II SHRT-45R test was simplified to an ULOF transient with an inputflowrate change curve by fitting from reference. With the neutron-photon coupled power calculationmodel, SARAX gave close results in both power fraction and peak power prediction to the referenceresults. The location of the hottest assembly from SARAX and reference are the same and the relativepower deviation of the hottest assembly is 2.6%. As for transient analysis, compared with experimentalresults and other calculated results, SARAX presents coincident results both in trend and absolute value. The minimum value of core net reactivity during the transient agreed well with the reported results,which ranged from 0.3$ to 0.35$. The results verify the models in SARAX, which are correct and ableto simulate the in-core transient with reliable accuracy.

        Sport Policy Development in China: Legacies of Beijing’s 2008 Summer Olympic Games and 2022 Winter Olympic Games

        The aim of this article is to explore Olympic-led sport policy changes (as part of Olympic legacy) for China triggered by the 2008 Summer Olympics and the 2022 Winter Olympic Games. Although there has been a burgeoning of research interest in analysing Olympic-triggered changes and legacies, with focus on various areas such as economic, sociocultural, and environmental issues, little is known about the changes that the hosting of the Olympics Games stimulates in a host nation’s sport policy. Drawing from policy document analysis, the paper reveals that the two Olympic Games collectively helped to expand the role and value of sport in China and to elevate the status of mass sport. In terms of policy, China’s two bidding experiences led to a streamlining of policy making and legacy planning processes for sporting mega events. The paper ends with a discussion of the implications associated with strategic policy planning for future Olympics hosts.

        Hepatocellular carcinoma prediction model performance decreases with long-term antiviral therapy in chronic hepatitis B patients

        Background/Aims: Existing hepatocellular carcinoma (HCC) prediction models are derived mainly from pretreatment or early on-treatment parameters. We reassessed the dynamic changes in the performance of 17 HCC models in patients with chronic hepatitis B (CHB) during long-term antiviral therapy (AVT). Methods: Among 987 CHB patients administered long-term entecavir therapy, 660 patients had 8 years of follow-up data. Model scores were calculated using on-treatment values at 2.5, 3, 3.5, 4, 4.5, and 5 years of AVT to predict threeyear HCC occurrence. Model performance was assessed with the area under the receiver operating curve (AUROC). The original model cutoffs to distinguish different levels of HCC risk were evaluated by the log-rank test. Results: The AUROCs of the 17 HCC models varied from 0.51 to 0.78 when using on-treatment scores from years 2.5 to 5. Models with a cirrhosis variable showed numerically higher AUROCs (pooled at 0.65–0.73 for treated, untreated, or mixed treatment models) than models without (treated or mixed models: 0.61–0.68; untreated models: 0.51–0.59). Stratification into low, intermediate, and high-risk levels using the original cutoff values could no longer reflect the true HCC incidence using scores after 3.5 years of AVT for models without cirrhosis and after 4 years of AVT for models with cirrhosis. Conclusions: The performance of existing HCC prediction models, especially models without the cirrhosis variable, decreased in CHB patients on long-term AVT. The optimization of existing models or the development of novel models for better HCC prediction during long-term AVT is warranted.

        Launch Vehicle software has played an important role in Launch Vehicle system. However, the reliability assessment of Launch Vehicle software is still a hard problem due to the complexity of Launch Vehicle software. Failure prediction can be an effective approach of Launch Vehicle reliability evaluation, whereas failure prediction of software has not yet been fully explored. In this paper, a Markov Bayesian Network model for Launch Vehicle software failure prediction (MBNG) is proposed. In MBNG, unique features of Launch Vehicle software is considered as an important part in the modeling process, which improves the effectiveness of this novel model. Experiments are conducted to verify the effectiveness of MBNG model and compare its performance with classic models.

        Inhibition of Invasion and Metastasis of MHCC97H Cells by Expression of Snake Venom Cystatin through Reduction of Proteinases Activity and Epithelial-Mesenchymal Transition

        Snake venom cystatin (sv-cystatin) is a member of the cystatin family of cysteine protease inhibitors. To further evaluate the possibility of sv-cystatin in cancer therapy, this study examined the effects of sv-cystatin on the invasion and metastasis of liver cancer cells (MHCC97H) in vitro and in vivo as well as the underlying mechanism. sv-cystatin cDNA was transfected into MHCC97H cells and the anti-invasion and antimetastasis effects of sv-cystatin were determined using migration and matrigel invasion assays and a lung-metastasis mice model. The results suggest that sv-cyst clone (sv-cystatin expression in MHCC97H cells) delayed the invasion and metastasis in vitro and in vivo compared to the parental, mock and si-sv-cyst clone cells (inhibited sv-cystatin expression by siRNA). The decreased activities of cathepsin B, MMP-2 and MMP-9 and EMT change index including higher E-cadherin, lower N-cadherin and decreased Twist activity were observed in the sv-cyst clone, which contributes to the change in invasion and metastasis ability of MHCC97H cells. This study provides evidence that expression of the sv-cystatin gene in MHCC97H cells inhibits tumor cell invasion and metastasis through the reduction of the proteinases activity and Epithelial-Mesenchymal Transition (EMT), which might contribute to the anticancer research of the sv-cystatin protein.

        Unsupervised Single Moving Object Detection Based on Coarse-to-Fine Segmentation

        An efficient and effective unsupervised single moving object detection framework is presented in this paper. Given the sparsely labelled trajectory points, we adopt a coarse-to-fine strategy to detect and segment the foreground from the background. The superpixel level coarse segmentation reduces the complexity of subsequent processing, and the pixel level refinement improves the segmentation accuracy. A distance measurement is devised in the coarse segmentation stage to measure the similarities between generated superpixels, which can then be used for clustering. Moreover, a Quadmap is introduced to facilitate the refinement in the fine segmentation stage. According to the experiments, our algorithm is effective and efficient, and favorable results can be achieved compared with state-of-the-art methods.

        Size Aware Correlation Filter Tracking with Adaptive Aspect Ratio Estimation

        Correlation Filter-based Trackers (CFTs) gained popularity recently for their effectiveness and efficiency. To deal with the size changes of the target which may degenerate the tracking performance, scale estimation has been introduced in existing CFTs. However, the variations of the aspect ratio were usually neglected, which also influence the size of the target. In this paper, Size Aware Correlation Filter Trackers (SACFTs) are proposed to deal with this problem. The SACFTs not only determine the translation and scale variations, but also take the aspect ratio changes into consideration, thus a better estimation of the size of the target can be realized, which improves the overall tracking performance. And competing results can be achieved compared with state-of-the-art methods according to the experiments conducted on two large scale datasets.

        Huber Second-order Variable Structure Predictive Filter for Satellites Attitude Estimation

        This work presents a novel filtering approach to the high-accuracy attitude estimation problem of satellites. A new second-order variable structure predictive filter is first designed with the measurement errors and theirdifference reduced. The key feature of this filter is that the noise handled is not constrained to be the Gaussianwhite noise. Hence, it is a new solution to filtering problem in the presence of modeling errors or heavy-tailednoise. Then, the robust version of the preceding filter is developed by using the Huber technique. This robustfilter can ensure great robustness and perfect estimation accuracy/precision for the satellite attitude. The Lyapunovstability analysis proves that the measurement error and its difference can be stabilized into a small set with a fasterrate of convergence. The effectiveness of the presented attitude estimation filters is validated via simulation bycomparing with the traditional cubature Kalman filter.
