        SERPINE1 as an Independent Prognostic Marker and Therapeutic Target for Nicotine-Related Oral Carcinoma

        Objectives. Nicotine is an ingredient of tobacco, and exposure to nicotine increases the risks of various cancers, includingoral cancer. Previous studies have focused on the addictive properties of nicotine, but its carcinogenic mechanism hasrarely been studied. We aimed to explore the key genes in the process through which nicotine promotes the occur-rence and development of oral cancer via data mining and experimental verification. Methods. This study involved three parts. First, key genes related to nicotine-related oral cancer were screened throughdata mining; second, the expression and clinical significance of a key gene in oral cancer tissues were verified by bio-informatics. Finally, the expression and clinical significance of the key gene in oral cancer were histologically investi-gated, and the effects of its expression on cell proliferation, invasion, and drug resistance were cytologically assessed. Results. SERPINE1 was identified as the key gene, which was upregulated in nicotine-treated oral cells and may be an in-dependent prognostic factor for oral cancer. SERPINE1 was enriched in various pathways, such as the tumor necro-sis factor and apelin pathways, and was related to the infiltration of macrophages, CD4+T cells, and CD8+T cells. Overexpression of SERPINE1 was associated with N staging and may be involved in hypoxia, angiogenesis, and me-tastasis. Knockdown of SERPINE1 in oral cancer cells resulted in weakened cell proliferation and invasion abilityand increased sensitivity to bleomycin and docetaxel. Conclusion. This study revealed SERPINE1 as a key gene for nicotine-related oral cancer, indicating that SERPINE1 maybe a novel prognostic indicator and therapeutic target for oral carcinoma.

      • Research on Dynamic Evaluation Method of Slope Stability Based on Monitoring Data

        Due to the frequent occurrence of slope instability caused by disasters in recent years, it is particularly important for monitoring the slope disasters and the dynamic evaluation of slope stability. The monitioring data is the most intuitive factor to reflect the stability of the slope. Therefore, it is more urgent to directly evaluate the stability of the slope through the monitoring data of the slope disaster. In this paper, based on BP neural network, the slope s tability dynamic evaluation system-DESlopeStability is developed by MATLAB. Then, the accuracy of the DESlopeStability is verified by the numerical simulation of ABAQUS, which is a three-dimensional finite element software. Finally, this system is applied to the Jietai Temple landslide project in Beijing. Through the above research, the BP neural network can find the nonlinear mapping between the monitoring data and the stability coefficient of the slope based on a large amount of data, and the DESlopeStability system meets the actual engineering requirements and it can be used as an auxiliary means to judge the stability of the slope.

        Phase structure, electrical properties, and stability of 0.96(K0.48Na0.52)1xLixNbO3e0.04Bi0.5Na0.5ZrO3 lead-free piezoceramics

        In this work, 0.96(K0.48Na0.52)1xLixNbO3e0.04Bi0.5Na0.5ZrO3 lead-free piezoceramics were prepared using the conventional solid state reaction method in order to attain both a high TC and an enhanced d33. The effect of Li content on their phase structure, electrical properties, and stability was systematically investigated. These results indicate that adding Li could result in the increase of TC and the decrease of TOeT. Here we obtain the ceramics with an orthogonaletetragonal phase boundary in the composition of x ¼ 0.02, and then enhanced piezoelectric properties and a high Curie temperature (i.e., d33 w 255 pC/N, kp w 41%, and TC w 360 C) were observed, showing the realization of our objective. In addition, a good stability of piezoelectric and ferroelectric properties has been shown in such a material system.

        Local Bearing Capacity of Steel Beams with Corrugated Webs

        Nine specimens were tested to investigate the local bearing capacity of steel beams with corrugated webs. Then, the infl uence of diff erent load distribution widths on local bearing capacity was studied. Results showed that the failure modes of the specimens were web yielding and changed from brittle failure to plastic failure at increased distribution width. The ultimate bearing capacities were higher than those of steel beams with plate webs. Meanwhile, the loading course, ultimate load and failure modes were closely simulated through the fi nite element method, and parameter analysis was performed, the result of which verifi ed the decreased capacity of the sparse waveforms. The design expression of the local bearing capacity under diff erent load distribution widths were obtained on the basis of these results and applied to crane beams with corrugated webs. The calculated results compared with fi nite element analysis and experimental results suggested the correctness and reliability of the design expression.

        Study on Damage of Shallow Foundation Building Caused by Surface Curvature Deformation in Coal Mining Area

        Coal mining can lead to various forms of surface deformation, the damage caused by curvature deformation is the most extensive, which has become the key to restricting the development and utilization of coal resources under villages. Therefore, it is very important to master the deformation and destruction mechanism of buildings under surface curvature deformation for the rational exploitation of coal resources under villages. In this paper, the deformation and damage of buildings caused by curvature deformation are studied with theoretical analysis and numerical simulation, aiming at the shallow foundation building in rural China, and the following objectives are achieved: 1) Based on the theory of elastic foundation beam, the longitudinal and transverse walls are considered as a whole and analyzed, and a mathematical model of the cooperative deformation in building walls under surface curvature deformation is established. 2) The failure mechanism of masonry buildings is analyzed, including the distribution of internal stress and the form of wall cracks. 3) The influence of various factors on the internal stress changes of buildings is discussed. 4) The results of numerical simulation of wall stress distribution are similar to theoretical analysis, and the influence of doors and windows on the additional stress distribution is analyzed emphatically. This study can provide important guidance for the optimization of coal mining design under villages and the protection of village buildings.

        A 32.2 GHz Full Adder Designed with TLE Method in a InP DHBT Technology

        Ultra-high-speed full adder is the bottleneck in a tens of GHz Direct digital synthesizer (DDS). In this paper, a 32.2 GHz, 1bit full adder in a 0.7 μm InP double hetero-junction bipolar transistor (DHBT) technology is presented. In such a high-speed circuit, signal integrity is a crucial issue. Therefore, a transmission line equivalent (TLE) method is proposed. With the TLE method, the design of the full adder could be simplified with good accuracy. The synchronous latch is combined with adding operation to improve the calculation speed. A single-level parallel-gated circuit is designed using majority decision algorithm to reduce power consumption. Measurement results show that the maximum clock frequency of the full adder is 32.2-GHz, and the overall power consumption is 350 mW. The full adder is successfully adopted in a 17 GHz, 8 bit DDS which can synthesize sin-wave outputs from 66.41 MHz to 8.5 GHz in 66.41 MHz steps with an average Spurious-Free Dynamic Range (SFDR) of -18.1 dBc.

        <P>We perform a systematic numerical study to characterize the tradeoff between the plasmonic enhancement and optical loss in periodically aligned, silicon nanowire (SiNW) arrays integrated with a silver back reflector (Ag BR). Optimizing the embedded depth of the wire bottoms into a silver reflector achieved a highly efficient SiNW solar cell. Compared to the SiNW solar cell employing a flat back reflector, the embedded depth of ~20 nm resulted in the relative increase of ~5% in ultimate solar cell efficiency.</P>

        Identification of copy number variations using high density whole-genome single nucleotide polymorphism markers in Chinese Dongxiang spotted pigs

        Objective: Copy number variations (CNVs) are a major source of genetic diversity complementary to single nucleotide polymorphism (SNP) in animals. The aim of the study was to perform a comprehensive genomic analysis of CNVs based on high density whole-genome SNP markers in Chinese Dongxiang spotted pigs. Methods: We used customized Affymetrix Axiom Pig1.4M array plates containing 1.4 million SNPs and the PennCNV algorithm to identify porcine CNVs on autosomes in Chinese Dongxiang spotted pigs. Then, the next generation sequence data was used to confirm the detected CNVs. Next, functional analysis was performed for gene contents in copy number variation regions (CNVRs). In addition, we compared the identified CNVRs with those reported ones and quantitative trait loci (QTL) in the pig QTL database. Results: We identified 871 putative CNVs belonging to 2,221 CNVRs on 17 autosomes. We further discarded CNVRs that were detected only in one individual, leaving us 166 CNVRs in total. The 166 CNVRs ranged from 2.89 kb to 617.53 kb with a mean value of 93.65 kb and a genome coverage of 15.55 Mb, corresponding to 0.58% of the pig genome. A total of 119 (71.69%) of the identified CNVRs were confirmed by next generation sequence data. Moreover, functional annotation showed that these CNVRs are involved in a variety of molecular functions. More than half (56.63%) of the CNVRs (n = 94) have been reported in previous studies, while 72 CNVRs are reported for the first time. In addition, 162 (97.59%) CNVRs were found to overlap with 2,765 previously reported QTLs affecting 378 phenotypic traits. Conclusion: The findings improve the catalog of pig CNVs and provide insights and novel molecular markers for further genetic analyses of Chinese indigenous pigs.

        Identification of loci affecting teat number by genome-wide association studies on three pig populations

        Objective: Three genome-wide association studies (GWAS) and a meta-analysis of GWAS were conducted to explore the genetic mechanisms underlying variation in pig teat number. Methods: We performed three GWAS and a meta-analysis for teat number on three pig populations, including a White Duroc${\times}$Erhualian $F_2$ resource population (n = 1,743), a Chinese Erhualian pig population (n = 320) and a Chinese Sutai pig population (n = 383). Results: We detected 24 single nucleotide polymorphisms (SNPs) that surpassed the genome-wide significant level on Sus Scrofa chromosomes (SSC) 1, 7, and 12 in the $F_2$ resource population, corresponding to four loci for pig teat number. We highlighted vertnin (VRTN) and lysine demethylase 6B (KDM6B) as two interesting candidate genes at the loci on SSC7 and SSC12. No significant associated SNPs were identified in the meta-analysis of GWAS. Conclusion: The results verified the complex genetic architecture of pig teat number. The causative variants for teat number may be different in the three populations
