        How to evaluate the adequacy of staging for nodal-negative epithelial ovarian cancer? Use of nodal staging score

        Objective: No guideline has been provided to assess the minimal number of lymph nodes (LNs) that should be dissected for accurate staging in patients with epithelial ovarian cancer (EOC). The aim of the study was to develop a nodal staging score (NSS) as an index to assess whether a pathologic (p)N0 EOC patient is indeed free of a nodal disease. Methods: A total of 16,361 EOC patients staged I–III between 2004 and 2013 were identified from the Surveillance, Epidemiology and End Result database. With a β-binomial model, NSS was calculated to assess the probability of true-negative findings of LN status. Results: With an increased number of LNs examined, the probability of missing a nodal disease decreased and varied among different pT stages. Given 1 LN examined, an NSS of 93.76% calculated could ensure a high adequacy of nodal-negative classification for pT1N0 EOC patients. For pT2N0 patients, 5 LNs examined could guarantee an NSS of 90% for adequate staging. Likewise, 11 and 29 LNs examined in pT3N0 patients could maintain NSS at the level of 80% and 90%, respectively. Our study suggested the optimal number of LNs that could be examined and stratified by the pT stages for EOC patients based on this statistical model derived from large pathologic data of clinical surgery patients. Conclusion: NSS, as an auxiliary tool, not only could assist the International Federation of Gynecology and Obstetrics staging more precisely, but also would provide a statistical basis for postoperative evaluation for further clinical decision-making.

        Coloring mechanism of celadon glazes from Yaozhou kiln

        The Five Dynasty sky-green, Song dynasty olive-green and Jin dynasty moon-white glazes are three typical glazes of theYaozhou celadon. In this work, the coloring mechanism of the three celadon glazes were compared and analyzed by usingcolorimeter, EDXRF, XRD, XPS and Raman spectrometer. The results indicate that the content of Fe2O3 is proportional tothe depth of the glaze colors. The Raman Ip value indicates that the firing temperature of the Song dynasty olive-green glazewas the highest, followed by the Five Dynasty, and then the Jin dynasty. The lower firing temperature and dense bubbles areimportant causes of the high opacification of the Jin moon-white glaze. The ratio of Fe2+ to Fe3+ in the glaze is directlyproportional to the b* value, which is the main reason for why the Yao celadon glaze colors are yellow-green in Song, butbluish in the Five Dynasty and Jin dynasty.

        Micro gadolinium oxide dispersed flexible composites developed for the shielding of thermal neutron/gamma rays

        In this study, a series of flexible neutron/gamma shielding composites are fabricated through the doping of Gd2O3 into the matrix of SEBS with (MGd2O3: MSEBS) % from 5% to 100%. Neutron transmittance test shows an exponential attenuation with the increase of areal density of Gd, in which the transmittance T ranges from 59.1440% to 35.3026%, with standard deviation less than 2.2743%, mass attenuation coefficient mm from 0.3194 cm2 /g to 0.4999 cm2 /g, and half value layer-HVL value from 2.4530 mm to 1.1313 mm. Shielding efficiency of the Gd2O3/SEBS composites is basically improved in comparison with that of B4C/SEBS. The transmittance T, mass/linear attenuation coefficient mm and m, HVL and effective atomic number Zeff for the shielding of g rays (39 keV, 59 keV and 122 keV) are measured and calculated with XCOM as well as MCX programs. Finally, plots of the three dimensional relationships between transmittance, doping amount and thickness are provided to the guidance for engineering shielding design. In summary, the Gd2O3/SEBS composite is proved to be an effective flexible neutron/low energy g rays shielding material, which could be of potential applications in the field of nuclear technology and nuclear engineering.

        FBXL20-mediated Vps34 ubiquitination as a p53 controlled checkpoint in regulating autophagy and receptor degradation

        <P>Vps34, the catalytic subunit in the class III phosphatidylinositol 3 kinase complexes, mediates the production of PtdIns3P, a key intracellular lipid involved in regulating autophagy and receptor degradation. Xiao et al. show that DNA damage-activated mitotic arrest and CDK activation lead to the phosphorylation of Vps34. This provides a signal to promote Vps34 ubiquitination and proteasomal degradation mediated by FBXL20, leading to inhibition of autophagy and receptor endocytosis. Importantly, they also find that expression of FBXL20 is regulated by p53-dependent transcription.</P><P>Vacuolar protein-sorting 34 (Vps34), the catalytic subunit in the class III PtdIns3 (phosphatidylinositol 3) kinase complexes, mediates the production of PtdIns3P, a key intracellular lipid involved in regulating autophagy and receptor degradation. However, the signal transduction pathways by which extracellular signals regulate Vps34 complexes and the downstream cellular mechanisms are not well understood. Here we show that DNA damage-activated mitotic arrest and CDK activation lead to the phosphorylation of Vps34, which provides a signal to promote its ubiquitination and proteasomal degradation mediated by FBXL20 (an F-box protein) and the associated Skp1 (S-phase kinase-associated protein-1)–Cullin1 complex, leading to inhibition of autophagy and receptor endocytosis. Furthermore, we show that the expression of FBXL20 is regulated by p53-dependent transcription. Our study provides a molecular pathway by which DNA damage regulates Vps34 complexes and its downstream mechanisms, including autophagy and receptor endocytosis, through SCF (Skp1–Cul1–F-box)-mediated ubiquitination and degradation. Since the expression of FBXL20 is regulated by p53-dependent transcription, the control of Vps34 ubiquitination and proteasomal degradation by FBXL20 and the associated SCF complex expression provides a novel checkpoint for p53 to regulate autophagy and receptor degradation in DNA damage response.</P>
