        Illuminating the hepatotoxic mechanism of norcantharidin in rats using metabolomics analysis

        Norcantharidin (NCTD) has multiple antitumor effects. However, NCTD can induce significant hepatotoxicity and the mechanism of hepatotoxicity is not clear for now.This study aimed to explore the hepatotoxicity of NCTD in rat by ultra-performance liquid chromatography (UPLC) quadrupole time-of-flight (Q-TOF)-MS (UPLC/Q-TOF-MS) metabolomics.Serum biochemical indices including alanine aminotransferase (ALT) and total bilirubin (T-BIL) were significantly increased. Histopathological and ultrastructure results revealed that hepatocytes were damaged. Furthermore, the metabolomics results showed that 11 metabolites in serum and 8 metabolites in liver were differential metabolites for NCTD hepatotoxicity. Four metabolic pathways including the sphingolipid metabolism, purine metabolism, arachidonic acid metabolism, and glycerophospholipid metabolism were the key metabolic pathways related to NCTD hepatotoxicity.The metabolomics analysis in this study reveal new clues on the hepatotoxicity mechanism of NCTD in rats. These findings have potential applications in the toxicity study of NCTD.

        Clinicopathological Features of Low-Grade Thyroid-like Nasopharyngeal Papillary Adenocarcinoma

        Purpose Primary low-grade thyroid-like papillary adenocarcinomas are extremely rare neoplasms that generally originate in the nasopharynx. Here, we describe a novel case of a 15-year-old Chinese girl who was diagnosed with low-grade thyroid-like papillary adenocarcinoma, including a brief review of the literature to reveal the clinicopathological features of lowgrade thyroid-like nasopharyngeal papillary adenocarcinoma. Materials and Methods Immunohistochemistry was used to evaluate the expression of pan-cytokeratin (CKpan), cytokeratin (CK) 7, thyroid transcription factor 1 (TTF-1), vimentin, epithelial membrane antigen (EMA), thyroglobulin, CD15, S100, P40, CK20, CDX-2, glial fibrillary acidic protein (GFAP), and Ki-67. Additionally, in situ hybridization investigation was utilized to identify the presence of small Epstein-Barr virus (EBV)–encoded RNA. Results Histopathological analysis revealed florid proliferation of papillary structures lined by columnar epithelial cells with fibrovascular cores. Immunohistochemically, the neoplastic cells were positive for CKpan, CK7, TTF-1, vimentin, and EMA, but negative for thyroglobulin, CD15, S100, P40, CK20, CDX-2, and GFAP. The Ki-67–labeling index reached 5% in the most concentrated spot. In situ hybridization for EBV was negative. Conclusion Due to the distinct rarity of low-grade thyroid-like papillary adenocarcinomas with a favorable clinical outcome, a nationwide effort to raise public awareness of this neoplasm is required.

      • Immunity : Inflammatory T Cell Responses Rely on Amino Acid Transporter ASCT2 Facilitation of Glutamine Uptake and mTORC1 Kinase Activation

        Glutamine has been implicated as an immunomodulatory nutrient, but how glutamine uptake is mediated during T cell activation is poorly understood. We have shown that naive T cell activation is coupled with rapid glutamine uptake, which depended on the amino acid transporter ASCT2. ASCT2 deficiency impaired the induction of T helper 1 (Th1) and Th17 cells and attenuated inflammatory T cell responses in mouse models of immunity and autoimmunity. Mechanistically, ASCT2 was required for T cell receptor (TCR)-stimulated activation of the metabolic kinase mTORC1. We have further shown that TCR-stimulated glutamine uptake and mTORC1 activation also required a TCR signaling complex composed of the scaffold protein CARMA1, the adaptor molecule BCL10, and the paracaspase MALT1. This function was independent of IKK kinase, a major downstream target of the CARMA1 complex. These findings highlight a mechanism of T cell activation involving ASCT2-dependent integration of the TCR signal and a metabolic signaling pathway.

        Characterization of a New Isolate of Chilli ringspot virus in Yunnan, China

        Virus diseases are the major limiting factors of chilli production in China. A virus isolate of chilli ringspot virus-Yunnan (ChiRSV-YN) from chilli (Capsicum annuum L.) showing mild mosaic, leaf crinkling, ringspot, and vein banding symptoms was recently identified in Yunnan, China. The electron microscopy confirmed the virus as a typical potyvirus having flexuous filamentous particles ca. 780 nm in length. Several chilli and tobacco varieties were tested through mechanical inoculation and found to be susceptible to the virus isolate ChiRSV-YN. The viral genome was sequenced through RNA sequencing. The complete genomic RNA of the isolate (GenBank Acc. No. KX258620) consisted of 9,652 nucleotides (nt) excluding the poly (A) tail at the 3ʹ end. The isolate contains a large ORF which encodes a polyprotein of 3,086 amino acids (aa) with Mr. of 349.21 kDa. It had a typical genomic organization of potyviruses. The complete nucleotide and polyprotein sequences of ChiRSV-YN had 87.6 and 93.2% identities with the ChiRSV-HN/14, respectively. The result showed ChiRSV-YN is a new isolate of ChiRSV and it is the first full-length genome characterization of ChiRSV in Yunnan, China. Understanding of the characteristics of this virus isolate might provide the basis for disease diagnosis and control in the future.

        Air Embolism during Upper Endoscopy: A Case Report

        Air embolism is a rare complication of upper endoscopy and potentially causes life-threatening events. A 67-year-old man with ahistory of surgery of cardiac carcinoma and pancreatic neuroendocrine tumor underwent painless upper endoscopy because oftarry stools. During the procedure, air embolism developed, which caused decreased pulse oxygen saturation and delayed sedationrecovery. He recovered with some weakness of the left upper limb in the intensive care unit without hyperbaric oxygen therapy. Theetiology, clinical manifestations, and treatments of air embolism are discussed based on the literature reports. Although air embolism isuncommon in endoscopic examinations, the patients’ outcomes could be improved if clinicians are alert to this potential complication,and promptly start proper diagnostic and therapeutic measures.
