      • Simultaneous Blockage of Epidermal Growth Factor Receptor and Cyclooxygenase-2 in a Human Xenotransplanted Lung Cancer Model

        The effects of erlotinib combined with celecoxib in a lung cancer xenograft model were here explored with a focus on possible mechanisms. A xenotransplanted lung cancer model was established in nude mice using the human lung cancer cell A549 cell line and animals demonstrating tumour growth were randomly divided into four groups: control, erlotinib, celecoxib and combined (erotinib and celecoxib). The tumor major axis and short diameter were measured twice a week and after 40 days tissues were collected for immunohistochemical analyses of Bcl-2 and Bax positive cells and Western-blotting analyses for the epidermal growth factor recepto (EGFR), P-EGFR, and cyclooxygenase-2 (COX-2). Tumor size in the combined group was smaller than in the others (p<0.01) and the percentage of Bcl-2 positive cells was fewer in most cases (p<0.01), while that of Bax positive cells was greater than in the erlotinib and celecoxib groups (P>0.05). Western blotting showed decreased expression of P-EGFR and COX-2 with both erlotinib and celecoxib treatments, but most pronouncedly in the combined group (P<0.05). Simultaneous blockage of the EGFR and COX-2 signal pathways exerted stronger growth effects in our human xenotransplanted lung cancer model than inhibition of either pathway alone. The anti-tumor effects were accompanied by synergetic inhibition of tumor cell apoptosis, activation of p-EGFR and expression of COX-2.

        魏晉寫槐骈赋研究

        홰나무를 소재로 한 변문은 위진시기 문인들에게 상당한 인기를 끌었으며 역대로 더욱 발전양상을 띄어 구식, 결구, 장법, 수사 등 여러 분야가 날로 성숙되었다. 홰나무에 대한 상징을 사용하는 일과 그 문화적 내함은 점차로 확립되어 당시의 심미관을 선명하게 체현하였다. 위진시기 홰나무 소재 변문 중 홰 나무 상징의 사용은 대략 세 가지 종류가 있다. 大槐安國(한 사람이 홰나무 밑에서 낮잠을 자다가 꿈에 대괴안국 왕의 사위가 되어 南柯郡을 20년 동안 다스리면서 부귀영화를 누리다가 꿈을 깨었다는 내용)의 정치적 우의, 槐社安民의 심리적 기탁, 그리고 槐仙之道의 현학적 함의이다. 일종의 상징 부호로서 홰나무는 위진시기에는 더 이상 자연지물에 대한 묘사는 아니었다. 부단한 전습과 변화로 다양한 측면의 인문 내함을 부여받게 되었고 농후한 역사적 연속성을 띄게 된 것이다.

        Establishment of a model to evaluate the nutritional quality of Bombyx mori Linnaeus (Lepidoptera, Bombycidae) pupae lipid based on principal components

        Silkworm pupae are a source of edible lipid with a high content of unsaturated fatty acids (UFAs) (70% of total lipid content) and are becoming a focus of pharmaceutical and dietary research in the functional oil field. To study the nutritional value of different silkworm strains, principal component analysis (PCA) was used to establish a mathematical model and cluster analysis on silkworm pupae lipid (SPL) in 90 strains based on Z-score. Single-factor correlation analysis indicated that the PUFAs content was significantly positively correlated with the α-linolenic acid (ALA) (ρ=0.98) and negatively correlated with the oleic acid (OA) (ρ=−0.73) contents, and comprehensive of principal components mathematical model (PC =0.148ZX1 +0.00 2ZX2 −0.197ZX3 −0.338ZX4 +0.220ZX5 +0.304ZX6 +0.314ZX7, ZXi is the standard score (Z-score) of X1, which was calculated as follow: ZXi = (Xi −−X )/S, where X1: palmitic acid, X2: palmitoleic acid, X3: stearic acid, X4: OA, X5: linoleic acid, X6: ALA, X7: PUFAs.−X is the arithmetic mean content of variable i, and S is the standard deviation of variable i for all samples included in the analysis.) showed the amount of ALA and PUFAs were the key factors determining the nutritional value of the pupae lipid. Cluster analysis on the composite scores calculated by the model of the principal component divided the 90 strains into three groups. A cluster of 6 strains (XHR, XIFF, ZX, YT, YO and BTN) with the best nutritional value was characterized by high contents of ALA and PUFAs and a low content of oleic acid.

        북한 歌劇 節歌의 문학형식 특징 연구

        본고는 북한 歌劇 節歌의 시문학형식 특징에 대한 연구이다. 서사, 주제, 극적 구조, 음악, 운율, 정서 등 북한 가극의 여러 가지 특징에 대한 연구의 한 부분으로서의 본고는 분절, 정형형식, 후렴 형식 세 가지 부분으로 절가의 문학형식 특징을 살펴보았다. 분절형식의 특징으로 볼 때, 사용 빈도의 순위가 2절 형식(123수), 1절 형식(45수), 3절 형식(26수), 4절 형식(5수), 6절 형식(1수)의 순서로서, 1절 형식이 3절 형식에 비해 월등이 많으며, 정형형식에 있어서 일반가요에서도 보편적으로 많이 쓰이는 4행 정형형식과 함께 2, 3, 5, 6, 7, 8, 9 등 형식이 다양하게 사용되었다. 그리고 후렴 형식도 가극 절가 시문학 형식의 중요한 특징의 하나이다. 이런 특징들은 모두 인물의 형상화, 환경 묘사, 극적 주제의 서술, 극정의 전개와 발전 등 가극의 극적 수요에 의한 것이며, 민요나 시조로부터의 계승과 발전이라는 민족문화유산의 계승과 발전 차원의 예술사적, 문학적, 정신사적 의의와 가치를 가진다. 최근의 한반도 평화통일 열망 속에, 서로의 이질성 극복과 동질성 회복이 무엇보다도 시급한 지금, 가극이라는 예술분야를 통한 본고의 연구가 조금이나마 도움이 되기를 기대한다. This paper mainly focuses on the research on the literary form characteristics of North Korea Opera section songs with segment, regular form and chorus forms as the main content. It is a part of the study of narrative, theme, structure, music, rhythm, aesthetic emotion and other characteristics of North Korean operas. The research results have three characteristics. One is the sectional characteristics, which ranks the first in the highest usage rate of the two- segment form, then, followed one- segment form, three- segment form, four- segment form and six-segment form in order. The two-segment is a regular form, in addition to quatrain; it has two lines, terza rima, limerick, sestet, heptastich, octave, and nine-line stanza characteristics. The three- segment is also one of the important characteristics of literary form characteristics of North Korea Opera section songs. The above characteristics are formed according to the needs of opera art, such as visualization of characters, the description of environment, the narration of themes, and development of the plot. It has the significance and value of inheriting and carrying forward the national cultural heritage, art history, literature history and spiritual history. It is expected that this paper would be beneficial to mutual recognition and overcoming.

        Alu Tandem Sequences Inhibit GFP Gene Expression by Triggering Chromatin Wrapping

        Alu elements belonging to the short interspersed nuclear elements (SINE) of repetitive elements are present in more than one million copies which altogether represent 10% of the whole human genome. In this study, the roles of Alu tandem sequences in the process of GFP gene (GFP) expression and packing into chromatin of its DNA were studied. To detect the effect of Alu repeats on gene expression, different copies of Alus were inserted GFP downstream respectively in pEGFP-C1 vector. We found that Alu sequences decreased the amount of GFP transcription, the percentage of GFP positive cells and the accessibility to DNase I in length-dependent manner. Inserting Alu caused the production of higher-molecular-mass RNA, indicating Alu sequence did not induce premature transcriptional termination. Tight packing chromatins keep silent and resist to DNase I digestion, which is a general phenomenon. We suggested that head and tail tandem Alu sequences suppressed GFP expression in length dependent manner by triggering chromatin packing.

      • rs10505474 and rs7837328 at 8q24 Cumulatively Confer Risk of Prostate Cancer in Northern Han Chinese

        Aims: Genome-wide association studies (GWAS) have identified several risk variants for prostate cancer (pCa) mainly in Europeans, which need to be further verified in other racial groups. We selected six previously identified variants as candidates and to define the association with PCa in Northern Han Chinese. Methods: 749 subjects from Beijing and Tianjin in Northern China were included. Six variants (rs10505474, rs7837328, rs4242384, rs7813, rs486907 and rs1058205) were genotyped by high resolution melting (HRM) assays. The individual and cumulative contribution for of the risk of PCa and clinical covariates were analyzed. Results: Among the six candidate variants, onlyrs10505474, and rs7837328, both locating at 8q24 region, were associated with PCa in our population.rs10505474 (A) was associated with PCa ($OR_{recessive}=1.56$, p=0.006); and rs7837328 (A) was associated with PCa ($OR_{dominant}=1.38$, p=0.042/$OR_{recessive}=1.99$, p=0.003). Moreover, we observed a cumulative effects between them ($p_{trend}=2.58{\times}10^{-5}$). The joint population attributable risk showed the two variants might account for 71.85% of PCa risk. In addition, we found the homozygotes of rs10505474 (A) and rs7837328 (A) were associated with PCa clinical covariants (age at onset, tumor stage, respectively) ($p_{age}=0.046$, $P_{tumorstage}=0.048$). Conclusion: rs10505474 (A) and rs7387328 (A) at 8q24 are associated with PCa and cumulatively confer risk, suggesting the two variations could determine susceptibility to PCa in the Northern Chinese Han population.

        Protective Effect of Dandelion Extracts on Ethanol-Induced Acute Hepatotoxicity in C57BL/6 Mice

        Dandelion (Taraxacum officinale) has been widely used as an anti-inflammatory agent in oriental medicine. In the current study, we investigated the protective effect, and the possible mechanism, of dandelion extracts against ethanol-induced acute hepatotoxicity in C57BL/6 mice. Dandelion water and ethanol extract was administered at 2 g/㎏ body weight (BW) once daily for 7 consecutive days, whereas control and ethanol groups received water by gavage. Ethanol (50% ethanol; 6 g/㎏ BW) was administered 12 hr before sacrificing the mice in order to generate liver injury. Significantly increased serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities as well as liver triglyceride (TG) and cholesterol levels were attenuated by dandelion supplementation. In addition, dandelion extracts not only enhanced alcohol dehydrogenase (ADH) and anti-oxidative enzyme activities, but reduced lipid peroxidation. Cytochrome P450 2E1 (CYP 2E1), one of the critical enzymes xenobiotic metabolism, expression was lower with ethanol treatment but restored by dandelion supplementation. These results were confirmed by improved histopathological changes in fatty liver and hepatic lesions induced by ethanol. In conclusion, dandelion could protect liver against ethanol administration by attenuating of oxidative stress and inflammatory responses.

        Treatment performance and N2O emission in the UASB-A/O shortcut biological nitrogen removal system for landfill leachate at different salinity

        Landfill leachate was treated using the upflow anaerobic sludge blanket (UASB) combined with anoxic/aerobic reactor (A/O) for shortcut biological nitrogen removal. The treatment performance and nitrousoxide (N2O) emission were studied under different salinity. When salinity increased from 10 to 35 g/L,the removal efficiency of ammonium nitrogen and total nitrogen decreased from 99.3 to 83.9% and from85.4 to 68.4%, respectively while the nitrite accumulation rate always remained high at more than 94.3%. The N2O conversion ratio increased by 14.7 times, from 0.3 to 4.7%. The concentration of biodegradableCOD in influent of the A/O reactor increased with the increase of salinity, which lead to the decrease ofdissolved oxygen and thus stimulated N2O emission remarkably during the aerobic zone. Meanwhile, theN2O emissions were likely to occur by a combination of pathways since the simultaneous nitritation anddenitritation was occurred in aerobic zone. The increase of NH4+-N concentration, NO2-N concentrationand pH caused by high salinity also promoted the N2O emission. The population of Nitrosomonaseuropaea increased with salinity, which was important for N2O emission.
