Laminin and Chromogranin Expression on the Korean Stomach and Colorectal Carcinoma Cell Lines

Laminin is a major basement membrane component synthesized by epithelial and endothelial cells. It is involved in embryonic differentiation, morphogenesis, cell migration, neurite growth and tumor metastasis. To investigate the role of laminin in tumor metastasis, we have used 10 cell lines derived from Korean stomach and colorectal carcinoma patients. The expression of laminin gene was analyzed by Northern and slot blot techniques. Elevated level of laminin mRNA was found in stomach cancer cell line, SNU-16 and colorectal cancer cell line, SNU-67. The high level of laminin expression found in these cell lines may be associated with the metastatic state of these tumors. In addition, we have analyzed the expression of chromogranin A gene on these cell lines using the same experimental techniques. Chromogranin A is a protein present in the secretory granules of endocrine and neuronal cells and can be used as marker for neuroendocrine lineage. The fact that a previously unrecognized subgroup of stomach and colorectal carcinomas has neuroendocrine features will improve tumor classification, diagnosis and treatment. Of the tested 10 tumor cell lines, SNU-16 (stomach), SNU-67 (colon), and SNU-C2A (colon) express chromogranin A gene. Thus these cell lines might be of neuroendocrine origin. Further, it seems reasonable that some subsets of colon and stomach adenocarcinomas might have neuroendocrine features.
Carotid Ultrasonography of Thyroid Incidentaloma

With the widespread use of a high-resolution sonographic equipment for breast ultrasound or carotid ultrasonography, the proportion of asymptomatic cases in which thyroid nodules are incidentally found has been increased. Given the above background, we attempted to assess the incidence of incidental thyroid cancers, which were incidentally detected while examining the thyroid gland, one of the anatomical structures that are adjacently located, during the carotid ultrasonography, and the rate of malignancy on ultrasonography. The current study was conducted in 258 patients who underwent a carotid ultrasonography. Of these, excluding patients who were aware that thyroid nodules were present in these nodules, 273 patients were finally selected for the incidental thyroid tumor. Then, based on the interpretation results, the incidence of nodules, size and the findings were thyroid nodules were classified into probably benign, indeterminate and suspiciously malignant nodule. Based on this classification, the rate of malignancy was evaluated. In our series, 49 of 237 patients (20.7%) had thyroid nodules detected. Mean size was 1.13 cm (range 0.3 cm~5.1 cm). The number of cases in which the size of thyroid nodules was smaller than 1 cm (60.4%). Of thyroid nodules, probably benign nodule of < 5 mm in size was not recorded on the ultrasonography. Besides, probably benign nodules were identified from 51.7% (30/58), indeterminate nodules were identified from 39.7% (23/58) and suspiciously malignant nodules were identified from 8.6% (5/58). These results indicate that the malignancy rate was relatively higher. It can therefore be inferred that even the incidental thyroid rumors should also be managed and then treated in the same manner a thyroid nodules which are found in a clinical setting.

Botulinum neurotoxin A (BoNT/A) has been used therapeutically to treat muscular hypercontractions and sudomotor hyperactivity and it has been reported that BoNT/A might have analgesic properties in headache. PEP-1 peptide is a known carrier peptide that delivers fulll-ength native proteins in vitro and in vivo. In this study, a BoNT/A gene were fused with PEP-1 peptide in a bacterial expression vector to produce a genetic in-frame PEP-1-BoNT/A fusion protein. The expressed and purified PEP-1-BoNT/A fusion proteins were efficiently transduced into cells in a time- and dose-dependent manner when added exogenously in a culture medium. In addition, immuno-histochemical analysis revealed that PEP-1-BoNT/A fusion protein efficiently penetrated into the epidermis as well as the dermis of the subcutaneous layer, when sprayed on mice skin. These results suggest that PEP-1-BoNT/A fusion protein provide an efficient strategy for therapeutic delivery in various human diseases related to this protein.
Random Permutation Test for Comparison of Two Survival Curves

There are many situations in which the well-known tests such as log-rank test and Gehan-Wilcoxon test fail to detect the survival differences. Assuming large samples, these tests are developed asymptotically normal properties. Thus, they shall be called asymptotic tests in this paper, Several asymptotic tests sensitive to some specific types of survival differences have been recently proposed. This paper compares by simulations the test levels and the powers of the conventional asymptotic tests and their random permutation versions. Simulation studies show that the random permutation tests possess competitive powers compared to the corresponding asymptotic tests, keeping exact test levels even in the small sample case. It also provides the guidelines for choosing the valid and most powerful test under the given situation.

<P>The chemically synthesized suppressor and leveler are added together with bis(3-sulfopropyl)disulfide (SPS) to galvanostatically fill up the trenches with the similar dimensions to those of the through silicon vias. In our previous study, the deposition of the coarse-grained Cu was indicated as a drawback of the synthesized additives, i.e., polyoxy polymer with amine terminal groups and pyridine derivatives containing additional amine groups. In this study, the modified chemistry of organic additives is used, enabling the bottom-up filling of trenches and improving the microstructure of the deposited Cu. The conversion of the functional groups from amine to hydroxyl groups in both the suppressor and leveler, and the uniform adsorption of modified suppressor improved the microstructure of the deposited Cu. The void-free trench filling is induced by the selective adsorption and accumulation of SPS at the bottom and negligible deposition on the top and side-walls of the trenches. Based on the filling mechanism, the trenches with 9 μm width and 50 μm depth are galvanostatically filled in ≤20 min.</P>
Highly efficient and bending durable perovskite solar cells: toward a wearable power source

<P>Perovskite solar cells are promising candidates for realizing an efficient, flexible, and lightweight energy supply system for wearable electronic devices. For flexible perovskite solar cells, achieving high power conversion efficiency (PCE) while using a low-temperature technology for the fabrication of a compact charge collection layer is a critical issue. Herein, we report on a flexible perovskite solar cell exhibiting 12.2% PCE as a result of the employment of an annealing-free, 20 nm thick, amorphous, compact TiO<SUB><I>x</I></SUB> layer deposited by atomic layer deposition. The excellent performance of the cell was attributed to fast electron transport, verified by time-resolved photoluminescence and impedance studies. The PCE remained the same down to 0.4 sun illumination, as well as to a 45° tilt to incident light. Mechanical bending of the devices worsened device performance by only 7% when a bending radius of 1 mm was used. The devices maintained 95% of the initial PCE after 1000 bending cycles for a bending radius of 10 mm. Degradation of the device performance by the bending was the result of crack formation from the transparent conducting oxide layer, demonstrating the potential of the low-temperature-processed TiO<SUB><I>x</I></SUB> layer to achieve more efficient and bendable perovskite solar cells, which becomes closer to a practical wearable power source.</P> <P>Graphic Abstract</P><P>We report annealing-free compact TiO<SUB><I>x</I></SUB> layer by atomic layer deposition for high efficiency flexible perovskite solar cells, and maintained 95% of the initial PCE after 1000 bending cycles with 10 mm bending radius. <IMG SRC='http://pubs.rsc.org/services/images/RSCpubs.ePlatform.Service.FreeContent.ImageService.svc/ImageService/image/GA?id=c4ee02441a'> </P>
Complete sequence-based screening of TPMT variants in the Korean population

Thiopurine S-methyltransferase (TPMT) is a cytoplasmic enzyme involved in the metabolism of thiopurine drugs and its activity is largely influenced by polymorphisms of the TPMT gene. To date, more than 35 TPMT variants are known to be associated with reduced enzyme activity, but most studies on the TPMT genotype have included only common nonfunctional variants, such as TPMT*2 and TPMT*3. In this study, we carried out a complete sequencing analysis to screen all TPMT variants in Korean patients. A total of 900 Korean patients were genotyped for TPMT and 30 patients (3.3%) had the known TPMT variant alleles. TPMT*3C was found in 25 patients (2.8%): 24 patients with TPMT*1/*3 and one with TPMT*3/*3. Rare TPMT variants including TPMT*6, TPMT*16, and TPMT*32 were detected in five patients (0.6%) and a novel variant, TPMT*38 (c.514T>C, p.S172P), was identified in two patients. This is the first complete sequence-based screening study evaluating all TPMT variants in Asian populations.
