Comparison of Chromosome Aberration Sister Chromatid Exchange for the Assement of Cadmium Toxicity

Induction of Chromosome Aberration and Sister Chromatid exchange by CdC1₂ in CHO-K, cells and human peripheral lymphocytes was studied in order to compare the applicability of them for the assessment of cytotoxicity of cadmium. The frequencies of chromosome aberrations in CHO cells treated with cadmium during G₁ and human lymphocytes during G_0 were increased with dose-dependent manner, and almost all aberrations were chromatid types suggesting that cadmium was S-dependent agent. On the other hand, no significant increase in frequencies of SCE both in CHO cells and in human lymphocytes was observed. These data implicate that chromosome aberration analysis is good for the assessment of cadmium toxicity but SCE analysis is not good for it.

In this study, the 2-mercaptopyridine (2-MPY) and 4-mercaptopyridine (4-MPY) on to silver mirror and silver sol substrates have been studied by surface enhanced Raman scattering (SERS). The surface enhanced properties of 2-MPY and 4-MPY showed that the substrate of silver mirror was superior to the sliver sol. The silver mirror and silver sol substrates were prepared by Tollen’s and gamma irradiation method respectively. UV-visible spectra indicate that Ag nanoparticles were produced by this method. Considerable differences were found in the SERS spectra of 2-MPY and 4-MPY molecules onto silver mirror and silver sol substrates. From the analysis of the SERS data, it was evidenced that 2-MPY was chemisorbed through its S atom to Ag surfaces at a perpendicular orientation, whereas 4-MPY was bonded to Ag surface via the S atom and N atom assuming a tilted orientation.

<P>Highly conductive indium oxide films, electrically more conductive than commercial sputtered indium tin oxide films films, were deposited using a new liquid precursor Et<SUB>2</SUB>InN(SiMe<SUB>3</SUB>)<SUB>2</SUB> and H<SUB>2</SUB>O by atomic layer deposition (ALD) at 225–250 °C. Film resistivity can be as low as 2.3 × 10<SUP>–4</SUP>–5.16 × 10<SUP>–5</SUP> Ω·cm (when deposited at 225–250 °C). Optical transparency of >80% at wavelengths of 400–700 nm was obtained for all the deposited films. A self-limiting ALD growth mode was found 0.7 Å/cycle at 175–250 °C. X-ray photoelectron spectroscopy depth profile analysis showed pure indium oxide thin film without carbon or any other impurity. Purpose: This study evaluated the predictive performance of a combination of self-report questionnaires, salivary hemoglobin levels, and age as a non-invasive screening method for periodontitis. Methods: The periodontitis status of 202 adults was examined using salivary hemoglobin levels, responses to 10 questions on a self-report questionnaire, and the Community Periodontal Index (CPI). The ability of those two variables and the combination thereof with age to predict the presence of CPI scores of 3-4 and 4 was assessed using logistic regression and receiver operating characteristic (ROC) curve analysis. Results: CPI scores of 3-4 and 4 were present among 79.7% and 46.5% of the sample, respectively. The area under the ROC curves (AUROCs) of salivary hemoglobin levels for predicting prevalence of CPI scores of 3-4 and 4 were 0.63 and 0.67, respectively (with sensitivity values of 71% and 60% and specificity values of 56% and 72%, respectively). Two distinct sets of five questions were associated with CPI scores of 3-4 and 4, with AUROCs of 0.73 and 0.71, sensitivity values of 76% and 66%, and specificity values of 63% and 69%. The combined model incorporating both variables and age showed the best predictive performance, with AUROCs of 0.78 and 0.76, sensitivity values of 71% and 65%, and specificity values of 68% and 77% for CPI scores of 3-4 and 4, respectively. Conclusions: The combination of salivary hemoglobin levels and self-report questionnaires was shown to be a valuable screening method for detecting periodontitis.

<P><B>Background/Aims</B></P><P>The purpose of this study was to investigate the malignant potential of aberrant crypt foci (ACF) by measuring the multiplicity of crypts and lectin expression in the early and late stages of 1,2-dimethylhydrazine (DMH)-induced colon carcinogenesis.</P><P><B>Methods</B></P><P>Six-week-old Wistar rats were injected subcutaneously with DMH for 27 weeks. We classified ACF according to the number of crypts per ACF as a few crypts (≤3 crypts, FC ACF) or numerous crypts (≥4 crypts, NC ACF). Immunohistochemistry was used to evaluate lectin expression.</P><P><B>Results</B></P><P>In the early stage, FC ACF (590/1,902, 31.0%) occurred more frequently than NC ACF (35/449, 7.8%); whereas in the late stage, NC ACF (176/449, 39.2%) occurred more frequently than FC ACF (324/1,902, 17.0%). The number of ACF peaked at 15 to 20 weeks. The ratio of NC/FC ACF increased gradually during carcinogenesis. The expression of both UEA1 and PNA was higher in NC ACF than FC ACF. Lectin expression increased in the late stage compared with the early stage.</P><P><B>Conclusions</B></P><P>The expression of lectin was higher in NC ACF and ACF in the late stage. Therefore, ACF with higher multiplicities in the late stage may have more malignant potential in DMH-induced colon carcinogenesis.</P>

<P><B>Abstract</B></P> <P>This study achieves a better understanding how <I>Scenedesmus quadricauda</I> microalgae participate in the phycoremediation of a reverse osmosis (RO) concentrate at a high salinity (8000mgCl<SUP>−</SUP> L<SUP>−1</SUP>). Algal treatment of the RO concentrate under continuous illumination resulted in a notable increase in the biodegradability of dissolved organic matter, subsequent removal of biodegradable fractions, and simultaneous removal of nutrients (N and P). This is the first time that <I>S. quadricauda</I> is shown to induce the degradation of polymeric organic matter in the RO concentrate, which is typically refractory to microbial decomposition. In this study, the mechanisms for algae-induced degradation were investigated by measuring the hydrogen peroxide (H<SUB>2</SUB>O<SUB>2</SUB>) released out of algal cells and dispersed in the aqueous phase. The algae-induced biodegradation process has an advantage over typical electrochemical oxidation technologies in that photo-sensitive living organisms are capable of self-repair, reproduction, and nutrient uptake. Our results indicate that the use of algae-induced oxidation is highly feasible as a safe, inexpensive technology to pre-treat non- or slowly-biodegradable organic matter in wastewater prior to downstream biological processing.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The robust phycoremediation of RO concentrate was demonstrated at a high salinity. </LI> <LI> <I>S. quadricauda</I> induced the degradation of refractory organics in the RO concentrate. </LI> <LI> Proposed method is promising to reduce harmful byproducts and energy consumption. </LI> <LI> Much work is needed to fully exploit the algae-induced oxidation characteristics. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

<B>Abstract</B><P>Homing of endothelial progenitor cells (EPCs) to the neovascular zone is now considered to be an essential step in the formation of vascular networks during embryonic development and also for neovascularization in postnatal life. We report here the prominent role of the insulin-like growth factor 2 (IGF2)/IGF2 receptor (IGF2R) system in promoting EPC homing. With high-level expression of IGF2R in EPCs, IGF2-induced hypoxic conditions stimulated multiple steps of EPC homing in vitro and promoted both EPC recruitment and incorporation into the neovascular area, resulting in enhanced angiogenesis in vivo. Remarkably, all IGF2 actions were exerted predominantly through IGF2R-linked G(i) protein signaling and required intracellular Ca2+ mobilization induced by the β2 isoform of phospholipase C. Together, these findings indicate that locally generated IGF2 at either ischemic or tumor sites may contribute to postnatal vasculogenesis by augmenting the recruitment of EPCs. The utilization of the IGF2/IGF2R system may therefore be useful for the development of novel means to treat angiogenesis-dependent diseases.</P>
