Identifying a Cinnamoyl Coenzyme A Reductase (CCR) Activity with 4-Coumaric Acid: Coenzyme A Ligase (4CL) Reaction Products in Populus tomentosa

        Dong-Dong Wang,Hua Bai,Wei-Qi Chen,Hai Lu,Xiang-Ning Jiang 한국식물학회 2009 Journal of Plant Biology Vol.52 No.5

A cinnamoyl coenzyme A reductase (CCR, EC 1.2.1.44), one of the key enzyme involved in lignin biosynthesis, was cloned from Populus tomentosa (Chinese white poplar). At the same time, a 4CL1 gene was cloned from P. tomentosa, too. The two genes were subcloned in pQE31 vector and expressed in Escherichia coli M15. Both of them were purified by Ni-NTA. Purified CCR protein was digested by trypsin and analyzed by HPLC-MS; the peptide segments had 27% similarity with the sequence of the CCR protein. 4CL was thought to be a neighbor enzyme of CCR in lignin biosynthesis. In this paper, a 4CL1 from P. tomentosa was cloned, and its enzyme reaction products were extracted for the substrates of CCR. Three 4CL1 enzyme reaction products were monitored by HPLC-MS and then the CCR enzyme reaction was detected by GC-MS. In the CCR reaction, the three corresponding aldehyde (p-coumaraldehyde, caffealdehyde, and coniferaldehyde) were detected and identified by Frontier3 software. The results showed that the CCR that we cloned from P. tomentosa had affinities with 4CL1 enzyme reaction products and a ptCCR that was cloned from aspen (Li et al., Plant Cell Physiol 46(7):1073–1082, 2005) only had affinity with feruloyl-CoA. The different results maybe depend on the different study method. The method of exacting 4CL enzyme products as the substrates of CCR in the paper was reliable and can be used in lignin biosynthesis network to detect the enzymes in the neighborhood that depended on the polarity of the substrates and products. This CCR gene had eight homology sequence CCR gene when a BLAST was conducted in Populus trichocarpa genome database. The CCR homology genes in Populus suggested that some CCRs may take part in the lignin biosynthesis, too. The gene family would be the hot spot in the future study.

Radiofrequency Ablation Combined with Transcatheter Arterial Chemoembolization Therapy Versus Surgical Resection for Hepatocellular Carcinoma within the Milan Criteria: A Meta-Analysis

        Wei-dong Wang,Li-hua Zhang,Jia-Yan Ni,Xiong-ying Jiang,Dong Chen,Yao-ting Chen,Hong-liang Sun,Jiang-hong Luo,Lin-feng Xu 대한영상의학회 2018 Korean Journal of Radiology Vol.19 No.4

Objective: To meta-analytically compare combined transarterial chemoembolization (TACE) plus radiofrequency ablation (RFA) and surgical resection (SR) for the treatment of hepatocellular carcinoma (HCC) within the Milan criteria. Materials and Methods: PubMed, Medline, Embase, and Cochrane Library were searched for studies comparing these two therapies that were published between January 2006 and August 2017. Overall survival rate (OS), recurrence-free survival rate (RFS), major complications and the average length of hospital stay were compared between these two therapies. Metaanalytic pooled odds ratio (OR) was calculated using TACE plus RFA as the base category. Results: Seven case-control studies and one randomized trial were identified. Meta-analytic results revealed that, compared with SR, TACE plus RFA had significantly higher 1-year OS (OR for survival = 0.50, p = 0.009) and lower major complications (OR = 1.88, p = 0.02) after therapy. Three studies reported on the length of hospital stay. The average length ± standard deviation reported in individual studies for SR and TACE plus RFA groups was 19.8 ± 8.4 days and 7.4 ± 2.2 days, respectively; 18.7 ± 4.9 days and 11.5 ± 6.9 days, respectively; and 16.6 ± 6.7 days and 8.5 ± 4.1 days, respectively (p < 0.0001 for all studies). Three or 5-year OS and 1-, 3-, or 5-year RFS did not significantly differ between the two therapies. Conclusion: Combined TACE plus RFA may be an alternative to SR for the treatment of patients with HCC within Milan the criteria. Non-randomized design in most of the original studies was a limitation.

      • Frequency Tracking of Power Grid based on Phase Difference and UKF

Wei Dong,Wei Dong,Shichao Jiang 보안공학연구지원센터 2015 International Journal of Security and Its Applicat Vol.9 No.12

For frequency tracking problem of non-stationary signals in power grid, we propose a unscented Kalman filter (UKF) and the weighted phase difference smoothing algorithm. It improved poor initial conditions sensitive situations of robustness UKF. In order to improve the accuracy of the algorithm, real-time and noise immunity, a suboptimal multiple fading factor and weighted smoothing method is added to improve UKF for frequency tracking of power signals. At last, The simulation results are given. It shows that this method has better accurate, real-time tracking of various frequency power signal and its performance is superior to similar documents tracking algorithm.

Biotransformation of Panax ginseng extract by rat intestinal microflora: identification and quantification of metabolites using liquid chromatography-tandem mass spectrometry

        Dong, Wei-Wei,Zhao, Jinhua,Zhong, Fei-Liang,Zhu, Wen-Jing,Jiang, Jun,Wu, Songquan,Yang, Deok-Chun,Li, Donghao,Quan, Lin-Hu The Korean Society of Ginseng 2017 Journal of Ginseng Research Vol.41 No.4

Background: In general, after Panax ginseng is administered orally, intestinal microbes play a crucial role in its degradation and metabolization process. Studies on the metabolism of P. ginseng by microflora are important for obtaining a better understanding of their biological effects. Methods: In vitro biotransformation of P. ginseng extract by rat intestinal microflora was investigated at $37^{\circ}C$ for 24 h, and the simultaneous determination of the metabolites and metabolic profile of P. ginseng saponins by rat intestinal microflora was achieved using LC-MS/MS. Results: A total of seven ginsenosides were detected in the P. ginseng extract, including ginsenosides Rg1, Re, Rf, Rb1, Rc, Rb2, and Rd. In the transformed P. ginseng samples, considerable amounts of deglycosylated metabolite compound K and Rh1 were detected. In addition, minimal amounts of deglycosylated metabolites (ginsenosides Rg2, F1, F2, Rg3, and protopanaxatriol-type ginsenosides) and untransformed ginsenosides Re, Rg1, and Rd were detected at 24 h. The results indicated that the primary metabolites are compound K and Rh1, and the protopanaxadiol-type ginsenosides were more easily metabolized than protopanaxatriol-type ginsenosides. Conclusion: This is the first report of the identification and quantification of the metabolism and metabolic profile of P. ginseng extract in rat intestinal microflora using LC-MS/MS. The current study provided new insights for studying the metabolism and active metabolites of P. ginseng.

C-type natriuretic peptide attenuates renal osteodystrophy through inhibition of FGF-23/MAPK signaling

        Dong Dong Zhang,Yang Fang Wu,Wei Xia Chen,Yao Xu,Si Yan Liu,Huang Huang Luo,Guang Mei Jiang,Yue Wu,Peng Hu 생화학분자생물학회 2019 Experimental and molecular medicine Vol.51 No.-

Renal osteodystrophy (ROD) occurs as early as chronic kidney disease (CKD) stage 2 and seems ubiquitous in almost all pediatric patients with CKD stage 5. Fibroblast growth factor (FGF)-23, a bone-derived endocrine regulator of phosphate homeostasis, is overexpressed in CKD and disturbs osteoblast differentiation and matrix mineralization. In contrast, C-type natriuretic peptide (CNP) acts as a potent positive regulator of bone growth. In the present study, we infused CNP into uremic rats and observed whether CNP could attenuate ROD through the inhibition of FGF-23 cascades. In uremic rats, CNP administration significantly alleviated renal dysfunction, calcium phosphate metabolic disorders, hypovitaminosis D, secondary hyperparathyroidism, the decrease in bone turnover markers and retarded bone pathological progression. More importantly, within FGF-23/mitogen-activated protein kinase (MAPK) signaling, the fibroblast growth factor receptor-1, Klotho and alternative (STAT-1/phospho-STAT-1) elements were upregulated by CNP, whereas FGF-23, RAF-1/phospho-RAF-1, and downstream (ERK/phospho-ERK and P38/phospho-P38) elements were paradoxically underexpressed in bone tissue. Therefore, CNP exerts a therapeutic effect on ROD through inhibition of FGF-23/MAPK signaling at the RAF-1 level.

Inhibition of microRNA-214 promotes epithelial mesenchymal transition process and induces interstitial cystitis in postmenopausal women by upregulating Mfn2

        Jian-Wei Lv,Wei Wen,Chen Jiang,Qi-Bo Fu,Yin-Jun Gu,Ting-Ting Lv,Zhen-Dong Li,Wei Xue 생화학분자생물학회 2017 Experimental and molecular medicine Vol.49 No.-

Our study aims to investigate the roles that microRNA-214 (miR-214) plays in the epithelial mesenchymal transition (EMT) process and the development of interstitial cystitis (IC) in postmenopausal women by targeting Mitofusin 2 (Mfn2). IC bladder tissues and adjacent normal bladder tissues were collected from postmenopausal women. Immunohistochemistry (IHC) staining was conducted. The target relationship between miR-214 and Mfn2 was determined by a dual luciferase reporter gene assay. Adipose-derived mesenchymal stem cells (ADMSCs) were extracted from postmenopausal rats and assigned to the blank, mimics, miR-214 inhibitors, mimics negative control (NC), inhibitors NC, Mfn2 siRNA, miR-214 inhibitors and Mfn2 siRNA groups. Exosomes secreted by transfected ADMSCs were instilled into the bladders of postmenopausal rats. The expression of miR-214 and Mfn2 mRNA and EMT markers was assessed by qRT-PCR and western blotting. It was confirmed that Mfn2 was the target gene of miR-214 in IC. Compared with the normal bladder tissues, miR-214 decreased, but Mfn2 increased in IC bladder tissues. Compared with the blank group, the expression of miR-214 and the expression levels of N-cadherin, Fibronectin, Twist1, Snail and Vimentin mRNA and protein increased, whereas the expression levels of Mfn2, E-cadherin and ZO-1 mRNA and protein decreased in the miR-214 mimics and Mfn2 groups. The expression of MiR-214 and the expression levels of N-cadherin, Fibronectin, Twist1, Snail and Vimentin mRNA and protein decreased, whereas the expression levels of Mfn2, E-cadherin and ZO-1 mRNA and protein increased in the miR-214 inhibitors group. Our findings indicate that the inhibition of miR-214 promotes the EMT process and contributes to bladder wall fibrosis by up-regulating Mfn2, thus leading to the occurrence of IC in postmenopausal women.

A path-dependent fatigue crack propagation model under non-proportional modes I and III loading conditions

        Mei, J.,Dong, P.,Kalnaus, S.,Jiang, Y.,Wei, Z. Pergamon Press 2017 Engineering fracture mechanics Vol.182 No.-

<P><B>Abstract</B></P> <P>It has been well established that fatigue damage process is load-path dependent under non-proportional multi-axial loading conditions. Most of studies to date have been focusing on interpretation of S-N based test data by constructing a path-dependent fatigue damage model. This paper presents a two-parameter mixed-mode fatigue crack growth model which takes into account of crack growth dependency on both load path traversed and a maximum effective stress intensity attained in a stress intensity factor plane ( e . g . , K I - K III plane). By taking advantage of a path-dependent maximum range (PDMR) cycle definition (Dong et al., 2010; Wei and Dong, 2010), the two parameters are formulated by introducing a moment of load path (MLP) based equivalent stress intensity factor range ( Δ K NP ) and a maximum effective stress intensity parameter K Max incorporating an interaction term K I · K III . To examine the effectiveness of the proposed model, two sets of crack growth rate test data are considered. The first set is obtained as a part of this study using 304 stainless steel disk specimens subjected to three combined non-proportional modes I and III loading conditions (i.e., with a phase angle of 0°, 90°, and 180°). The second set was obtained by Feng et al. (2007) using 1070 steel disk specimens subjected to similar types of non-proportional mixed-mode conditions. Once the proposed two-parameter non-proportional mixed-mode crack growth model is used, it is shown that a good correlation can be achieved for both sets of the crack growth rate test data.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Presented a two-parameter mixed-mode fatigue crack growth model. </LI> <LI> Both load path dependency and mean stress effect are considered. </LI> <LI> Validated by non-proportional mixed mode I and III crack growth tests on 304 steel. </LI> <LI> Good correlations are achieved both for test data obtained in this study and from others. </LI> </UL> </P>

Thermodynamic simulation and structural optimization of the collimator in the drift duct of EAST-NBI

        Tang Ning,Hu Chun-dong,Xie Yuan-lai,Wei Jiang-long,Cui Zhi-Wei,Xie Jun-Wei,Pan Zhuo,Jiang Yao 한국원자력학회 2022 Nuclear Engineering and Technology Vol.54 No.11

The collimator is one of the high-heat-flux components used to avoid a series of vacuum and thermal problems. In this paper, the heat load distribution throughout the collimator is first calculated through experimental data, and a transient thermodynamic simulation analysis of the original model is carried out. The error of the pipe outlet temperature between the simulated and experimental values is 1.632%, indicating that the simulation result is reliable. Second, the model is optimized to improve the heat transfer performance of the collimator, including the contact mode between the pipe and the flange, the pipe material and the addition of a twisted tape in the pipe. It is concluded that the convective heat transfer coefficient of the optimized model is increased by 15.381% and the maximum wall temperature is reduced by 16.415%; thus, the heat transfer capacity of the optimized model is effectively improved. Third, to adapt the long-pulse steady-state operation of the experimental advanced superconducting Tokamak (EAST) in the future, steady-state simulations of the original and optimized collimators are carried out. The results show that the maximum temperature of the optimized model is reduced by 37.864% compared with that of the original model. The optimized model was changed as little as possible to obtain a better heat exchange structure on the premise of ensuring the consumption of the same mass flow rate of water so that the collimator can adapt to operational environments with higher heat fluxes and long pulses in the future. These research methods also provide a reference for the future design of components under high-energy and long-pulse operational conditions
