DNA methylation and mRNA expression of COL6A3 in antler mesenchyme of female and male reindeer

        JianCheng Zhai,Ruo‑Bing Han,Sheng‑Nan Wang,Qiang‑Hui Wang,Yan‑Ling Xia,Wei‑Shi Liu,Ya‑Jie Yin,He‑Ping Li 한국유전학회 2019 Genes & Genomics Vol.41 No.9

Backgroud Reindeer is the only deer species that both male and female produce antlers, which provides a particularly interesting case in studying the differences between antlers of the two sexes. Alpha 3(VI) Collagen Gene (COL6A3), forms a microfibrillar network associated with the structural integrity and biomechanical properties, has been found to be one of the differentially expressed genes in antler mesenchyme of female and male reindeer. Objective and Methods The promoter sequence of reindeer COL6A3 gene was obtained using the cloning technology and analyzed by the bioinformatics methods. Bisulfite sequencing PCR (BSP) was used to detect the methylation status of the COL6A3 promoter in reindeer antler mesenchyme. Real-time quantitative PCR was used to detect COL6A3 expression in the antler mesenchyme of female and male reindeer. Results Sequence analysis revealed that the reindeer COL6A3 partial promoter sequence was 983 bp including the possible promoter region at + 105 bp to + 155 bp. Homology and phylogenetic analysis indicated that the COL6A3 promoter of reindeer had the closest genetic distance with Bos taurus, Capra hircus and Ovis aries. BSP results indicated that the methylation level of COL6A3 promoter in the female reindeer antler mesenchyme was significantly higher than in the male. Correlating with increased methylation status, we also found that COL6A3 mRNA expression in female reindeer antler mesenchyme was significantly lower than in the male. Conclusion The higher methylation level of the COL6A3 gene in female reindeer antler mesenchyme coincides with decreased COL6A3 mRNA expression, thereby affecting the transposon silencing mechanism and possibly contributing to apparent differences of antlers in female and male reindeer.

      • The genome of the mesopolyploid crop species Brassica rapa

        Wang, Xiaowu,Wang, Hanzhong,Wang, Jun,Sun, Rifei,Wu, Jian,Liu, Shengyi,Bai, Yinqi,Mun, Jeong-Hwan,Bancroft, Ian,Cheng, Feng,Huang, Sanwen,Li, Xixiang,Hua, Wei,Wang, Junyi,Wang, Xiyin,Freeling, Michael Nature Publishing Group, a division of Macmillan P 2011 Nature genetics Vol.43 No.10

We report the annotation and analysis of the draft genome sequence of Brassica rapa accession Chiifu-401-42, a Chinese cabbage. We modeled 41,174 protein coding genes in the B. rapa genome, which has undergone genome triplication. We used Arabidopsis thaliana as an outgroup for investigating the consequences of genome triplication, such as structural and functional evolution. The extent of gene loss (fractionation) among triplicated genome segments varies, with one of the three copies consistently retaining a disproportionately large fraction of the genes expected to have been present in its ancestor. Variation in the number of members of gene families present in the genome may contribute to the remarkable morphological plasticity of Brassica species. The B. rapa genome sequence provides an important resource for studying the evolution of polyploid genomes and underpins the genetic improvement of Brassica oil and vegetable crops.

Field Emission from TiO2/Ti Nanotube Array Films Modified with Carbon Nanotubes

        Cheng-Wei Wang,Rui-Sheng Guo,Jian-Biao Chen,Yan Li,Jian Wang,Wei-Min Liu 한국물리학회 2009 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.55 No.6

Well-aligned TiO2/Ti nanotube array films were synthesized by using anodic oxidation of titanium in 0.5-wt% HF and were modified with carbon nanotubes (CNTs) assembled by using catalyzed chemical vapor deposition (CVD) at 650 ˚C. The morphology and the quality of the films were assessed with field emission scanning electron microscopy and Raman spectrometry (325 nm). The field emission characteristics of such TiO2/Ti nanostructures were investigated before and after being modified with CNTs. The results show that the TiO2/Ti nanotube arrays possess a moderate turn-on electric field of 7.4 V/µm, a field emission current density of 2.7 mA/cm2 at 23 V/µm. It is noteworthy that the turn-on field of the TiO2/Ti nanotube arrays modified with CNTs is decreased significantly to 1.3 V/µm, and the emission current density is increased to 10 mA/cm2 at 5.6 V/µm. This novel structure shows high emission efficiency as a field emitter. • Which is the Best Chinese Herb Injection Based on the FOLFOX Regimen for Gastric Cancer? A Network Meta-analysis of Randomized Controlled Trials

Wang, Jian-Cheng,Tian, Jin-Hui,Ge, Long,Gan, Yu-Hong,Yang, Ke-Hu Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.12

Background: Few studies have directly compared clinical efficacy and safety among Chinese herb injections (CHIs) for gastric cancer (GC). The present study aimed to compare CHIs combined with FOLFOX regimens for GC to show which provides the best CHIs results. Materials and Methods: 9 electronic databases and 6 gray literature databases were comprehensive searched in April 20, 2013. According to inclusion and exclusion criteria, two reviewers independently selected and assessed the included trials. The risk of bias tool described in the Cochrane Handbook version 5.1.0 and CONSORT statement were used to assess the quality of the trials. All calculations and graphs were performed and produced using ADDIS 1.16.5 software. Results: A total of 541 records were searched and 38 RCTs met the inclusion criteria (2,761 participants), involving 10 CHIs. The results of network meta-analysis showed that compared with FOLFOX alone, combinations with Kanglaite, Astragalus polysaccharides, Cinobufacini, or Yadanziyouru injections could furthest strengthen ORR, improve the quality of life, reduce nausea and vomiting, and reduce the incidence of leukopenia (III-IV). Conclusions: Kanglaite injection, Astragalus polysaccharides injection, Yadanziyouru injection were superior to other CHIs in clinical efficacy and safety for GC. The conclusions now need to be confirmed by large sample size direct head-to-head studies.

Alternating Copolymer of Sulfonated Poly(ether ether ketone-benzimidazole)s (SPEEK-BI) Bearing Acid and Base Moieties

        Wang, Jianli,Song, Yilin,Zhang, Cheng,Ye, Zhibin,Liu, Hui,Lee, Myong-Hoon,Wang, Dehai,Ji, Jianbing WILEY-VCH Verlag 2008 Macromolecular Chemistry and Physics Vol.209 No.14

<P>A novel alternating copolymer of sulfonated poly(ether ether ketone-benzimidazole)s (SPEEK-BI) was synthesized by polycondensation of aromatic bisphenol containing benzimidazole and a mixture of sulfonated and non-sulfonated difluorobenzophenones. By varying the amount of two difluorobenzophenones, the ratio of sulfonic acid unit to imidazole base was controlled straightforwardly. The polymer structure was characterized by 1H NMR and IR spectroscopy. Two different methods of ion exchange capacity measurement supported a strong inter- and intra-molecular interaction between acid and base units, which resulted in an improved mechanical strength as well as avoiding phase separation. Tough membranes with good mechanical stability in hot water (80 °C) and alcohol were obtained by solution casting, which is potential for applications in polymer electrolyte membrane fuel cells and membranes for separation of liquids or gases.</P><P> <img src='wiley_img/10221352-2008-209-14-MACP200700610-gra001.gif' alt='wiley_img/10221352-2008-209-14-MACP200700610-gra001'> </P>

Structure-based functional site recognition for p21-activated kinase 4

        Jian Wang,Gang Wang,Yu Sha,Dong-Mei Zhao,Feng Li,Mao-Sheng Cheng 대한약학회 2013 Archives of Pharmacal Research Vol.36 No.12

Recently, many molecular modeling methodsare being developed to better understand the principlesunderlying protein folding. In the present study, fullyflexible dinucleotides d(pApA), d(pApC), d(pApG),d(pApT), d(pCpA), d(pCpC), d(pCpG), d(pCpT), d(pGpA),d(pGpC), d(pGpG), d(pGpT), d(pTpA), d(pTpC), d(pTpG)and d(pTpT) were docked onto the surface of p21-activatedkinase 4 (PAK4) kinase domain. The results showedthat automated docking was a useful tool to identify thefunctional sites of PAK4 and it may provide a theoreticalbasis for the interaction data obtained from previousexperiments. Therefore, structure-based docking with fullyflexible dinucleotide probes might be a good tool to predictand annotate the functional sites of enzymes.

      • Analytical behavior of built-up square concrete-filled steel tubular columns under combined preload and axial compression

        Jian-Tao Wang,Fa-Cheng Wang 국제구조공학회 2021 Steel and Composite Structures, An International J Vol.38 No.6

This paper numerically investigated the behavior of built-up square concrete-filled steel tubular (CFST) columns under combined preload and axial compression. The finite element (FE) models of target columns were verified in terms of failure mode, axial load-deformation curve and ultimate strength. A full-range analysis on the axial load-deformation response as well as the interaction behavior was conducted to reveal the composite mechanism. The parametric study was performed to investigate the influences of material strengths and geometric sizes. Subsequently, influence of construction preload on the full-range behavior and confinement effect was investigated. Numerical results indicate that the axial load-deformation curve can be divided into four working stages where the contact pressure of curling rib arc gradually disappears as the steel tube buckles; increasing width-to-thickness (B/t) ratio can enhance the strength enhancement index (e.g., an increment of 1.88% from B/t=40 to B/t=100), though ultimate strength and ductility are decreased; stiffener length and lip inclination angle display a slight influence on strength enhancement index and ductility; construction preload can degrade the plastic deformation capacity and postpone the origin appearance of contact pressure, thus making a decrease of 14.81%~27.23% in ductility. Finally, a revised equation for determining strain εscy corresponding to ultimate strength was proposed to evaluate the plastic deformation capacity of built-up square CFST columns.

        중국의 拘束制度에 대한 개혁

        汪建成(Wang Jian-Cheng),金昌遵(번역자) 동아대학교 법학연구소 2009 東亞法學 Vol.- No.43

The arrest in China consists of the act of arrest and the subsequent detention. The requirements for arrest are that there are evidences showing the facts of offense, that the offense is punishable by imprisonment and more severe punishments, and that it is necessary to have the offender arrested. The procuratorate has the power of approving arrests; it makes the decision to approve or disapprove after examining the facts. The problems concerning arrest in China are that the requirements for arrest are not quite operative, that the rate of approval and the rate of custody are always very high, and that the rate of guaranteed pending trial is low. China may experiment to innovate conditioned arrest. The academic circle raises diverse voices on its connotation, legality and the range of “major cases”. The conditioned arrest itself, however, is an innovation on the method of examination and approving, which does not lower the standards of arrest requirements. It is favorable for the establishment of case diversion system and it is possible to accelerate the advent of judicial review in China. During the course of reform, the standardization must be achieved; the extension of application should be avoided, and the relationship between the procuratorate and public security organ should be properly dealt with.

      • Growth, Clonability, and Radiation Resistance of Esophageal Carcinoma-derived Stem-like Cells

        Li, Jian-Cheng,Liu, Di,Yang, Yan,Wang, Xiao-Ying,Pan, Ding-Long,Qiu, Zi-Dan,Su, Ying,Pan, Jian-Ji Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.8

Objective: To separate/enrich tumor stem-like cells from the human esophageal carcinoma cell line OE-19 by using serum-free suspension culture and to identify their biological characteristics and radiation resistance. Methods: OE-19 cells were cultivated using adherent and suspension culture methods. The tumor stem-like phenotype of CD44 expression was detected using flow cytometry. We examined growth characteristics, cloning capacity in soft agar, and radiation resistance of 2 groups of cells. Results: Suspended cells in serum-free medium formed spheres that were enriched for CD44 expression. CD44 was expressed in 62.5% of suspended cells, but only in 11.7% of adherent cells. The suspended cells had greater capacity for proliferation and colony formation in soft agar than the adherent cells. When the suspended and adherent cells were irradiated at 5 Gy, 10 Gy, or 15 Gy, the proportion of CD44+ suspended cells strongly and weakly positive for CD44 was 77.8%, 66.5%, 57.5%; and 21.7%, 31.6%, 41.4%, respectively. In contrast, the proportion of CD44+ adherent cells strongly positive for CD44 was 18.9%, 14.%, and 9.95%, respectively. When the irradiation dose was increased to 30 Gy, the survival of the suspended and adherent cells was significantly reduced, and viable CD44+ cells were not detected. Conclusion: Suspended cell spheres generated from OE-19 esophageal carcinoma cells in serum-free stem medium are enriched in tumor stem-like cells. CD44 may be a marker for these cells.

Melatonin Attenuates Mitochondrial Damage in Aristolochic Acid-Induced Acute Kidney Injury

        Sun Jian,Pan Jinjin,Liu Qinlong,Cheng Jizhong,Tang Qing,Ji Yuke,Cheng Ke,wang Rui,Liu Liang,Wang Dingyou,Wu Na,Zheng Xu,Li Junxia,Zhang Xueyan,Zhu Zhilong,Ding Yanchun,Zheng Feng,Li Jia,Zhang Ying,Yua 한국응용약물학회 2023 Biomolecules & Therapeutics(구 응용약물학회지) Vol.31 No.1

Aristolochic acid (AA), extracted from Aristolochiaceae plants, plays an essential role in traditional herbal medicines and is used for different diseases. However, AA has been found to be nephrotoxic and is known to cause aristolochic acid nephropathy (AAN). AA-induced acute kidney injury (AKI) is a syndrome in AAN with a high morbidity that manifests mitochondrial damage as a key part of its pathological progression. Melatonin primarily serves as a mitochondria-targeted antioxidant. However, its mitochondrial protective role in AA-induced AKI is barely reported. In this study, mice were administrated 2.5 mg/kg AA to induce AKI. Melatonin reduced the increase in Upro and Scr and attenuated the necrosis and atrophy of renal proximal tubules in mice exposed to AA. Melatonin suppressed ROS generation, MDA levels and iNOS expression and increased SOD activities in vivo and in vitro. Intriguingly, the in vivo study revealed that melatonin decreased mitochondrial fragmentation in renal proximal tubular cells and increased ATP levels in kidney tissues in response to AA. In vitro, melatonin restored the mitochondrial membrane potential (MMP) in NRK-52E and HK-2 cells and led to an elevation in ATP levels. Confocal immunofluorescence data showed that puncta containing Mito-tracker and GFP-LC3A/B were reduced, thereby impeding the mitophagy of tubular epithelial cells. Furthermore, melatonin decreased LC3A/B-II expression and increased p62 expression. The apoptosis of tubular epithelial cells induced by AA was decreased. Therefore, our findings revealed that melatonin could prevent AA-induced AKI by attenuating mitochondrial damage, which may provide a potential therapeutic method for renal AA toxicity.
