Purpose: The consequences of precipitously rising allergic skin inflammation rates worldwide have accelerated the risk of atopic dermatitis (AD). Natural product-based agents with good efficacy and low risk of side effects offer promising prevention and treatment strategies for inflammation-related diseases. We have already reported that Platycodon grandiflorum root-derived saponins (Changkil saponins, CKS) have many pharmacological effects, including anti-inflammatory and anti-allergic effects, but its influence on AD remains unclear. Therefore, we evaluated the inhibitory effect of CKS, mainly platycodin D, on AD-like skin symptoms in mice and the possible mechanisms in cells. Methods: Mice were sensitized and challenged with 2,4-dinitrochlorobenzene (DNCB). Four weeks after challenge, mice were treated with oral administration of CKS for 4 weeks. In addition, cells were used to evaluate the effect of CKS, mainly platycodin D, on the TARC expression regulated mechanism. Results: CKS attenuated DNCB-induced dermatitis severity, serum levels of IgE and TARC, and mRNA expression of TARC, TNF-α, IFN-γ, IL-4, IL-5, and IL-13 in mice. Histopathological examination showed reduced thickness of the epidermis/dermis and dermal infiltration of inflammatory cells and mast cells in the ears. Moreover, CKS and platycodin D inhibited TNF-α/IFN-γ-induced TARC expression through the suppression of NF-κB and STAT1 and induction of Nrf2/ARE-mediated hemeoxygenase-1 (HO-1) expression in cells. Conclusion: We suggest that CKS and platycodin D inhibited the development of AD-like skin symptoms by regulating cytokine mediators and may be an effective alternative therapy for AD-like skin symptoms.ⓒ2014 Elsevler Gmbh. 키넥트 센서를 이용한 고령자 대상의 선자세 균형능력 평가

Portable low-cost Kinect sensor was used to analyze standing balance ability of the elderly. Eighty subjects who can walk alone and have a normal cognitive level participated in this experiment. Based on Berg Balance scale (BBS) test with 52 points, subjects were divided into Healthy older (HO: 46 persons, BBS: 53.80 ± 1.19) and Impaired older (IO: 34 persons, BBS: 49.06 ± 2.03) group. Each subject performed 30 seconds four different standing balance tests (EO: Eyes Open, EC: Eyes Close, EOf: Eyes Open on foam, ECf: Eyes Close on foam). Five variables (Mean distance, Range of distance, Root mean square, Mean velocity, 95% ellipse area) were calculated from the hip joint center movement of Kinect sensor. Results showed that there were significant differences between groups for four different standing tests. Calculated variables from kinect sensor showed significant correlation with BBS score. Especially, mediolateral mean distance, mediolateral root mean square, mediolateral range of distance and 95% ellipse area showed discriminative ability for all tests. Mean values of variables of IO were higher than those of HO, which means the decreased balance ability in IO compared with HO. Therefore, it was possible to estimate simple balance assessment of the elderly using portable low-cost Kinect sensor.

Like-charge ion pairs of hydronium and hydroxide in aqueous solution?

<P>The like-charge ion pairings of hydronium and hydroxide were investigated using both ab initio cluster calculations and QM/MM-MD aqueous simulations. While only a two-water-bridged H3O+(H2O)(2)H3O+ is found in hydronium cluster calculations, three clusters of HO-(H2O)(2)HO-, HO-(H2O)(3)HO- and HO-(H2O)(4)HO- are stable dihydroxide aggregates. In addition, an interesting yet very stable parallelogram structure of [O-H...H-O](2-) without any bridging water was also discovered using QM/MM-MD simulations. According to our analysis, its unique structure reduces the electrostatic repulsion and allows stable coordination with solvents at the same time. In conclusion, hydroxide can formstronger like-ion pairs than hydronium in aqueous solutionmostly due to its versatile coordination ability with solvents.</P>

■ Objectives The purpose of this case study is to report that a hypertension with intracerebral hemorrhage of tae-eumin was treated with 'Cheongsim Yeunja-tang' and then the symptoms were improved and blood pressure was decreased. ■ Methods Although the patient who had hypertension with intracerebral hemorrhage took antihypertension drugs, intermittent increase of blood pressure was shown. We diagnosed him as Tae-eumin and treated with Cheongsim Yeunja-tang. We daily checked blood pressure and evaluated the symptoms. ■ Results After the treatment with Cheongsim Yeunja-tang, blood pressure was decreased and symptoms were improved. ■ Conclusion The result shows Cheongsim Yeunja-tang has antihypertensive effect and improve hypertension symptoms of Tae-eumin patient with intracerebral hemorrhage.

'1998 한국인 우울증 선별의 날' 참가자들의 사회인구통계학적 특성과 우울증 선별검사 결과

연구목적 : 1998년 정신건강의 날 기념 한국인 우울증 선별의 날 행사에 자발적으로 참여한 참가자들의 사회인구 통계학적 특성을 분석하고 우울증 선별검사 결과를 평가하기 위하여 본 연구를 시행하였다. 방 법 : 8개 병원에서 얻어진 619명의 자료를 사용하여 선별검사로 밝혀진 우울증의 유병율과 사회인구통계학적 특성 및 응답자의 정신과적 치료를 포함한 과거력 등을 조사하였다. 결 과 : 우울증 척도 점수는 응답자 전체 평균 57.5±13.8로 경도의 우울 증상이 있는 것에 해당되었다. 전체 응답자의 69.5%(430명)가 최소한 경도 이상의 우울 증상을, 43.1%(267명)가 중등도 이상의 우울 증상을, 18.4%(114명)가 고도의 우울 증상을 보였다. 고도의 우울 증상이 있는 응답자의 56.8%와 중등도의 우울 증상이 있는 응답자의 63.9%가 정신과 치료를 받은 적이 전혀 없었다. 나이가 29세 이하인 것과 전임근무 중인 직업상태는 각각 나이가 60세 이상인 것과 직업이 없는 것에 비교하여 우울증으로부터 보호하는 인자임을 시사하였다. 결 론 : '1998 한국인 우울증 선별의 날' 행사를 통하여 많은 우울증 환자들을 선별해낼 수 있었고, 이들의 우울 증상은 몇몇 사회인구통계학적 특성과 유의한 관계가 있음을 확인하였다. 우울증 선별의 날 행사를 통한 우울증에 대한 교육과 우울증 선별검사는 치료받지 못하고 있는 우울증 환자 및 기존의 치료 중인 환자 모두에게 유용한 프로그램임을 알 수 있었다. Objectives : This study was performed to analyze the sociodemographic characteristics of participants in '1998 Korean Depression Screening Day' and to evaluate the results of the screening test. Methods : By using the survey results of 619 volunteers from 8 hospitals, the authors examined the prevalence of depression detected at the screening test and sociodemographic characteristics and the psychiatric treatment history of respondents. The assessment measure was the Zung Self-Rating Depression Scale. Results : The mean depression score of all participants was 57.5±13.8 and it came under mild depression. Of all participants, 69.5%(N=430) had at least mild depressive symptoms, 43.1%(N=267) had at least moderate symptoms, and 18.4%(N=114) had severe symptoms. Never have 56.8% of respondents in the severely depressed range and 63.9% of those in the moderately depressed range had psychiatric treatment. The results suggest that the age group of 29-year-old or younger(relative to 60-year-old or older group) and full-time employment status(relative to unemployment) are protective factors of depression. Conclusions : By '1998 Korean Depression Screening Day', many depressed patients were detected and their depressive symptoms had statistically significant relationships with some sociodemographic characteristics. The results suggest that the education and screening test programs for depressive illness facilitated by Depression Screening Day are useful to the patients regardless of being under current treatment or not.

구강암에서 microsatellite 부위 게놈의 불안정성과 p53유전자변이분석

구강암환자의 조기진단 및 예방과 치료에 도움이 되고자 구강편평상피세포암 14례에서 종양억제유전자인 p53,DCC,p16,APC 등이 위치하는 유전좌와 인접 유전좌를 포함한 8부위를 대상으로 구강암조직과 정상조직간의 Microsatellite Instability(이하 MI)분석을 이용하여 게놈의 불안정성을 조사하였으며,대표적인 종양억제유전자인 p53단백의 발현정도를 면역조직화학법을 이용하여 조사하고 p53유전자의 돌연변이양상과 구강암과의 관계를 규명하기 위하여 p53유전자의 exon부위 중 돌연변이가 90%이상 분포하는 exon 5-8을 대상으로 polymerase chain reaction-single stranded conformation polymorphism (PCR-SSCP)분석을 시행하고 band 이동의 이상이 관찰될 경우 직접염기서열분석을 시행하여 돌연변이 양상을 조사하고 이들과 음주,흡연 등과의 관련성을 살펴보았다.14명의 구강편평상피세포암환자의 임상병리학적 소견은 남자가 9례,여자가 5례였으며 평균연령은 60세였다.발생부위는 연구개가 6례,치은 3례,설부 3례,구강저 1례,협점막 1례로 나타났다.병리학적 분화도는 고분화가 8례,중증도분화가 5례,저분화가 1례였으며 남자환자의 경우 8명이 흡연과 음주기간이 최소한 20년이상 되었으며 조직학적으로 임파선으로의 전이가 확인된 환자는 5명 이었다.MI조사에서 1명의 환자에서 조사한 8부위 모두에서 microsatellite 불안정성이 관찰되었으며 11명의 환자(79%)에서 최소한 1유전좌 이상 MI가 관찰되었다.PCR-SSCP분석에서는 14례중 6례(43%)에서 p53유전자의 돌연변이가 관찰되었다.Exon 5에서는 6,7,8,9번 환자에서 band 이동의 변화가 관찰되었으며,exon6의 경우 전 시료에서 band 이동의 변화가 관찰되지 않았다.Exon7의 경우 5,8번 환자에서 band의 이상이동이 관찰되었고,exon8의 경우 7,10번 환자에서 band 이동이상이 관찰되었다.p53 면역조직화학검사에서는 14례 중 5례(36%)에서 p53유전자의 과발현이 관찰되었다.임파절 전이가 일어난 4례는 p53의 과발현이나 돌연변이가 관찰되었다.6례는 면역조직화학검사법과 PCR-SSCP분석에서 단백의 과발현이나 band이동의 변화가 관찰되지 않았으며,3례에서 단백의 과발현과 band 이동이상이 모두 관찰되었으며 5례에서 두 검사방법중 한 방법에서 이상이 관찰되었다.p53 유전자의 돌연변이 양상은 점돌연변이에 의한 nonsense,missense돌연변이 3례외에 삽입과 결손에 의한 frameshift 4례와 splicing miss1례 등 다양하게 관찰되었다.흡연 및 음주경력과 구강암 발생과의 연관성에서는 9명의 남자환자중 8명이 최소한 20년이상의 흡연과 음주경력을 보여 구강암발생과의 깊은 연관성을 보이고 있으며,MI분석에서 흡연자 8명중 7명이 유전자의 불안정성을 보였으며,8명중 6명에서 p53단백의 과발현이나 p53 exon5-8에서 천이에 의한 missense mutation,삽입,결손,splicing miss등의 다양한 형태의 돌연변이양상을 보였다.

魚油함유 飼料에 첨가한 α- 토코페롤의 含量이 흰쥐 血液의 肢質過酸化反應에 미치는 影響

어유와 돈지의 혼합지질(3 : 1, wt/wt: P/S : 1.6)에 α-토코페롤을 용량별(0, 50, 100, 200, 400, 800mg/kg diet)로 첨가하여 조제한 어유 함유 사료로써 7주간 사육하여 이 사료에 혼합한 α-토코페롤의 첨가량이 혈청 중의 지질대사에 미치는 영향을 검토하였다. 중성지질(TG)과 인지질의 함량은 α-토코페롤 200mg 이상의 첨가군은 대조군에 비하여 유의적으로 감소하였고, 또한 총콜레스테롤의 함량은 α-토코페롤의 첨가에 따라 감소하는 경향이었지만 낮은 첨가농도에서만 유의성이 인정되었다. HDL-콜레스테롤의 함량은 100mg 이상의 α-토코페롤 첨가군에서 총콜레스테롤에 대한 HDL-콜레스테롤의 비(HDL-chol/T. chol)도 α-토코페롤의 모든 첨가군에서 유의적으로 증가효과가 인정되었다. 따라서 동맥경화 지표가 α-토코페롤 첨가군에서 유의적으로 감소하였다. 중성지질에 대한 과산화지질(TBA value/TG)의 비는 10mg 이상의 α-토코페롤의 첨가군에서 유의적으로 억제되???고, 중성지질에 대한 α-토코페롤(α-tocopherol/TG)의 비는 100mg 이상의 α-토코페롤 첨가군에서 유의적으로 증가되었다(p<0.001). 어유 함유 사료에 α-토코페롤을 첨가함으로써 C20 : 5/C20 : 4의 비를 효과적으로 억제할수 있었다. 따라서 혈액 중의 적절한 지질대사를 위해서는 이들 어유 함유 사료에 100mg 이상의 α-토코페롤이 첨가되어야 할것으로 생각된다. To evaluate the vitamin E requirement, Fischer 344 male rats were fed diets containing the fish oil-lard (3 : I, wt/wt: P/S ratio : 1.6) supplemented with 0, 50, 100, 200. 400 and 800 mg/kg diet of a-tocopherol for 7 weeks. The effects of a-tocopherol levels supplemented to fish oil rich diets on the lipid metabolism in serum were investigated. Triglyceride (TG) and total cholesterol significantly decreased by supplementation of a-tocopherol. The a-tocopherol supplementation of abme 100 mg diet caused an incressis in HDL-cholesterol. and HDL-cholesterol : T. chol ratio, and a decrease in the atherogenic index. The lipid peroxide : TG ratio significantly inhibited by a-tocopherol supplementation of above 100 mg. The ratios of α-tocopherol/TG or n-3 fatty acid (EPA+ DHA) significantly increased in all α-tocopherol supplemented groups. The ratio of eicosapentaenoic acid : arachidonic acid effectively inhibited by a-tocopherol supplementation of above 100 mg. The results suggest that a-tocopherol supplementation of above 100 mg to fish oil-rich diets map be requisite for adequate lipid metabolism.

This experiment was done on 30 fencers that had joined the national competition and had a research on theft heart rates during the fencing bout and got the following results. 1. In average, a male fencer have a height of 178.40㎝, weight of 72.50㎏, the height when sitting of 93.70㎝, the chest perimeter of 94.70㎝, arm length of 56.50㎝, and thigh parameter of 60.20㎝. In average, for a female fencer, had a height of 167.98㎝, weight of 59.00㎏, the height when sitting of 87.00㎝, chest perimeter of 84.50㎝, the thigh perimeter of 58.30㎝, and the arm length of 50.90㎝. From the result of testing a fencer's endurance by general footwork, it was proofed that the evaluation of respiratory function was needed more. 3. When comparing the heart rates of male fencers in the fencing bout of fleuret and epee, we found out that the fleuret fencer had a higher heart rate, in average of 17.06beat/min, than the epee fencer. However, there was not much difference with the heart rates during the bouts of sabre and fleuret Clearly, the bout during epee showed lowest heart rate among these three subjects. 4. The heart rates during female's fencing bout, there was not much difference with the heart rates in the male's fencing bout, and the heart rate during a bout of epee was about 18.59beat/min lower than fleuret.

Tsukamurella sp. 26A에 의한 생물계면활성제의 생산

The strain producing biosurfactant was isolated from soil. The isolated strain was identified as the genus Tsukamurella through its morphological, cultural, physiological, menaquinone type, fatty acid composition characteristics. The highest biosurfactant production by Tsukamurella sp. 26A was observed after 4 days cultivation in the culture medium containing n-hexadecane 7%, NaNO₃0.2%, KH₂PO₄0.001%, K₂HPO₄0.02%, MgSO₄·7H₂O 0.02%, CaCl₂·2H₂O 0.02%, yeast extract 0.02%(pH 6.8-7.0, 30℃). The surface and interfacial tension of an aqueous solution reached 30 mN/m and 1.5 mN/m, respectively. The biosurfactant stabilized oil-in-water emulsion with a variety of hydrocarbones, edible oils and petroleum oils.
