        Effects of Tween 80 on In Vitro Fermentation of Silages and Interactive Effects of Tween 80, Monensin and Exogenous Fibrolytic Enzymes on Growth Performance by Feedlot Cattle

        The effects of monensin, Tween 80 and exogenous fibrolytic enzymes on ruminal fermentation and animal performance were studied in vitro and in vivo. In Expt 1, the effects of the surfactant Tween 80 (0.2% wt/wt, DM basis) on ruminal fermentation of alfalfa, corn and orchardgrass silages were investigated using in vitro gas production techniques. Tween 80 did not affect (p>0.05) cumulative gas production at 24 h, but it reduced (p<0.05) the lag in fermentation of all three silages. With corn silage and orchardgrass silage, gas production rates and concentrations of total volatile fatty acids (VFA) were increased (p<0.05) by Tween 80; with alfalfa silage, they were reduced (p<0.05). Tween 80 increased (p<0.05) the proportion of propionate in total VFA, and reduced (p<0.05) acetate to propionate ratios (A:P) with all three silages. In Expt 2, exogenous fibrolytic enzymes (E; at 0, 37.5 or 75 g/tonne DM), monensin (M; at 0 or 25 ppm and Tween 80 (T; at 0 or 2 L/tonne DM) were added alone or in combination to backgrounding and finishing diets fed to 320 crossbred steers in a feeding trial with a $3{\times}2{\times}$2 factorial arrangement of treatments. The backgrounding and finishing diets contained barley grain and barley silage in ratios of 57.8:42.2 and 93.5:6.5 (DM basis), respectively. Added alone, none of the additives affected DM intake (p>0.1) in the backgrounding or in the finishing period, but interactive $M{\times}T$ effects were observed in the finishing period (p=0.02) and overall (p=0.04). In the finishing period, T without M tended to reduce DM intake (p=0.11), but T with M increased (p=0.05) DM intake. Monensin increased average daily gain (ADG) during backgrounding (p=0.07) and finishing (p=0.01), and this ionophore also improved overall feed efficiency (p=0.02). Warm carcass weight was increased (p<0.001) by M, but dressing percentage was reduced (p=0.07). In the backgrounding period, T increased ADG by 7% (p=0.06). Enzymes increased (p=0.07) ADG by 5 and 6% (low and high application rates, respectively) during backgrounding, but did not affect (p>0.10) ADG during finishing, or overall feed efficiency. Whereas T enhanced the positive effects of M on ADG during backgrounding (p=0.04) and overall (p=0.05), it had no impact (p>0.1) on the effects of E. Interactions between M and T suggest that the surfactant may have potential for enhancing the positive effects of monensin on beef production, but this requires further research.

        Milk Conjugated Linoleic Acid (CLA) Profile and Metabolic Responses of Dairy Cows Fed with High-temperature-micro-time (HTMT) Treated Diets Containing High Quantity Extruded Soybean (ESB)

        A feeding trial was conducted to examine the effect of high-temperature-micro-time (HTMT) processing of diets containing extruded soybean (ESB) in high quantity on milk fat production, metabolic responses, and the formation of conjugated linoleic acid (CLA) and trans-vaccenic acid (TVA). Twenty-one multiparous Holstein cows in mid-lactation were blocked according to milk yield in the previous lactation. Cows within each block were randomly assigned to either normal concentrate or HTMT treated diets containing ESB (7.5% HTMT-ESB and 15% HTMT-ESB). It was hypothesized that the HTMT-ESB would affect the undegradable fatty acids in the rumen and, thus, would modify the fatty acid profile of milk fat. Both 7.5% and 15% HTMT-ESB did not affect milk yield, fat, protein, lactose and solid-not-fat (SNF), but the proportion of cis-9, trans-11 CLA in milk fat was significantly increased by these treatments. Content of TVA in milk fat was not affected by HTMT-ESB. The HTMT-ESB influenced the fatty acid profile in milk fat, but there was little difference between 7.5% and 15% of supplementation. HTMT-ESB feeding significantly decreased the concentration of plasma insulin and glucose, while plasma growth hormone (GH), triglyceride (TG), non-esterified fatty acid (NEFA) and HDLcholesterol were increased by 7.5% and 15% ESB-HTMT supplementation in comparison to the control group (p<0.05). However, no significant difference was observed in plasma LDL-cholesterol, insulin like growth factor (IGF)-1, T3, T4, and leptin concentrations among treatments (p>0.05). The present results showed that cis-9, trans-11 CLA production was increased by HTMT treatment of dietary ESB without reduction of milk fat, and the unchanged milk fat and yield was assumed to be associated with the constant level of thyroid hormones, leptin, and IGF-1.

      • Development of A Chimeric Antigen Receptor Targeting C-Type Lectin-Like Molecule-1 for Human Acute Myeloid Leukemia

        <P>The treatment of patients with acute myeloid leukemia (AML) with targeted immunotherapy is challenged by the heterogeneity of the disease and a lack of tumor-exclusive antigens. Conventional immunotherapy targets for AML such as CD33 and CD123 have been proposed as targets for chimeric antigen receptor (CAR)-engineered T-cells (CAR-T-cells), a therapy that has been highly successful in the treatment of B-cell leukemia and lymphoma. However, CD33 and CD123 are present on hematopoietic stem cells, and targeting with CAR-T-cells has the potential to elicit long-term myelosuppression. C-type lectin-like molecule-1 (CLL1 or CLEC12A) is a myeloid lineage antigen that is expressed by malignant cells in more than 90% of AML patients. CLL1 is not expressed by healthy Hematopoietic Stem Cells (HSCs), and is therefore a promising target for CAR-T-cell therapy. Here, we describe the development and optimization of an anti-CLL1 CAR-T-cell with potent activity on both AML cell lines and primary patient-derived AML blasts in vitro while sparing healthy HSCs. Furthermore, in a disseminated mouse xenograft model using the CLL1-positive HL60 cell line, these CAR-T-cells completely eradicated tumor, thus supporting CLL1 as a promising target for CAR-T-cells to treat AML while limiting myelosuppressive toxicity.</P>

        In the last ten years there has been a dramatic change in the electrical industry regarding its technology and capability. VLSI circuits technologies have brought about the develoment of computers, digital communications, digital control systems, and others. The microprocessor now is appearing in the simplest appliances as well as in complex systems, permeating all of engineering. This paper is presented as a proposal for a more improved education courses for technical high school students preparing to enter this dynamic electronic industry. The current curricula, and their syllabuses, of the electronic deparments of typical technical high schools were analyzed, and their teachers and instructors, as well as technicians in electronic companies, were consulted about the education courses. The results of this study are follow: 1. The fields of digital circuits and systems using IC and LSI should be emphasized in the current curricula and the subject of microprocessor applications should be added. 2. The current ratio of the theoretical; subject hours vs laboratory /shop practice hours should be rearranged at about same ratio through effective laboratory systems. 3. A retraining course for experimental instructors and supervisors should be provided, similar to that provided for teachers. 4. Technical high school education should avoid the erroneous philosophy of training the student only sufficiently to pass the qualification examination.

        <P>Development of advanced high energy density lithium ion batteries is important for promoting electromobility. Making electric vehicles attractive and competitive compared to conventional automobiles depends on the availability of reliable, safe, high power, and highly energetic batteries whose components are abundant and cost effective. Nickel rich Li[NixCoyMn1−x−y]O2 layered cathode materials (<I>x</I> > 0.5) are of interest because they can provide very high specific capacity without pushing charging potentials to levels that oxidize the electrolyte solutions. However, these cathode materials suffer from stability problems. We discovered that doping these materials with tungsten (1 mol%) remarkably increases their stability due to a partial layered to cubic (rock salt) phase transition. We demonstrate herein highly stable Li ion battery prototypes consisting of tungsten-stabilized Ni rich cathode materials (<I>x</I> > 0.9) with specific capacities >220 mA h g<SUP>-1</SUP>. This development can increase the energy density of Li ion batteries more than 30% above the state of the art without compromising durability.</P>

        Ultrafast Pump-Probe Study of Phase Separation and Competing Orders in the Underdoped(Ba,K)Fe2As2Superconductor

        <P>We report measurements of quasiparticle relaxation dynamics in the high-temperature superconductor (Ba,K)Fe2As2 in optimally doped, underdoped, and undoped regimes. In the underdoped sample, spin-density wave (SDW) order forms at approximately 85 K, followed by superconductivity at approximately 28 K. We find the emergence of a normal-state order that suppresses SDW at a temperature T{*} approximately 60 K and argue that this normal-state order is a precursor to superconductivity.</P>

        Association between Genetic Polymorphism in the Swine Leukocyte Antigen-DRA Gene and Piglet Diarrhea in Three Chinese Pig Breeds

        The swine leukocyte antigen (SLA)-DRA locus is noteworthy among other SLA class II loci for its limited variation and has not been investigated in depth. This study was investigated to detect polymorphisms of four exons of SLA-DRA gene and its association with piglet diarrhea in Landrace, Large White and Duroc pigs. No polymorphisms were detected in exon 3, while 2 SNPs (c.178G>A and c.211T>C), 2 SNPs (c.3093A>C and c.3104C>T) and 5 SNPs (c.4167A>G, c.4184A>G, c.4194A>G, c.4246A>G and c.4293G>A) were detected in exon 1, exon 2 and exon 4 respectively, and 1 SNP (c.4081T>C) in intron 3. Statistical results showed that genotype had significant effect on piglet diarrhea, individuals with genotype BC had a higher diarrhea score when compared with the genotypes AA, AB, AC and CC. Futhermore, genotype AC had a higher diarrhea score than the genotype CC in exon 1 (p<0.05); diarrhea scores of genotype AA and BB were higher than those of genotypes AC and CC in exon 2 (p<0.05); individuals with genotype AA had a higher diarrhea score than individuals with genotype AB and BB in exon 4 (p<0.05). Fourteen common haplotypes were founded by haplotype constructing of all SNPs in the three exons, its association with piglet diarrhea appeared that Hap2, 5, 8, 10, and 14 may be the susceptible haplotypes and Hap9 may be the resistant haplotype to piglet diarrhea. The genetic variations identified of the SLA-DRA gene may potentially be functional mutations related to piglet diarrhea.

        Influence of residual stress on the adhesive behavior of detonation gun sprayed WC-Co coatings

        The residual stresses generated during D-Gun spraying process act as pre-existing stresses and seriously affect the various types of coating performance. To produce a reliable coating, it is necessary to know the influence of residual stress on the adhesive behavior of coating/substrate system. Based on the residual stress formula for thick coatings and accurate curvature measurements, the residual stresses of D-Gun sprayed WC-Co coatings were obtained. And a strong link was built between the residual stress and the adhesive strength of the coating, namely, the compressive stress in the coating could significantly improve the adhesion, whereas the tensile stress weakened the adhesion. The larger compressive residual stress is in the coating, the stronger adhesion between the coating and substrate.

        Proteomic Analysis Reveals PGAM1 Altering cis-9, trans-11 Conjugated Linoleic Acid Synthesis in Bovine Mammary Gland.

        <P>cis-9, trans-11 Conjugated linoleic acid (CLA) is one of the most extensively studied CLA isomers due to its multiple isomer-specific effects. However, the molecular mechanisms of cis-9,trans-11 CLA synthesis in ruminant mammary gland are still not clearly understood. This process may be mediated, to a certain extent, by trans-11 C18:1 regulated by stearoyl-CoA desaturase-1 (SCD1) and/or its syntrophic proteins. This study aimed to investigate the effects of TVA on SCD1-mediated cis-9,trans-11 CLA synthesis in MAC-T cells and its potential molecular mechanism. Results showed that trans-11 C18:1 was continually taken up and converted into cis-9,trans-11 CLA in MAC-T cells during the 4-h incubation of 50?μM trans-11 C18:1. SCD1 protein expression increased more than twofold at 2?h (P?<?0.01) and 2.5?h (P?<?0.05) before decreasing to less than half of the normal level at 4?h (P?<?0.05). One up-regulated (RAS guanyl releasing protein 4 isoform 1 [RASGRP4]) and six down-regulated proteins (glucosamine-6-phosphate deaminase 1 [GNPDA1], triosephosphate isomerase [TPI1], phosphoglycerate mutase 1 [PGAM1], heat shock protein beta-1 [HSPB1], annexin A3 [ANXA3], thiopurine S-methyltransferase [TPMT]) were found in MAC-T cells treated with trans-11 C18:1. Of these seven identified proteins, the presence of GNPDA1 and PGAM1 was verified in several models. More trans-11 C18:1 was taken up after PGAM1 knockdown by small interfering RNA (siRNA). In conclusion, our data suggested that PGAM1 may have a negative relationship with SCD1 and seemed to be involved in cis-9, trans-11 CLA synthesis by facilitating the absorption of trans-11 C18:1 in the bovine mammary gland.</P>
