집중호우가 발생할 때 유역의 특성을 고려한 재해 대책이 필요하다. 침수구역을 객관적으 로 평가하기 위해 지형 특성과 지형 정보를 활용하였다. 적정한 침수구구역의 설정은 침수 방지 시설을 갖추기 위한 선행 작업이다. 이 작업을 위해 최근에 개발된 Arc-Gis를 이용하 여 침수구역을 설정하였다. 이 방법을 증암천 유역에 적용하여 위험구역을 설정하였다. There is require disaster prevention considering the characteristics of the watershed when heavy rains occur. We have to objectively evaluate the flooded area using topographic characteristics and geographic information. A set of appropriate flooded area should be preceded for facilities of flood prevention. We have established a flooded area using the latest version Arc-Gis. This software was applied to the classification for flood hazard area at Jeungam-stream basin.

<P>Pooled CRISPR screens based on lentiviral systems have been widely applied to identify the effect of gene knockout on cellular phenotype. Although many screens were successful, they also have the limitation that genes conferring mild phenotypes or those essential for growth can be overlooked, as every genetic perturbation is incorporated in the same population. Arrayed screens, on the other hand, incorporate a single genetic perturbation in each well and could overcome these limitations. However, arrayed screens based on siRNA-mediated knockdown were recently criticized for low reproducibility caused by incomplete inhibition of gene expression. To overcome these limitations, we developed a novel arrayed CRISPR screen based on a plasmid library expressing a single guide RNA (sgRNA) and disrupted 1514 genes, encoding kinases, proteins related to endocytosis, and Golgi-localized proteins, individually using 4542 sgRNAs (three sgRNAs per gene). This screen revealed host factors required for infection by coxsackievirus B3 (CVB3) from Picornaviridae, which includes human pathogens causing diverse diseases. Many host factors that had been overlooked in a conventional pooled screen were identified for CVB3 infection, including entry-related factors, translational initiation factors, and several replication factors with different functions, demonstrating the advantage of the arrayed screen. This screen was quite reliable and reproducible, as most genes identified in the primary screen were confirmed in secondary screens. Moreover, ACBD3, whose phenotype was not affected by siRNA-mediated knockdown, was reliably identified. We propose that arrayed CRISPR screens based on sgRNA plasmid libraries are powerful tools for arrayed genetic screening and applicable to larger-scale screens.</P>

We investigated the (ballistic) current-voltage (I-V ) characteristics in a 100-nm ..-gate AlGaAs/InGaAs Pseudomorphic high-electron-mobility transistor (pHEMT) as a function of temperature for temperatures ranging from 300 K to 10 K and analyzed the transport in terms of carrier backscattering. We observed that the drain current in the linear region dramatically increased below 60 K due to reduced phonon scattering. The critical temperature where the drain current started to rapidly increase shifted to a higher value as the drain voltage was increased. These results are in good agreement with the ballistic transport theory in which low temperatures and high electric fields along the channel lead to enhanced ballistic transport due to a reduction in the backscattering probability. We also carried out self-consistent Schrodinger-Poisson band profile calculations to understand the experimental data.

본 논문에서는 한국형발사체 2단에 대한 부분체 구조 시험치구의 기본설계 결과를 표현하고 있다. 단일하중 100톤, 최대 500톤 규모의 강도 및 강성시험을 수행할 수 있도록 설계되었고, 시험체 접근성과 Setup 편리성 극대를 위한 Handling Device가 설치되었다. 기본설계를 통해 개발된 기술은 하중의 일정분포를 유지하는 상/하부 하중부가장치 기술와 복합하중 및 중량보상장치, 시험장비 구성설계와 시험체 안전장치 기술이다. 본 기본설계 결과는 시험수행을 통해 유효성이 확인될 것을 기대한다.
Evaluation of Geographic Indices Describing Health Care Utilization

Objectives: The accurate measurement of geographic patterns of health care utilization is a prerequisite for the study of geographic variations in health care utilization. While several measures have been developed to measure how accurately geographic units reflect the health care utilization patterns of residents, they have been only applied to hospitalization and need further evaluation. This study aimed to evaluate geographic indices describing health care utilization. Methods: We measured the utilization rate and four health care utilization indices (localization index, outflow index, inflow index, and net patient flow) for eight major procedures (coronary artery bypass graft surgery, percutaneous transluminal coronary angioplasty, surgery after hip fracture, knee replacement surgery, caesarean sections, hysterectomy, computed tomography scans, and magnetic resonance imaging scans) according to three levels of geographic units in Korea. Data were obtained from the National Health Insurance database in Korea. We evaluated the associations among the health care utilization indices and the utilization rates. Results: In higher-level geographic units, the localization index tended to be high, while the inflow index and outflow index were lower. The indices showed different patterns depending on the procedure. A strong negative correlation between the localization index and the outflow index was observed for all procedures. Net patient flow showed a moderate positive correlation with the localization index and the inflow index. Conclusions: Health care utilization indices can be used as a proxy to describe the utilization pattern of a procedure in a geographic unit.
Rhythmic control of AANAT translation by hnRNP Q in circadian melatonin production.

<P>The circadian rhythm of pineal melatonin requires the nocturnal increment of serotonin N-acetyltransferase (arylalkylamine N-acetyltransferase [AANAT]) protein. To date, only limited information is available in the critical issue of how AANAT protein expression is up-regulated exclusively at night regardless of its species-specific mRNA profiles. Here we show that the circadian timing of AANAT protein expression is regulated by rhythmic translation of AANAT mRNA. This rhythmic control is mediated by both a highly conserved IRES (internal ribosome entry site) element within the AANAT 5' untranslated region and its partner hnRNP Q (heterogeneous nuclear ribonucleoprotein Q) with a peak in the middle of the night. Consistent with the enhancing role of hnRNP Q in AANAT IRES activities, knockdown of the hnRNP Q level elicited a dramatic decrease of peak amplitude in the AANAT protein profile parallel to reduced melatonin production in pinealocytes. This translational regulation of AANAT mRNA provides a novel aspect for achieving the circadian rhythmicity of vertebrate melatonin.</P>

<P><B>Purpose</B></P><P>To evaluate the impact of temporary internal ureteral stents on the surgical outcomes of dismembered pyeloplasty in children.</P><P><B>Materials and Methods</B></P><P>The medical records of 70 children (76 renal units) who underwent dismembered pyeloplasty for ureteropelvic junction (UPJ) obstruction at at Asan Medical Center between January 2005 and December 2010 were retrospectively reviewed. We classified the renal units into the stented group (22 renal units) and the nonstented group (54 renal units). Fifty-four of 70 patients were male and their mean age was 2.2±3.8 years old. The mean follow-up period was 29.6±16.8 months.</P><P><B>Results</B></P><P>Sixty-four children had unilateral UPJ obstruction. The mean stent duration was 31.9 days. As shown by evaluation of radiologic images, there were no significant differences between the stented group and the nonstented group during the follow-up period (p>0.05). The mean preoperative and postoperative anteroposterior pelvic diameters (APPDs) of the nonstented group were 31.3 mm and 15.1 mm, respectively (p<0.001). The preoperative and postoperative grades of hydronephrosis were 3.9 and 2.9, respectively (p=0.037). The mean preoperative and postoperative APPDs of the stented group were 36.4 mm and 15.6 mm, respectively (p<0.001). The preoperative and postoperative grades of hydronephrosis were 4 and 3.1, respectively (p<0.001). Repeat obstruction was shown in 4 subjects as a postoperative complication (5.7%). Two children from each group had recurrent UPJ obstruction, with percentages of 3.7% and 9%, respectively (p=0.575).</P><P><B>Conclusions</B></P><P>In a comparison of nonstented and stented groups during pediatric dismembered pyeloplasty for UPJ obstruction, no significant differences were found in the resolution of hydronephrosis or overall postoperative complications.</P>

<P>Diacylglycerol acyltransferase 2 (DGAT2), which catalyzes the final step in triacylglycerol (TG) synthesis, is a key enzyme associated with hepatic steatosis and insulin resistance. Here, using an in vitro screen of 20000 molecules, we identified a class of compounds with a substituted 1H-pyrrolo[2,3-b]pyridine core which proved to be potent and selective inhibitors of human DGAT2. Of these compounds, H2-003 and -005 exhibited a considerable reduction in TG biosynthesis in HepG2 hepatic cells and 3T3-L1 preadipose cells. These compounds exert DGAT2-specific-inhibitory activity, which was further confirmed in DGAT2- or DGAT1-overexpressing HEK293 cells. In addition, these compounds almost completely abolished lipid droplet formation in 3T3-L1 cells when co-treated with a DGAT1 inhibitor, which was not attained using either a DGAT2 or DGAT1 inhibitor alone. Collectively, we identified two DGAT2 inhibitors, H2-003 and -005. These compounds will aid in DGAT2-related lipid metabolism research as well as in therapeutic development for the treatment of metabolic diseases associated with excessive TG.</P>
