This study was performed to select single nucleotide polymorphisms (SNPs) related to the body burden of heavy metals in Koreans, to provide Korean allele frequencies of selected SNPs, and to assess the difference in allele frequencies with other ethnicities. The candidate-gene approach method and genome-wide association screening were used to select SNPs related to the body burden of heavy metals. Genotyping analysis of the final 192 SNPs selected was performed on 1,483 subjects using the VeraCode Goldengate assay. Allele frequencies differences and genetic differentiations between the Korean population and Chinese (CHB), Japanese (JPT), Caucasian (CEU), and African (YIR) populations were tested by Fisher’s exact test and fixation index (FST), respectively. The Korean population was genetically similar to the CHB and JPT populations (FST < 0.05, for all SNPs in both populations). However, a significant difference in the allele frequencies between the Korean and CEU and YIR populations were observed in 99 SNPs (60.7%) and 120 SNPs (73.6%), respectively. Ten (6.1%) and 26 (16.0%) SNPs had genetic differentiation (FST > 0.05) among the Korean-CEU and Korean-YIR comparisons, respectively. The SNP with the largest FST value between the Korean and African populations was cystathionine-β-synthase rs234709 (FST: KOR-YIR, 0.309; KOR-CEU, 0.064). Our study suggests that interethnic differences exist in SNPs associated with heavy metals of Koreans, and it should be considered in future studies that address ethnic differences in heavy-metal concentrations in the body and genetic susceptibility to the body burden of heavy metals.
Fraxinellone Attenuates Rheumatoid Inflammation in Mice

<P>This study aimed to evaluate the therapeutic effect of fraxinellone on inflammatory arthritis and identify the underlying mechanisms. Fraxinellone (7.5 mg/kg) or a vehicle control was injected into mice with collagen-induced arthritis (CIA). The severity of arthritis was evaluated clinically and histologically. The differentiation of CD4<SUP>+</SUP> T cells and CD19<SUP>+</SUP> B cells was investigated in the presence of fraxinellone. Osteoclastogenesis after fraxinellone treatment was evaluated by staining with tartrate-resistant acid phosphatase (TRAP) and by measuring the mRNA levels of osteoclastogenesis-related genes. Fraxinellone attenuated the clinical and histologic features of inflammatory arthritis in CIA mice. Fraxinellone suppressed the production of interleukin-17 and the expression of <I>RAR-related orphan receptor γ t</I> and phospho-signal transducer and activator of transcription 3 in CD4<SUP>+</SUP> T cells. CD19<SUP>+</SUP> B cells showed lower expression of <I>activation-induced cytidine deaminase</I> and <I>B lymphocyte-induced maturation protein-1</I> after treatment with fraxinellone. The formation of TRAP-positive cells and the expression of osteoclastogenesis-related markers were reduced in the presence of fraxinellone. Inhibition of interleukin-17 and osteoclastogenesis was also observed in experiments using human peripheral mononuclear cells. Fraxinellone alleviated synovial inflammation and osteoclastogenesis in mice. The therapeutic effect of fraxinellone was associated with the inhibition of cellular differentiation and activation. The data suggests that fraxinellone could be a novel treatment for inflammatory arthritis, including rheumatoid arthritis.</P>
Rapid identification and preparative isolation of antioxidant components in licorice

<P>This study employed the online HPLC-2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonate radical cation (ABTS<SUP>+</SUP><SUP>·</SUP>) bioassay to rapidly determine antioxidant compounds occurring in the licorice extract of Glycyrrhiza uralensis. The negative peaks of the ABTS<SUP>+·</SUP> radical scavenging detection system, which indicated the presence of antioxidant activity, were monitored by measuring the decrease in absorbance at 734 nm. The ABTS<SUP>+</SUP>-based antioxidant activity profile showed that three peaks exhibited antioxidant activity, and then the high-speed counter-current chromatography technique of preparative scale was successfully applied to separate the three peaks I-III in one step from the licorice extract. The high-speed counter-current chromatography was performed using a two-phase solvent system composed of n-hexane–ethyl acetate–methanol–water (6.5:5.5:6:4, v/v). Yields of the three peaks, dehydroglyasperin C (I, 95.1% purity), dehydroglyasperin D (II, 96.2% purity), and isoangustone A (III, 99.5% purity), obtained were 10.33, 10.43, and 6.7% respectively. Chemical structures of the purified dehydroglyasperin C (I), dehydroglyasperin D (II), and isoangustone A (III) were identified by ESI-MS and <SUP>1</SUP>H- and <SUP>13</SUP>C-NMR analysis.</P>
A Bayesian Approach to Replacement Policy Based on Cost and Downtime

This paper considers a Bayesian approach to replacement policy model with minimal repair. We use the criterion based on the expected cost and the expected downtime to determine the optimal replacement period. To do so, we obtain the expected cost rate per unit time and the expected downtime per unit time, respectively. When the failure time is Weibull distribution with uncertain parameters, a Bayesian approach is established to formally express and update the uncertain parameters for determining an optimal maintenance policy. Especially, the overall value function suggested by Jiagn and Ji(2002) is applied to obtain the optimal replacement period. The numerical examples are presented for illustrative purpose.

Antioxidant and hepatoprotective activities of the fruiting bodies of Paecilomyces japonica (PJ) grown on silk-worm larvae and of Cordyceps sinensis (CS), a wild form of Cordyceps, were investigated. The 70% ethanol extracts of both fungi, PJ and CS, were found to exhibit free radical scavenging effects on 1,1-diphenyl-2-picrylhydrazyl (DPPH). The inhibitory potency of PJ (IC?? = 163 ㎍ㆍ㎖?¹) was approximately 4.5 folds that of CS (IC?? = 731 ㎍ㆍ㎖?¹). They exhibited a moderate decrease in serum transaminase activities elevated by hepatic damage induced by carbon tetrachloride (CCl₄)-intoxication in rats. Both fungal extracts also exhibited not only significant increases in hepatic superoxide dismutase (SOD) and catalase at a dose of 300 ㎍ㆍ㎏?¹ in CCl₄-intoxicated rats, but also a significant decrease in malondialdehyde (MDA) production in a thiobarbituric acid (TBA) reactant assay in rats. Although no significant glutathione peroxidase (GSH-px) activity was detected, SOD and catalase elevations were obtained.
Adhesion Change of AZO/PET Film by ZrCu Insertion Layer

In order to form an aluminum-doped zinc oxide (AZO) transparent electrode film on a polyethylene terephthalate (PET) substrate used for a flexible display substrate, the AZO transparent electrode was produced at low temperature without substrate heating. Even though the produced electrode showed characteristic optical transmittance of 90 % (at 550 nm) and sheet resistance within 100 Ω/sq, cracks occurred 10 minutes after loading applied 2 mm radius of curvature, and the sheet resistance increased linearly. An insertion layer of ZrCu was formed between the AZO film and the PET substrate to suppress the generation of cracks on the AZO film. It was verified that the crack was not generated 30 minutes after the loading of 2 mm radius of curvature, and no increase in sheet resistance was recorded. There was also not cracks in the dynamic bending test of 4 mm radius, but surface resistance was slightly increased. As a result, the ZrCu insertion film improved the interfacial adhesion between the substrate and AZO film layer without increasing sheet resistance and decreasing transmittance.
