Morphological and Molecular Classifications of Genus Pholis

Morphological and molecular classifications were attempted in an effort to establish species-specific classifications of three species of the genus Pholis in Korea; these species were subjected to morphological and molecular methodologies using body measurements, RFLP, RAPD, and phylogenetic trees using the nucleotide sequences of mitochondrial 16S and 12S ribosomal DNAs, cytochrome c oxidase I, and cytochrome b. The data demonstrated that the three species of genus Pholis are distinct from each other, both morphologically and genetically.

<B>ABSTRACT</B><P>A nested PCR assay was developed for the detection of spotted fever group (SFG) rickettsiae in serum samples. The assay was based on specific primers derived from the rickettsial outer membrane protein B gene (<I>rompB</I>) of <I>Rickettsia conorii</I>. An SFG rickettsia-specific signal is obtained from <I>R. akari</I>, <I>R. japonica</I>, <I>R. sibirica</I>, and <I>R. conorii</I>. Other bacterial species tested did not generate any signal, attesting to the specificity of the assay. As few as seven copies of the <I>rompB</I> gene of <I>R. conorii</I> could be detected in 200 μl of serum sample. The assay was evaluated with a panel of sera obtained from patients with acute-phase febrile disease tested by immunofluorescent antibody assay (IFA). The SFG rickettsia-specific DNA fragment was detected in 71 out of 100 sera, which were proven to have immunoglobulin M antibodies against SFG rickettsial antigen by IFA. The results were further confirmed by restriction fragment length polymorphism and sequencing analysis of the DNA fragments. The results indicated that this PCR assay is suitable for the diagnosis of spotted fever group rickettsiosis in Korea.</P>

To investigate the function of the Src Homology 2 (SH2) domains found in various non-receptor protein tyrosine kinases (PTKs) and many other cytoplasmic signalling proteins, the partial sequence of the v-yes oncogene containing the SH2 domain has been expressed in E. coli as a fused protein. The 456 by Sau3AI-HaeIII fragment of v-yes was inserted into BamHI-SmaI site of the pGEX-2T vector which has tac promoter and makes the fused proteins with glutathione S-transferase (GST). The fused protein was induced by IPTG and purified from E.coli crude extracts with glutathione-agarose beads. The GST moiety of the fused protein was excised by thrombin treatment. To assay the affinity of the expressed SH2 domain to the tyrosine-phosphorylated proteins, horse raddish peroxidase (HRP)-conjugated SH2 domain was used in detecting phosphotyrosines of enzymatically phosphorylated glutamate-tyrosine copolymers (PGT).

<P><B>Abstract</B></P><P>S-1 is a novel, oral fluoropyrimidine and a known radiosensitizer. We conducted a phase I trial to establish a schedule of S-1/irinotecan with standard pelvic radiotherapy as a preoperative treatment of locally advanced rectal cancer. Our findings suggest that this new combination is feasible and well tolerable.</P>

<P>Thymosin beta 4 (T beta 4) is known to inhibit an inflammatory response and to increase the survival of osteoblasts on titanium (Ti) surfaces. Ti is the most widely used graft material in dentistry; however, an inflammatory response induced following implant placement results in the generation of reactive oxygen species (ROS). The oxidative stress from the production of ROS such as nitric oxide (NO) and hydrogen peroxide (H2O2) can damage surrounding cells, resulting in implant failure by decreasing cell viability. Thus, the aim of this study was to determine the biological effects of T beta 4 on the oxidative stress induced to MC3T3-E1 preosteoblasts on the Ti surface. Based on an MTT assay and bromodeoxyuridine immunofluorescence staining, T beta 4 was found to increase the proliferation of the H2O2-exposed MC3T3-E1 cells on Ti discs. Reverse transcription-polymerase chain reaction and western blot analyses showed that T beta 4 decreased the mRNA and protein expression levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in H2O2-exposed MC3T3-E1 cells on the Ti discs. T beta 4 inhibited the synthesis of intracellular ROS and the secretion of NO and prostaglandin E-2 (PGE(2)) from H2O2-exposed MC3T3-E1 cells on the Ti discs. In conclusion, T beta 4 inhibits H2O2-induced iNOS and COX-2 expression with a decrease in ROS, NO, and PGE(2) synthesis, which leads to improved cell survival with low cytotoxicity under an oxidative stress condition in MC3T3-E1 cells on the Ti surface. This suggests that T beta 4 may be a crucial molecule to reduce oxidative stress-induced cell damage or hypoxia, leading to promoted osseointegration on the Ti surface during implant placement.</P>

We have studied the motion of charged micro-particles that are immersed in a nematic liquid crystal (LC) and controlled by in-plane field. The LC is an anisotropic liquid such that the viscosity of the LC depends on flow direction, phase of the LC, and temperature, which affects the motion of the charged particles under the influence of electric field. This study shows that the motion of charged particles mainly depends on the applied voltage and the LC phase, but does not show any significant influence from the initial alignment of LC, although one may expect directional difference in drag force due to interaction between LC and particle. The viscosity changes due to temperature variations in nematic phase also show no signification influence on particle velocity when compared to the effect from varying in-plane field strength.

An efficient plant regeneration of C. asiatica was achieved from organogenesis using petiole explants of in vitro plantlet on MS basal medium controled with different plant growth regulators (NAA,2,4-D, IAA kinetin, and BA). Best results that 50%, efficiency of regeneration per explant for regeneration were obtained with IAA $17.13\;{\mu}M$ and BA $8.9\;{\mu}M$. Formation of adventitious shoots via organogenesis from the petiole explant was verified by histological sectioning of plantlets. Regenerated plants were transplanted into soil.
A case of eccrine porocarcinoma

Eccrine porocarcinoma is a rare type of skin cancer arising from sweat glands, represents only 0.005% of epithelial cutaneous neoplasms. It generally present as a single, asymptomatic, nondescript cutaneous lesion on the lower extremities of older adults. The exact etiology of porocarcinoma is unknown. However, it is believed to either arise de novo or by malignant degeneration of a longstanding poroma. Risk factors for eccrine porocarcinoma include family history, immunosuppression, and ultraviolet radiation (UVR). The only way to be sure of the diagnosis is to take biopsy of the tumor. Primary treatment consists of excising the tumor. Surgery is curative in up to 80% of cases. However, about 20% of patients will have local recurrence and metastasis. A 49-year-old male patient visited our department with 2x1.5x1cm sized black colored verrucous surfaced mass on suprapubic region. The onset was about 4years ago. He had no symptoms. Biopsy specimen showed irregular cords and lobules of polygonal basaloid tumor cells that proliferate within the dermis in an asymmetric fashion, also presence of ductal and eccrine differentiation and cytological pleomorphisms. There is no demonstrable metastatic lesion on imaging study. We consulted to department of general surgery for total excision. Herein, we report a rare case of eccrine porocarcinoma.
