Metarrhizium anisopliae(Metschn.) Sorok이 생산하는 Biopolymer YU-122의 생산과 그 특성

토양으로부터 분리한 Metarrhizium anisopliae (Metschn.) Sorok이 생산하는 biopolymer를 정제하여 분석하고, 그의 생산조건을 검토하였다. 탄소원으로 mannitol, 질소원으로는 polypepton을 사용했을 때 가장 많은 biopolymer를 생산하였고 K_2HPO_4 및 CaCl_2를 첨가함으로서 생산량이 더욱 증가하였다. 또한 이 biopolymer를 정제하여 그 분자량과 구성성분을 검토한 결과 Metarrhizium anisopliae(Metschn.) Sorok이 생산하는 Biopolymer YU-122는 분자량이 1.7×10 exp (6)이고, C, H의 구성비가 1:2이며 미량의 N이 검출되었으나, 주 구성성분은 glucose와 galactose의 환원당으로 이루어져있는 전혀 새로운 biopolymer인 것으로 나타났다. To produce biopolymer, Metarrhizium anisopliae (Metschn.) Sorok was cultured in a medium containing glucose 1.0%, sucrose 2.0%, soluble starch 1.0%, yeast extract 0.5%, polypeptone 0.5%, K_2HPO_4 0.1%, MgSO_4·7H_2O 0.02%. The culture broth was centrifuged and the polymer was harvested by adding methanol to the culture supernatant. When three times of methanol was added, the polymer was coagulated and precipitated. Then it was further purifued through successive SK-1B, SA-20P, HP-20 column chromatographies. This polymer was designated as Biopolymer YU-122. C:H ratio of this Biopolymer YU-122 was 1:2 and small amount of N is detected by CHN analyzer. Glucose and galactose are main components of this polymer. Average molecular weight of this biopolymer was 1.7×10 exp (6) by Sepharose 4B gel permeation chromatography. Optimal condition for biopolymer production was investigated. When 5% of mannitol was used as a carbon source, and polypepton as a N source, highest productivity of biopolymer was achieved. C/N ratio as nutrient was also a major factor in polymer production and its optimal ratio was 3.
삼상 순환유동층의 상승관에서 연속 액상의 축방향 혼합특성

직경이 0.102 m이고 높이가 3.5 m인 기체-액체-고체 순환 유동층의 상승관에서 액상의 축방향 혼합특성을 고찰하였다. 기체유속(0.01~0.07 ㎧), 액체유속(0.25~0.31 ㎧) 그리고 고체순환속도(2~8 ㎏/㎡s)를 실험변수로 선정하였으며 이들 변수들이 액상의 축방향 분산계수에 미치는 영향을 고찰하였다. 축방향의 추적자 농도분포로부터 분산모델을 사용하여 액상의 축방향 분산계수를 구하였다. 액상의 축방향 분산계수는 기체유속과 고체순환속도가 증가함에 따라서 증가하였으나 액체의 유속의 증가에 따라서는 약간 감소하였다. 또한, 액상의 축방향 분산계수는 등방성 난류 모델을 적용한 무차원군으로 표현되는 상관식으로 나타낼 수 있었다. Characteristics of liquid dispersion in the axial direction were investigated in the riser of a three-phase circulating fluidized bed whose diameter is 0.102 m and 3.5 m in height. Effects of gas (0.01-0.07 m/sl and liquid (0.25-0.31./s) velocities and the circulation rate of solids (2∼8 kg/m^(2)s) on the dispersion coefficient of the continuous liquid phase in the axial direction were determined. A dispersion model was employed to obtain the axial dispersion coefficient of the liquid phase from the radial concentration profiles of tracer with variations of axial position. The axial dispersion coefficient of the liquid phase increased with increasing gas velocity or solid circulation rate, however, decreased slightly with increasing liquid velocity in the riser bed. The axial dispersion coefficient of the liquid phase was well correlated in terms of dimensionless groups based on the isotropic turbulence theory.

A fundamental experiment was carried out to confirm the behaviour of Bastard halibut Paralichthys olivaceus to audible underwater sound in a breeding ground around the coast of Cheju Island. To find characteristics of breeding water tank and experiment water tank. ambient noise, undcrwater noise and feeding sound of fish were measured and analyzied respectively. The results of measurement are as follows: 1. The maximum sound pressure level of ambient noise in breeding water tank was 81dB. average maximum sound level of underwater noise was 81dB at 120Hz. 2. The maximum sound pressure level of ambient noise in experiment water tank was 76dB and underwater noise was 84.2dB at 130Hz. 3. The maximum sound pressure level of feeding sound was 85dB at 120Hz.

Hepatic angiomyolipoma is a rare benign tumor of the liver composed of blood vessel, smooth muscle cells, fat and myelocomponent. The preoperative diagnosed of the lesion is important because of its therapeutic implications. Radiologic findings are not specific because the composed elements are variable in proportion and distribution. Thus, the findings at computed tomography, ultrasono graphy and magnetic resonance imaging may be only suggestive and its definitive diagnosed requires histologic confirmation. We experienced a case of hepatic angiomyolipoma in patient with chronic hepatitis diagnosed preoperatively by fine-needle aspiraton biopsy under ultrasound guidance. To our knowledge, this is the first case of hepatic angiomyolipoma diagnosed preoperatively in Korea. We report it with review of literatures.

사카린 유도체는 제초제, 항독성, 살균제 등에 광범위하게 연구되어져 오고 있다. 새로운 농략 활성을 가진 물질을 합성하기 위해 먼저 선도물질로 사카린 1을 산화, 염소화 시킨 3-chloro-1,2-benzisothiazol-1,1-dioxide(BID-CI) 2와2cholromethy-1,2-benzisohizol-3-one1,1-dioxide 4를 합성하였다. 선도물질, amines, imidazoles, trones등과 반응시켜 다음과 같은 물질들을 얻었다. ; 3-cyclohexylamino-1,2-benzisothiazol-1,1-dioxide 6, 3-furylamino-1,2-benzisothiazol-1,1-dioxide 8, 3-(3-imidazol)-1,2-benzisothiazol-1,1-dioxide 10, 3-(2-methy-3-imidazol)-1,2-benzisothaizol-1,1-dioxide 12, 2-(3-imidazol)methy-1-1,2-benzisothiazol-3-one-1,1-dioxide 13, and 2-(-3-imidazol)methyl- 1,2-benzisothiazol-1,1-dioxide 14.
A novel prognostic factor for hepatocellular carcinoma: protein disulfide isomerase

<P><B>Background/Aims</B></P><P>Protein disulfide isomerase (PDI) has been implicated in the survival and progression of some cancer cells, by compensating for endoplasmic reticulum stress by upregulating the protein-folding capacity. However, its prognostic role in patients with hepatocellular carcinoma (HCC) has not been investigated.</P><P><B>Methods</B></P><P>We collected HCC tissues from 83 HCC patients who underwent surgical resection for an immunohistochemical study of PDI. Overall survival (OS) was measured from the date of surgical resection until the date of death from any cause. Radiological progression was evaluated using the modified Response Evaluation Criteria in Solid Tumors in an independent radiological assessment.</P><P><B>Results</B></P><P>PDI expression was found to be increased in human HCC compared to adjacent nontumor tissues. Increased immunopositivity for PDI was associated with a high Edmondson-Steiner grade (<I>p</I> = 0.028). Univariate analysis of patients who had undergone surgical resection for HCC showed that tumor PDI upregulation is a significant risk factor for poor OS (<I>p</I> = 0.016; hazard ratio [HR], 1.980) and time to progression (TTP; <I>p</I> = 0.007; HR, 1.971). Multivariate analyses revealed that high PDI expression was an independent predictor of a shorter TTP (<I>p</I> = 0.015; HR, 1.865) and poor OS (<I>p</I> = 0.012; HR, 2.069).</P><P><B>Conclusions</B></P><P>Upregulated PDI expression is associated with aggressive clinicopathological features of HCC; thus, PDI might serve as an independent prognostic factor and a potential therapeutic target for HCC patients.</P>

Background: ent-Sauchinone is a polyphenolic compound found in plants belonging to the lignan family. ent-Sauchinone has been shown to modulate the expression of inflammatory factors through the nuclear factor-kappa B (NF-κB) signaling pathway. It is well known that neuroinflammation is associated with amyloidogenesis. Thus, in the present study, we investigated whether ent-Sauchinone could have anti-amyloidogenic effects through the inhibition of NF-κB pathways via its anti-inflammatory property. Methods: To investigate the potential effect of ent-Sauchinone on anti-neuroinflammation and anti-amyloidogenesis in in vitro studies, we used microglial BV-2 cells and cultured astrocytes treated with ent-Sauchinone (1, 5, and 10 μM) for 24 hours. For the detection of anti-neuro-inflammatory responses, reative oxygen species (ROS) and Nitric oxide (NO) generation and inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression were measured with assay kits and western blotting. β-secretase and β-secretase activities and β-amyloid levels were determined for measuring the anti-amyloidogenic effects of ent-Sauchinone by enzyme assay kits. NF-κB and STAT3 signals were detected with electromobility shift assay (EMSA) to study the related signaling pathways. The binding of ent-Sauchinone to STAT3 was evaluated by a pull-down assay and by a docking model using Autodock VINA software (Hoover`s Inc., Texas, United states).
