<P>Catalytic asymmetric synthesis of N-heterocyclic glycosides free of protecting and directing groups is reported. The key reaction is highlighted by the atom-efficient and regioselective addition of unprotected pyrimidines to highly functionalized alkoxyallene. Numerous acyclic and cyclic N-heterocyclic glycosides are accessed with minimal formation of organic byproducts. The synthetic utility of the reaction is demonstrated by the first catalytic asymmetric synthesis of anticancer pharmaceutical (-)-Tegafur and stereoselective synthesis of an oxepane nucleoside derivative.</P>

<P><B>Purpose</B></P><P>Overexpression of cyclooxygenase 2 (COX-2) is thought to promote survival of transformed cells. Transforming growth factor β (TGF-β) exerts anti-proliferative effects on a broad range of epithelial cells. In the current study, we investigated whether TGF-β can regulate COX-2 expression in A549 human lung adenocarcinoma cells, which are TGF-β-responsive and overexpress COX-2.</P><P><B>Materials and Methods</B></P><P>Western blotting, Northern blotting, and mRNA stability assays were performed to demonstrate that COX-2 protein and mRNA expression were suppressed by TGF-β. We also evaluated the effects of tristetraprolin (TTP) on COX-2 mRNA using RNA interference.</P><P><B>Results</B></P><P>We demonstrated that COX-2 mRNA and protein expression were both significantly suppressed by TGF-β. An actinomycin D chase experiment demonstrated that COX-2 mRNA was more rapidly degraded in the presence of TGF-β, suggesting that TGF-β–induced inhibition of COX-2 expression is achieved via decreased mRNA stability. We also found that TGF-β rapidly and transiently induced the expression of TTP, a well-known mRNA destabilizing factor, before suppression of COX-2 mRNA expression was observed. Using RNA interference, we confirmed that increased TTP levels play a pivotal role in the destabilization of COX-2 mRNA by TGF-β. Furthermore, we showed that Smad3 is essential to TTP-dependent down-regulation of COX-2 expression in response to TGF-β.</P><P><B>Conclusion</B></P><P>The results of this study show that TGF-β down-regulated COX-2 expression via mRNA destabilization mediated by Smad3/TTP in A549 cells.</P>
한국어 교사 대상 온라인 재교육 프로그램의 내용 분석과 개선 방향 : <한국 문화 활용 교수법>을 중심으로

본 연구는 한국어 교사의 문화 교수 능력을 신장시키기 위해 제공되는 온라인 재교육 프로그램의 내용을 분석하는 데 목적이 있다. 분석 결과를 토대로 향후 문화 교육 영역에서 한국어 교사를 위해 어떤 내용이 더 보완되고, 교사의 전문적인 지식 가운데 어떤 요소가 더 고려되어야 하는지를 탐색하여 방향성을 제언하였다. 분석 대상은 현재 국립국어원에서 제공하고 있는 한국 문화 활용 교수법 교과목을 중심으로 문화 영역 PCK에 근거하여 교육 내용을 분석하였다. 분석 결과 형식적인 측면에서 이론적 설명과 사진이 함께 제시되면서 내용 정리가 잘 되어 있었으며, 교과 내용지식과 교수학적 지식에 대한 부분이 교사가 이해하기 쉽도록 재교육 환경을 제공하고 있다. 다만, 현재 체계적이지 않은 지식 체계의 전달로 인해 실제적인 교수 활동을 통해 얻을 수 있는 전문성이 제대로 갖춰질지 의문스러운 상황이었다. 특히, 상황적 지식과 학습자의 문화 지식, 학습 평가 지식 등에 대해서는 온라인 재교육에서도 여전히 교사의 전문적인 지식을 제공하지 못한다는 문제를 안고 있었다. 이 연구는 현재 한국어 교사를 대상으로 온라인을 통해 제공되는 재교육 프로그램의 내용을 보다 더 구체적으로 내용 분석을 시도하였으며 향후 제공되는 프로그램에 대한 시사점을 제공했다는 점에서 의의가 있다. This study aims to analyze the content of online re-education programs offered to enhance Korean language teachers’ ability to teach culture. Based on the analysis results, the direction was suggested by exploring which content should be further supplemented for Korean language teachers in the future cultural and educational areas and which of the teachers’ professional knowledge should be considered more. The subjects of the analysis were based on the PCK in the cultural field, focusing on the teaching methods for utilizing Korean culture currently provided by the National Institute of Korean Language. As a result of the analysis, the theoretical explanations and photographs were presented together in terms of formality, the contents were well organized, and the parts of the curriculum content knowledge and teaching knowledge provided a re-education environment for teachers to understand easily. However, it was questionable whether the expertise gained through practical teaching activities would be properly provided due to the delivery of an unsystematic knowledge system. In particular, concerning the contextual knowledge, the learner’s cultural knowledge, and the knowledge of learning evaluation, there was a problem that online retraining still did not provide the teacher’s professional knowledge. The study is meaningful in that it attempted to analyze the contents of the current online re-training program for Korean language teachers more specifically and provided implications for future programs.
자외선 흡광도를 이용한 유전자재조합의약품 함량 결정과정 분석

This study summarizes the theoretical background of the protein concentration determination process using UV absorbance and proposes the standard calculation formular. We analyzed the protein concentration determination process of recombinant DNA-derived biologics that is inspected by the Ministry of Food and Drug Safety for the last three years (2017-2019) and we found some errors in calculation formular. In addition, we analyzed parameters and acceptance criteria of system suitability test (SST), and we proposed the direction for setting the SST. If the results of this study are widely understood and utilized by product manufacturers and research developers, the calculation of protein concentration using UV absorbance for each product would be simplified and standardized, and contribute to efficient communication between product manufacturers and regulatory authority.
