        3분과 : 가선집재작업에서의 작업 생산성 및 비용 분석

        본 연구에서는 임목수확작업에 사용되는 집재기계의 작업효율성을 분석하여 경사가 급한(20° 이상) 작업조건에 적합한 가선계 집재기계를 선정하는데 기초 정보를 제공하고자, 타워야더(RME-300T), 트랙터부착형 집재기(일명: 춘천집재기), 트랙터윈치(FARMI) 등에 의한 집재작업에서의 작업생산성 및 비용을 비교 분석하였다. 평균 흉고직경이 20 cm, 집재거리 60 m, 가로집재거리 10 m, 그리고 기계이용률이 70%인 동일한 작업 조건에서 작업이 이루어질 경우, 타워야더의 1일 집재작업 생산성은 33.04 m3/일, 트랙터부착형 집재기는 38.47 m3/일, 그리고 트랙터윈치는 14.17 m3/일으로 분석되었으며, 이에 따른 집재작업비용은 각각 25,105원/m3, 20,520원/m3, 37,835원/m3으로 나타났다. 동일한 작업조건에서 집재거리에 따른 각 집재기계별 작업효율성을 비교하였을 경우, 집재거리가 40 m 이내로 짧을 경우에는 트랙터윈치에 의한 집재작업이 효율적이며, 40~140 m의 집재거리에서는 춘천집재기가, 140 m 이상의 장거리 집재작업에서는 타워야더가 가장 효율적인 것으로 나타났다. This study was conducted to provide field-based harvesting study information which can be used to select an optimal cable system for certain work conditions on steep grounds (>20° ground slope) in Korea. Toaccomplish this study objectives, we evaluated three cable yarding systems (RME-300T tower-yarder, Chuncheontower-yarder, FARMI tractor winch) working in typical work conditions for their yarding productivity and operational efficiency. Those yarders are commonly used for removing logs or trees on steep grounds in Korea. Under the same work conditions (average DBH of tree to be cut, 20 cm; yarding distance, 60 m; lateral yardingdistance, 10 m; and machine utilization rate, 70%), the average productivities were 33.04 m3/day, 38.47 m3/day, and 14.17 m3/day for RME-300T, Chuncheon tower-yarder, and FARMI, respectively. Our standardized cost comparison study also showed that the yarding cost was highest at 37,835 won/m3 with FARMI, followed byRME-300T at 25,105 won/m3 for the same work conditions. We found the lowest yarding cost with the Chuncheon tower-yarder at 20,520 won/m3 which was resulted primarily from high yarding productivity at theyarding distance (60 m). Our analysis suggested that a small machine such as FARMI could be a low-costyarding machine option for a cable yarding job with a short yarding distance (40 m or less). The Chuncheontower-yarder is well suited for a mid-range yarding distance job in Korea, ranged between 40 to 140 m. Ifyarding distance were longer than 140 m, the RME-300T tower yarder appears to be most cost-effective.

        Long-term postresection prognosis of primary neuroendocrine tumors of the liver

        Purpose: Primary hepatic neuroendocrine tumor (PHNET) is a very rare neoplasm, requiring strict exclusion of metastasis from possible extrahepatic primary sites for its diagnosis. Methods: We reviewed our clinical experience of 13 patients with primary hepatic NET who underwent liver resection from January 1997 to December 2015. Results: The mean age of the 13 patients (8 males and 5 females) was 51.1 ± 12.8 years; the most common clinical manifestation was vague, nonspecific abdominal pain (n = 9). Of them, 11 patients underwent preoperative liver biopsy, 7 of which correctly diagnosed as neuroendocrine tumor (NET). Ten patients underwent R0 resection, and 3 underwent R1 resection. Diagnosis of PHNET was confirmed both immunohistochemically and by absence of extrahepatic primary sites. All tumors were single lesions, with a mean size of 9.6 ± 7.6 cm and a median size of 4.3 cm; all showed positive staining for synaptophysin and chromogranin. During a mean follow-up period of 95.1 ± 86.6 months, 7 patients died from tumor recurrence, whereas the other 6 remain alive to date, making the 5-year tumor recurrence rate 56.0% and the 5-year patient survival rate 61.5%. When confined to R0 resection, 5-year recurrence and survival rates were 42.9% and 70.0%, respectively. Univariate analysis showed that Ki-67 proliferative index was the only risk factor for tumor recurrence. Conclusion: PHNET is a very rare tumor with no specific clinical features, and its final diagnosis depends primarily on pathology, immunohistochemistry, and exclusion of metastasis from other sites. Aggressive surgical treatment is highly recommended for PHNET because of acceptably favorable postresection outcomes

      • Benzyl Isothiocyanate Inhibits Prostate Cancer Development in the Transgenic Adenocarcinoma Mouse Prostate (TRAMP) Model, Which Is Associated with the Induction of Cell Cycle G1 Arrest

        <P>Benzyl isothiocyanate (BITC) is a hydrolysis product of glucotropaeolin, a compound found in cruciferous vegetables, and has been shown to have anti-tumor properties. In the present study, we investigated whether BITC inhibits the development of prostate cancer in the transgenic adenocarcinoma mouse prostate (TRAMP) mice. Five-week old, male TRAMP mice and their nontransgenic littermates were gavage-fed with 0, 5, or 10 mg/kg of BITC every day for 19 weeks. The weight of the genitourinary tract increased markedly in TRAMP mice and this increase was suppressed significantly by BITC feeding. H and E staining of the dorsolateral lobes of the prostate demonstrated that well-differentiated carcinoma (WDC) was a predominant feature in the TRAMP mice. The number of lobes with WDC was reduced by BITC feeding while that of lobes with prostatic intraepithelial neoplasia was increased. BITC feeding reduced the number of cells expressing Ki67 (a proliferation marker), cyclin A, cyclin D1, and cyclin-dependent kinase (CDK)2 in the prostatic tissue. <I>In vitro</I> cell culture results revealed that BITC decreased DNA synthesis, as well as CDK2 and CDK4 activity in TRAMP-C2 mouse prostate cancer cells. These results indicate that inhibition of cell cycle progression contributes to the inhibition of prostate cancer development in TRAMP mice treated with BITC.</P>

        Byr4p , Possible Regulator of Mitosis and Cytokinesis in Fission Yeast , Localizes to the Spindle Pole Body by its C-Terminal Domains

        Cytokinesis and septation should be coordinated to nuclear division in the cell division cycle for precise transmission of the genome into daughter cells. byr4, an essential gene in fission yeast Schizosaccharomyces pombe, regulates the timing of cytokinesis and septation in a dosage-dependent manner. We examined the intracellular localization of the Byr4 protein by expressing byr4 as a fusion of green fluorescence protein (GFP). The Byr4 protein localizes as a single dot on the nuclear periphery of interphase cells, duplicates before mitosis, and the duplicated dots segregate with the nuclei in anaphase. The behavior of Byr4p throughout the cell cycle strongly suggests that Byr4p is localized to the spindle pole body (SPB), a microtubule organizing center (MTOC) in yeast. The presence of the Byr4 protein in the SPB is consistent with its function to coordinate mitosis and cytokinesis. We also mapped the domains of Byr4p for its proper localization to SPB by expressing various byr4 deletion mutants as GFP fusions. Analyses of the diverse byr4 deletion mutants suggest that the indirect repeats and the regions homologous to the open reading frame (ORF) YJR053W of S. cerevisiae in its C-terminus are essential for its localization to the SPB.

        Integrin-Linked Kinase Is Required in Hypoxic Mesenchymal Stem Cells for Strengthening Cell Adhesion to Ischemic Myocardium

        <P>Mesenchymal stem cells (MSCs) therapy has limitations due to the poor viability of MSCs after cell transplantation. Integrin-mediated adhesion is a prerequisite for cell survival. As a novel anti-death strategy to improve cell survival in the infarcted heart, MSCs were genetically modified to overexpress integrin-linked kinase (ILK). The survival rate of ILK-transfected MSCs (ILK-MSCs) was augmented by about 1.5-fold and the phosphorylation of ERK1/2 and Akt in ILK-MSCs were increased by about three and twofold, respectively. ILK-MSCs demonstrated an increase of twofold in the ratio of Bcl-2/Bax and inhibited caspase-3 activation, compared with hypoxic MSCs. The adhesion rate of ILK-MSCs also had a 32.2% increase on the cardiac fibroblast-derived three-dimensional matrix and ILK-MSCs showed higher retention by about fourfold compared to unmodified MSCs. Six animals per group were used for the in vivo experiments analyzed at 1 week after occlusion of the left coronary artery. ILK-MSC transplanted rats had a 12.0% +/- 3.1% smaller infarct size than MSC-treated rats after ligation of left anterior descending coronary artery. Transplantation of ILK-MSCs not only led to a 16.0% +/- 0.4% decrease in the fibrotic heart area, but also significantly reduced the apoptotic positive index by two-thirds when compared with ligation only. The mean microvessel count per field in the infarcted myocardium of ILK-MSCs group was increased relative to the sham group and MSCs group. In conclusion, the ILK gene transduction of MSCs further assisted cell survival and adhesion, and improved myocardial damage when compared with MSC only after transplantation.</P>

        Forage yield, nutritional value, soil chemical composition, and soil microbial abundance under maize–legume intercropping systems in a paddy field

        This study aimed to evaluate the forage production, nutritional value, soil chemical properties, and rhizosphere microorganism composition of maize–legume intercropping systems in fallow paddy felds. Before seeding, the felds were sprayed with 1000 kg ha−1 of nitrogen composite fertilizer (NPK: 21-17-17). From 2020 to 2021, maize was intercropped with seven diferent legume cultivars at the National Institute of Animal Science, Cheonan, Republic of Korea. Of the seven legumes, the intercropped lablab (Lablab purpureus [L.] Sweet) and cowpea (Vigna unguiculata L. Walp) showed the greatest plant heights because the maize plants supported their climbing habit. The forage productivity of the intercropped soybean cultivars, Chookdu 1 and 2, lablab, and cowpea, was relatively high compared with those of the other legumes. Chookdu 1 and 2 showed higher forage productivity in monoculture than when intercropped. When intercropped with maize, the Daewon and Daepung soybean cultivars showed the lowest neutral detergent fber and acid detergent fber contents and the highest crude protein in 2020. However, the forage productivity of Daewon and Daepung was low, indicating unsuitability for intercropping with maize in paddy felds. The microorganism composition in the root rhizosphere showed a higher proportion of Proteobacteria under legume than maize cultivation. Proteobacteria were substantially more abundant in soils intercropped with lablab than in the lablab monoculture. Maize and cowpea appeared to be the best intercropping combination considering forage productivity and feed value. However, the Chookdu 1 and 2 cultivars and lablab also showed potential for intercropping with maize in paddy felds.

        Piceatannol suppresses the metastatic potential of MCF10A human breast epithelial cells harboring mutated H-ras by inhibiting MMP-2 expression

        <P>Metastasis is one of the most threatening features of the oncogenic process and the main cause of cancer-related mortality. Several studies have demonstrated that matrix metalloproteinases (MMPs) are critical for tumor invasion and metastasis. Resveratrol (3,5,4′-trihydroxystilbene), a phenolic compound of red wine, has been reported to be a natural chemopreventive agent. However, the cancer preventive effects of piceatannol (3,5,3′,4′-tetrahydroxystilbene), a metabolite of resveratrol and the underlying molecular mechanisms have not yet been fully elucidated. In this study, we report that piceatannol inhibits H-ras-induced MMP-2 activity and the invasive phenotype of MCF10A human breast epithelial cells harboring mutated H-ras (H-ras MCF10A cells) more effectively than resveratrol. Piceatannol attenuated the H-ras-induced phosphorylation of Akt in a time- and dose-dependent manner, whereas resveratrol, at the same concentrations, did not exert an inhibitory effect. <I>In vitro</I> kinase assays demonstrated that piceatannol significantly inhibited phosphatidylinositol 3-kinase (PI3K) activity and suppressed phosphatidylinositol (3,4,5)-trisphosphate (PIP<SUB>3</SUB>) expression in the H-ras MCF10A cells. <I>Ex vivo</I> pull-down assays revealed that piceatannol directly bound to PI3K, inhibiting PI3K activity. Data from molecular docking suggested that piceatannol is a more tight-binding inhibitor than resveratrol due to the additional hydrogen bond between the hydroxyl group and the backbone amide group of Val882 in the ATP-binding pocket of PI3K.</P>

        Purification and Characterization of a Serine Proteinase from Acanthamoeba culbertsoni

        A serine proteinase was purified from Acanthamoeba culbertsoni by 41∼80 ammonium sulfate fractionation, ion exchange chromatography, affinity chromatography and gel filtration chromatography. The molecular weight of the purified enzyme was estimated to be 108.0 kDa by gel filtration chromatography and 54.0 kDa by SDS-PAGE_ Therefore, the purified enzyme seemed to be a dimer. Isoelectric point was 4.5. The enzyme activity was highly inhibited by the serine proteinase inhibitors diisopropyl fluorophosphate (DFP) and phenylmethyl sulfonylfluoride (PMSF). It had a narrow pH optimum of 6.5∼7.5 with a maximum at pH 7.0. These data suggested that the purified enzyme was a neutral serine proteinase. Optimal temperature was 37℃. It was stable for at least 16 h at 4℃ and 37℃, but it was rapidly inactivated at 65℃. The activity of the purified enzyme was not influenced significantly by Mg^(2+). Mn^(2+). Zn^(2+) or Ca^(2+). However, the enzyme activity was highly inhibited by Hg^(2+). The enzyme degraded type I collagen and fibronectin, but not BSA, hemoglobin, lysozyme, immunoglobulin A or immunoglobulin G.
