        Analysis of Population Structure and Genetic Diversity in Balloon Flower (Platycodon grandiflorum (Jacq.) A. DC.) Germplasm Using Simple Sequence Repeat (SSR) Markers

        Balloon flower (Platycodon grandiflorum A. DC) is widely distributed in South Korea and there are some local landraces that are cultivated as a vegetable crop or medicinal plant. Making use of the gene resources of wild-type and landraces is a way to increase the genetic diversity of the cultivars. However, few tools or information are available on an efficient identification system for maintaining and management of these landraces. To improve the genetic resources for balloon flower, 22 simple sequence repeat (SSR) markers, also known as microsatellite markers, were evaluated in a collection of 42 balloon flower landraces, 34 of which were from Korea and eight from China. All microsatellite markers produced the 107 alleles ranging from 2 to 10 with a mean of 4.864 alleles per each locus ($N_A$). The values of observed heterozygosity ($H_O$) and expected heterozygosity ($H_E$) ranged from 0.00 to 0.667 (mean of 0.285) and from 0.024 to 0.741 (mean of 0.416), respectively. An average value of polymorphic information contents (PIC) were 0.382 with a range of 0.023 to 0.703. Results of population structure and phylogenetic and principal coordinate analysis (PCoA) indicated that P. grandiflorum germplasm formed two largely distinct clusters according to their origins and the genetic differentiation. There was a high level of genotypic diversity at broad geographic regions between Korea and China, but the low genetic differentiation was found within the collections from Korea. The results of the genetic diversity will be useful for the selection of the parents for developing balloon flower breeding and the multi-locus SSR markers developed herein will be a valuable resource for germplasm assessments, evaluation of genetic diversity, and population genetic studies of balloon flower.

      • Analysis of customers demands for effective management and distribution of plant genetic resources

        The need to develop work on conservation of crop genetic diversity is emphasized in change of agricultural environment, such as climate change, turn of dietary habits, use of cereal to bioenergy production. To secure provide the germplasm and to derive improve quality, customers demands was surveyed in 2011 and the results are as follows. In the questionnaire on genetic resources necessary in the future, Korean landrace was the highest as 23.8%, followed by genetic resources of international agricultural research institutions as 21.4% and genetic resources of foreign countries of 21.4%. Followed by 42.8% for genetic resources from foreign countries, response ratio were 10.7%, 9.5% and 9.5% for wild or wild-relative species, advanced cultivar or breeders’ line from foreign countries, and advanced cultivar or breeders’ line from Korea, respectively. In addition, consumers desired different genetic resources according to their institutes. Satisfaction on seed purity, germination rate and expected characteristic expression with the genetic resources was higher than normal by 87.5%, 88.9%, 84.2% of respondents, respectively. In order to improve the quality of conserved resources, it is necessary to select the resources having useful characteristics from the time of introduction, and to facilitate the evaluation of the preserved accessions in the near future. Efforts for expansion and opening of information on genetic resources are required.

        The world-wide capacity of genebanks for ex-situ conservation of crop genetic resources has increased greatly since the 1970s, improving the access of crop breeders to landraces, and wild relatives (Wright, 1997). In south Korea, systematic seed germplasm management was begun at the Rural Development Administration (RDA) in 1985 and consequently genebank system was established in 1988. The RDA genebank, National Agrobiodiversity Center (NAC) of south Korea, preserve 173,217 accessions of plant seed germplasm and 10.7% (18,509 accessions) of whole preserved germplasm is occupied by horticultral PGR by 2011. Horticultural PGR preserved in NAC is composed with 67 genera of crops including pepper (3,817 accessions), tomato (2,040 accessions), cabbages (1,690 accessions) and etc. According to horticultural plant sub-group, fruit-vegetable is the biggest group with 60% of accession share and leafy vegetable (19%), root vegetable (10%), bulb vegetable (8%), and ornamental (3%) follow subsequently. In aspect of PGR status, landrace is dominant part of conserved accession and then developed cultivar, wild relatives, cultivated material, wild, and weedy type follow subsequently. Recently characterization and evaluation of horticultural PGR have been activated for 3 years in NAC, which are useful for breeders and users focused on disease-resistance or active ingredient. It will promote utilization of horticultural PGR and development of horticultural crop breeding program for both human being and sustainable agriculture in near future.

        10세 미만아 마취의 임상적 고찰 : 과거 10년간 Past 10 years

        The authors analyzed statistically 1309 cases of pediatric anesthesia (under 10 years old) from January, 1967 to December, 1976 at the central operating theatre, Busan National University Hospital and the result were as follows. 1. 264 cases (20.2%) were under 1 year old, the most in group, and 55 cases (4.2%) were 1 years old as the least. 2. Sexual distribution was 853 males (62.5%) and 456 females (34.8%). 3. Anesthesias for the operations of general surgery were the most as 444 cases (33.9%). 4. Elective surgeries were 811 cases (62.0%) and the remainder were emergency operations (498 cases, 38.0%). 5. The techniques of anesthesia were 577 cases (44.1%) of infantile circle system with endotracheal intubation and 489 cases (37.4%) of non-rebreathing system. 6. The main anesthetics were 694 cases (53.0%) of halothane and 571 cases (43.6%) of ether. 7. Intra and postoperative death cases were 58 cases (4.4%) and of these, 27 cases (2.1%) were under-1 year old.

        심 자창환자 마취의 임상적 고찰

        The authors studied 11 cases of anesthesia for stab wound in the heart which were performed in the Busan national university hospital from 1973 to 1976. The results were as follows: 1. Male vs. female ratio was 9:2 and the second to third decade of age had the highest number in the age groups. 2. The causes of wounds were 9 cases by knife, one by traffic accident and one by explosion. 3. Fatal cases were 2 in 11 cases, one during operation due to cardiac arrest and the otehr, on the seventh postoperative day in the ward, due to asphyxia from bleeding of the tracheostomized wound.

        <P>Although polymorphisms in glutathione S-transferase (GST) have been associated with the risk of bladder cancer (BC), few reports provide information about the development of BC. The aim of the present study was to investigate the effect of homozygous glutathione S-transferase-μ (GSTM1) and glutathione S-transferase-&phis; (GSTT1) deletions as prognostic markers in non-muscle-invasive bladder cancer (NMIBC). A total of 241 patients with primary NMIBC were enrolled in this study. GSTM1 and GSTT1 polymorphisms were analyzed by multiplex polymerase chain reaction (PCR) using blood genomic DNA. The results were compared with clinicopathological parameters. The prognostic significance of the GSTs was evaluated by Kaplan-Meier and multivariate Cox regression model. A statistically significant association between genotype and histopathological parameter was not observed. The patients with the GSTT1-positive genotype had significantly reduced recurrence- and progression-free survival than those with the GSTT1-null genotype (log-rank test, <I>p</I> < 0.05, respectively). Recurrenceand progressionfree survival were not related to the GSTM1 genotypes. In multivariate regression analysis, the GSTT1positive genotype was the independent predictor for recurrence [hazard ratio (HR), 1.631; <I>p</I> = 0.043] and progression (HR, 3.418; <I>p</I> = 0.006). These results suggested that the GSTT1 genotype could be a useful prognostic marker for recurrence and progression in NMIBC. </P>

        현재 일반적으로 사용되는 표면 결함 탐상 관련기술은 일반적으로 초음파 탐상과 와전류 탐상으로 행해지고 있다. 이 두 가지 방법은 방법의 원리적인 한계 및 장치 구성상의 한계상 검출시간 및 검출율에 한계가 있으며 비용면에서도 고가의 장비라는 면에 단점을 가지고 있다. [4~5] 이에 반해 교류누설 자속법은 아직 그 원리적 이해가 부족한 상황이지만 특성상 표면의 미세결함에 대해 매우 높은 정밀도를 가지며 시스템 제작이 용이하다는 장점이 있다. [4] 또한 탐촉자를 여러개 배열 하므로써 동시에 넓은 영역의 검사가 가능해 탐상시간이 기종의 방법에 비해 매우 단축된다. 또한, 그 탐상 결과를 정량화 시킬 수 있다는 잇점이 있다 본 논문에서는 이와 같은 장점을 갖는 AC magnetic leakage flux method을 채용한 system을 개발하여 Reference를 X축 Y축에 대하여, Search coil sensor를 이동시켰다 그리고, 그 이동경로에 대한 Reference의 결함Data를 직접적인 육안으로, 즉 Real-Time으로 Scan 하게금 설계하였다. 그리고, 저장의 용이성을 위해 data를 불러들일 수 있도록 프로그래밍 하였으며, 동시에 Data만을 가지고 결함을 찾지 못하는 일반인들을 위하여 Math-lab을 이용하여 Real-time 2차원 및 3차원 영상을 보여주게금 하였다. Generally, The present related technology of crack surface scanning is being done by sona-scanning testing and eddy-current scanning within the domestic. These two method has some disadvantage like that cost high price to accomplish, limitation of detection time, detection rate by the limitation of principal and device organization. On the contrary, ac magnetic flux scanning is short of principal understanding. but has advantage that is easy to make system, measure highly precise in the respect of minut-est crack on surface. Further, as arrangement of many poll, it can examine wide area at a time. for that reason scanning time is very short for established method. And it has advantage that change result of scanning to a fixed quantity. In this paper, we design system that use these advantage of AC magnetic leakage flux method. Designed system can scan crack-data that move search coil sensor around x and y axis and check movement path about reference in real time. And We program it can open data with excel for ease to save, can show Real-time 2 dimension and 3 dimension image by using Math-lab for novice who can not find a fault in data alone.
