        대두 칼모듈린 단백질, GmCaM-4를 발현하는 형질전환 감자의 무름병 저항성 확인

        Calmodulin (CaM) mediates cellular Ca²⁺ signals in the defense responses of plants. We previously reported that GmCaM-4 and 5 are involved in salicylic acidindependent activation of disease resistance responses in soybean (Glycine max). Here, we generated a GmCaM-4 cDNA construct under the control of the cauliflower mosaic virus (CaMV) 35S promoter and transformed this construct into potato (Solanum tuberosum L.). The constitutive over-expression of GmCaM-4 in potato induced high-level expression of pathogenesis-related (PR) genes, such as PR-2, PR-3, PR-5, phenylalanine ammonia-lyase (PAL), and proteinase inhibitorII (pinII). In addition, the transgenic potato plants exhibited enhanced resistance against a bacterial pathogen, Erwinia carotovora ssp. Carotovora (ECC), that causes soft rot disease and showed spontaneous lesion phenotypes on their leaves. These results strongly suggest that a CaM protein in soybean, GmCaM-4, plays an important role in the response of potato plants to pathogen defense signaling.

        식용 고구마 신품종 ‘다호미’

        ‘Dahomi’, a new sweetpotato variety, was developed for table use by the sweetpotato breeding team of Bioenergy Crop Research Institute, National Institute of Crop Science (NICS), RDA in 2012. This variety was derived from the cross between ‘Muan4’ and ‘Jinhongmi’ in 2005. The seedling and line selections were performed from 2007 to 2008, preliminary and advanced yield trials were carried out from 2009 to 2010, and the regional yield trials was conducted at five locations from 2011 to 2012 and it was named as ‘Dahomi’. This variety has three or five lobes leaf, green leaf, stem, and petiole. Storage root of ‘Dahomi’ was elliptic, red skin, light orange flesh. This variety is resistant to fusarium wilt, but susceptible to root-knot nematode. Texture of steamed storage root was intermediate, and degree Brix of steamed storage root was 24.8 °Brix. The total sugar content of raw and steamed storage root was 9.7, 32.0/100 dry weight (g), respectively, which was higher than that of ‘Yulmi. Storage root yield of ‘Dahomi’ was 26.3 MT/ha under the early seasonal cultivation, which was 52% higher than that of ‘Yulmi’. Storage root yield of ‘Dahomi’ was 21.9 MT/ha under the optimum and late seasonal cultivation, which was 35% higher than that of ‘Yulmi’. The number of storage roots over 50 g per plant was 2.8 and the average weight of storage root was 138 g under the optimum and late seasonal cultivation.

      • Role of S100A9 in the development of neutrophilic inflammation in asthmatics and in a murine model

        <P><B>Abstract</B></P> <P>S100A9 is an endogenous danger signal that promotes and exacerbates the neutrophilic inflammatory response. To investigate the role of S100A9 in neutrophilic asthma, S100A9 levels were measured in sputum from 101 steroid-naïve asthmatics using an ELISA kit and the levels were significantly correlated with percentages of neutrophils in sputum. Intranasal administration of recombinant S100A9 markedly increased neutrophil numbers at 8h and 24h later with concomitant elevation of IL-1β, IL-17, and IFN-γ levels. Treatment with an anti-S100A9 antibody restored the increased numbers of neutrophils and the increased airway resistance in OVA/CFA mice toward the levels of sham-treated mice. Concomitantly, the S100A9 and neutrophil elastase double positive cells were markedly reduced with attenuation of IL-1β, IL-17, and IFN-γ levels by the treatment with the anti-S100A9 antibody. Our data support a role of S100A9 to initiate and amplify the neutrophilic inflammation in asthma, possibly via inducing IL-1β, IL-17 and IFN-γ.</P> <P><B>Highlight</B></P> <P> <UL> <LI> S100A9 is an endogenous danger signal that promotes and exacerbates the neutrophilic inflammatory response. </LI> <LI> Our data support a role of S100A9 to initiate and amplify the neutrophilic inflammation via inducing IL-1β, IL-17 and IFN-γ. </LI> <LI> Modulation of S100A9 in the airway may be a therapeutic strategy to control neutrophilic inflammation in asthma. </LI> </UL> </P>

        유전 및 육종 : 국내산 더러브렛 경주마의 씨수말 평가 기준으로 이용되는 연간수득상금 분석

        본 연구는 연간수득상금형질을 이용한 국내 씨수말 평가시스템을 분석하여 주파기록과의 관계를 구명하고자 수행되었다. 씨수말 당 평균 자마수와 출주수는 각각 34두와 221회였다. 반면에 평균 2세 자마수와 출주수는 각각 9두와 25회였으며, 2세 자마들이 벌어들인 상금은 연간 수득상금의 8.3%였다. 연간수득상금에 대한 자마수와 출주수간의 단순상관계수는 각각 0.922와 0.934, 그리고 자마수와 출주수간의 단순상관계수는 0.985로 매우 높게 나타났다. 씨수말의 검정경력 첫해의 평균 자마수는 6두, 출주수는 17회로 매우 낮았으며, 검정경력에 따른 자마수와 연간수득상금은 매우 밀접한 관계를 나타내었다. 검정경력 4년차에서 40두 이상의 자마수를 확보하는 것으로 나타났으며, 자마수가 증가할수록 평균수득상금지수는 일괄적으로 증가하였다. 자마수가 30두 이하의 씨수말들의 평균수득상금지수는 1.00 보다 낮았으며, 자마수가 10두 이하인 경우에는 0.06에서 0.13의 범위를 나타내어 자마수는 씨수말의 순위 결정에 매우 큰 영향을 미치고 있었다. 주파기록에 대한 육종가와 출주당 수득상금의 상관계수는 다른 형질들에 비하여 가장 높은 -0.524에서 -0.633의 범위를 나타내었다. This study was conducted to analyze demerits of the sire evaluation system using annual earnings and to examine relationship between annual earnings and finish time in home-produced thoroughbred racehorses. The average number of progenies and number of starts per sire were 34 heads and 221 times, respectively. On the other hand, the number of progenies with the average age of 2 years and the number of starts were 9 heads and 25 times, respectively. The earnings of the horses with the age of 2 years accounted for 8.3% of annual earnings. The simple correlation coefficient between the number of progenies and the number of starts in annual earnings were 0.922 and 0.934, respectively. The correlation coefficient between the number of progenies and the number of starts was very high (0.985). The number of progenies and starts of sires for the first year of test career were very low (6 heads and 17 times), and there was very close relationship between number of progenies and annual earnings by the year of test career. The number of progenies was over 40 heads during the first 4 years of test career, and as the number of progenies increased the average earning index increased. The average earning index of sires with less than 30 progenies was lower than 1.00. When the number of progenies was less than 10, the average earning index was in the range of 0.06~0.13, indicating that the number of progenies affects much for determining the ranking of sires. The correlation coefficient between breeding value for finish time and annual earnings per start was very high(-0.524~-0.633) compared with other traits.

        2차유동이 평판후류의 난류열전달에 미치는 영향

        The effect of secondary flow on the heat transfer of a turbulent wake generated by a flat plate was experimentally investigated. The secondary flow was induced in a curved duct in which the flat plate wake generator was installed. All three components of turbulent heat flux were measured in the plane containing the mean radius of curvature of the curved duct. The results showed that mean temperature profiles deviate from the similarity of the straight wake because of the cold fluid transported from the free-stream. The half-width of the mean temperature profile increased rapidly by upwash motion of the secondary flow. The changes to turbulence structure caused by the secondary flow show more pronounced effect on heat transport than on momentum transport. This is because the response to the variation of flow conditions is delayed in temperature field. Negative production of the turbulent heat flux is observed in the inner wake region. From the conditional averaging, it has been found that the negative production of the turbulent heat flux is generated due to a mixing process between the hot and low momentum eddies occupied in the inner wake region and the cold and high momentum eddies in the potential region.

      • Impact of Visceral Fat on the Postprandial Triglyceride Clearance after Ingestion of Fast Food

        Background: The exaggerated postprandial lipemia is an important stimulus for atherogenic disease progression because delayed removal of blood-borne triglyceride (TG)-rich lipoproteins encourages direct deposition of lipoprotein remnants on the arterial wall and increases the likelihood that low-density lipoprotein cholesterol will undergo disease-accelerating oxidative modification. Objective: This study was performed to evaluate the impact of visceral fat on postprandial TG clearance in non-diabetic Korean men. Design: Fifty-one men (mean age: 47 years) referring to the same occupational district, not on hypolipidemic drugs and without any other chronic disease, performed multiple TG profile (at 8 hours-fasting, and every 30min after breakfast for 8 hours) after ingestion of fast food (Big Mac, French fries, and Coca-Cola). The visceral fat (VF) measured by abdominal computed tomography. Results: TG values (mg/dl) were 124.2 (low visceral fat group, VF < 130 cm2) and 179.4 (high visceral fat group, VF > 130 cm2) at fasting (p=0.005), peak TG values were 265.3 vs. 349.6 (p=0.02) and reached at 4-4.5h after breakfast, and area under curve (AUC) were 1514 vs. 2929 8h after meal. Remnant lipoprotein-cholesterol values (mg/dL) were 26.1 vs. 35.6 (p=0.007) at fasting, and 29.3 vs. 39.6 (p=0.006) at 3.5h after meal. Fasting blood glucose (mg/dL), and insulin levels (pmol/L) were 97 vs. 112 (p=0.004), and 36.7 vs. 58.2 (p=0.004). Glucose and insulin AUC 8h after meal were 823 vs. 957.9 (p=0.013) and 118.4 vs. 225.1 (p=0.000). And Log HOMA_insulin resistance were 0.03 vs. 0.27 (p=0.01). Conclusions: We observed persistence of elevated TG level in the blood after ingestion of a high-fat test meal. The increments are directly determined by fasting TG level and the amount of visceral fat. Fasting TG level seems the major nutritional determinant of postprandial TG clearance.

        Stall inception in a high-speed centrifugal compressor has been examined. The main objective was to find stall precursor and to develop a reliable stall warning method. For stall warning, a method which uses the spectrum at impeller frequency is suggested. The use of the spectrum at impeller frequency as a stall warning method showed a warning time of about two hundreds impeller revolutions. This method uses only one sensor that it has made the stall warning method more useful. And the well-known traveling wave energy method proved to be a good method for stall warning also in a high-speed centrifugal compressor. The warning time was about one hundred impeller revolutions at lower speeds, and about one thousand impeller revolutions at higher speeds. The stall warning methods used here were found to be robust and reliable. Therefore, it seems to be promising to set up a reliable stall avoidance control based on this analysis.

      • Reduction Kinetics of Zinc Powder from Brass Converter Slag by Pyrometallurgical Method Using Hydrogen Gas

        <P>A pyrometallurgical reduction process for the recovery of copper and zinc from brass secondary slag (BSS) was studied. Speciically, the effect of reduction temperature and time on the conversion to metallic phases was investigated. The brass secondary slag was characterized by X-ray diffraction, inductively coupled plasma-atomic emission spectrometry, automatic elemental analysis, thermogravimetric analysis, and field emission scanning electron microscopy. A two-step reduction of BSS was identiied. The step 1 comprised the reduction of ZnO, while the step 2 featured the reduction of ZnO center dot Al2O3. Furthermore, the application of a first-order reaction model with Arrhenius analysis, indicated a conversion of ZnO to Zn(g) that had a rate constant increasing from 1.4 +/- 0.13 x 10(-3) s(-1) at 900 degrees C to 2.18 +/- 0.15 x 10(-2) s(-1) at 1050 degrees C. This reaction had an activation energy of 233.2 +/- 26.1 kJ/mol. Secondly, the conversion of ZnO center dot Al2O3 to Zn(g) and Al2O3 increased from 1.54 +/- 0.21 x 10(-5) s(-1) at 900 degrees C to 1.09 +/- 0.19 x 10(-3) s(-1) at 1050 degrees C, and the activation energy was 376.7 +/- 22.4 kJ/mol. This reaction mechanism and its associated kinetic data can be applied to optimize the operation conditions of recycling processes for Cu-containing wastes.</P>
