        BMP3 promoter hypermethylation in plasma-derived cell-free DNA in colorectal cancer patients

        Detecting cfDNA in plasma or serum could serve as a ‘liquid biopsy’, for circulating tumor DNA with aberrant methylation patterns offer a possible method for early detection of several cancers which could avoid the need for tumor tissue biopsies. Bone Morphogenetic Protein 3 (BMP3) was identified as a candidate tumor suppressor gene putatively down-regulated in colorectal cancer (CRC). In this study, we aimed to assess the potential role of BMP3 promoter methylation changes in plasma DNA for detection of colorectal cancerous and precancerous lesions. Plasma DNA samples were extracted from 50 patients with histologically diagnosed polyps or tumor and 50 patients reported negative for polyps or tumors. The procedure consists of bisulfite conversion of the extracted DNA, purification of bis-DNA, and BMP3 methylation status analysis by using the bisulfite specific high resolution melting analysis. This study demonstrated that there was a significantly higher frequency of BMP3 methylated DNA in plasma in patients with polyps versus healthy controls with a sensitivity and specificity of 40 and 94%, respectively. In conclusion, our results demonstrated that BMP3 DNA methylation in plasma had not have sufficient sensitivity and it should be used in combination with other biomarkers for the detection of CRC.

        Simultaneous topology and size optimization of locomotive structure using multinary genetic algorithms

        Structural optimization of a typical locomotive carbody subjected to various operational loads is investigated using a simultaneous topology and size optimization approach. The proposed approach builds on a string representation of the structure in which the presence or absence of a structural element between specific nodes, as well as its cross-section, are represented by integers. Strings representing various structure topologies are then evolved according to a modified Genetic algorithm with improved genetic operators. The efficiency of the proposed model is verified through applying it to optimize the carbody structure of ER24PC locomotive. Operational load cases and performance criteria are adopted from the European standard EN12663. It is shown that the results based on the present optimal design have higher safety factors compared to the original design without a significant increase in weight. Besides, the computational cost of the optimization process is shown to be considerably less than that of the classical binary Genetic algorithm.

        Design and Implementation of a Novel Vertical Precision Positioner

        Vertical precision positioning is distinguished from positioning in horizontal plane due to the presence of gravitational forces that work differently in “lifting” and “lowering” modes. Vertical positioners also need a “locking mechanism” to avoid the unintentional lowering of the platform. We present a vertical precision positioner that works according to the “stick–slip” phenomenon. Using an adjustable off set coupling, the linear motion of a piezoelectric actuator is converted to the rotational motion of a low friction lead screw which results in the vertical motion of the screw. A microcontroller can adjust the grip, and hence the friction, between the coupling and the lead screw to ensure the uniformity of motion in lifting and lowering modes. The dynamic model of the mechanism is extracted, and simulations are performed to discover how the peak voltage and frequency of the saw-tooth signal affect the stick–slip motion of the mechanism. Additionally, equations are verified by comparing simulation results and experiments. They can be used in the control system design in future works.

        Electrochemical and anticorrosion behavior of functionalized graphite nanoplatelets epoxy coating

        Functionalized graphite nanoplatelets (FGNP) were used as efficient and compatible nano-particles toproduce homogenous epoxy nano-coating with impressive anticorrosion behavior for carbon steel. Thecharacterizations of the nano-particle and nano-coating were carried out by SEM, FT-IR, XRD, TEM andpull-off test. Fine distribution of nano-particles in the cured nano-coating with particle sizes of 20–40 nm was obtained. Electrochemical experiments, salt spray and X-ray fluorescence showed that thenano-coatings protect the metal substrate by formation of passive layer and physical barriercharacteristics. Three samples (0.25%, 0.5% and 1%) of FGNP-epoxy coatings were prepared that 0.5%showed better anticorrosion properties.

      • Evaluation of MT1XT20 Single Quasi-Monomorphic Mononucleotide Marker for Characterizing Microsatellite Instability in Persian Lynch Syndrome Patients

        Background: Colorectal malignancies with high microsatellite instability (MSI-H), either hereditary (Lynch syndrome) or sporadic, demonstrate better prognosis and altered response to 5FU chemotherapy. It is now recommended to perform MSI testing for all new cases of colorectal cancer regardless of being categorized as hereditary or sporadic. For MSI detection, immunohistochemistry or PCR-based protocols using a cohort of various sets of STR markers are recommended. Here we aimed to evaluate a simplified protocol using just a single STR marker, MT1XT20 mononucleotide repeat, for detection of MSI in Lynch syndrome patients. A Promega five-marker MSI testing panel and immunohistochemistry (IHC) were used as the gold standard in conjunction with MT1XT20. Materials and Methods: Colorectal patients with a positive history of familial cancers were selected by evaluating medical records. Based on Amsterdam II criteria for Lynch syndrome 20 families were short listed. DNA was extracted from formalin fixed paraffin embedded tumour and adjacent normal tissues resected from the index case in each family. Extracted DNA was subjected to MT1XT20 mononucleotide marker analysis and assessment with a commercially available five marker MSI testing kit (Promega, USA). IHC also was performed on tissue sections and the results were compared with PCR based data. Results: Eight (40%), seven (35%) and five (25%) cases were MSI positive using with the Promega kit, IHC and MT1XT20, respectively. Among the markers included in Promega kit, BAT26 marker showed instability in all 8 samples. NR24 and NR21 markers showed instability in 7 (87.5%), and BAT25 and MONO 27 in 6 (75%) and 5 (62.5%). Conclusions: Although MT1XT20 was earlier reported as a valid standalone marker for MSI testing in CRC patients, we could not verify this in our Iranian patients. Instead BAT26 among the markers included in Promega MSI testing kit showed instability in all 8 MSI-H CRC samples. Therefore, it seems BAT26 could act well as a single marker for MSI testing in Iranian CRC patients.
