Objective: Oocyte in vitro maturation is frequently used for assisted reproduction since established in human and rodent. To improve the current infertility treatment, such procedures as in vitro maturation of early follicles are popularly attempted. At follicular isolation stage, unexpected rupture of follicles yields extra oocytes. Those oocytes obtained during follicle isolation from primate ovary can be used for early follicle maturation studies. Methods: Marmoset (Callithrix jacchus) is a good model used for reproductive biomedical investigation, since its reproductive physiology is very similar to that of human. After the ovaries of female marmosets were surgically retrieved, and the excess oocytes released during follicle isolation were collected without gonadotropin induction. Oocytes were matured in vitro for 72 h in the presence of various concentrations of epidermal growth factor and human chorionic gonadotropin (hCG), and the optimal maturation conditions were assessed. Each oocyte was individually prepared for RT-PCR and the expression of mRNAs and microRNAs (miRs) were evaluated. Results: hCG concentrations significantly influenced on the maturation rate of oocytes. Expression of Figla, Cja-let-7b, and Cja-mir-27a, was highest when the oocytes were matured using 300 mIU/mL hCG. From these results, these up-regulated miRs were considered to regulate the maturation of oocytes. Protein network analysis suggested an association of Figla with Nobox, GDF9 and DAZL. Conclusion: To summarize, these results suggested optimal maturation conditions of marmoset extra oocytes without using in vivo gonadotropin treatment, and showed the specific roles of miRs during early primate oocyte maturation (2016R1E1A1A01943455).

Objectives : To assess the prevalence of autoantibodies in patients with endometriosis. Materials and Methods : The prevalence of autoantibodies were investigated in 93 patients with endometriosis diagnosed by laparoscopy or laparotomy. Autoantibodies, such as antithyroglobulin antibody, antimicrosomal antibody, antinuclear antibody (ANA), antiphospho -lipid antibody (APA), lupus anticoagulant (LAC), anti-dsDNA, and rheumatoid factor, were measured in sera of the subjects. The prevalence and distribution of autoantibodies were analyzed according to the stage of endometriosis. Results : All seven autoantibodies were checked in 73 patients out of 93 patients. Autoantibodies were identified in 23 of 73 patients with endometriosis (31.5%). Antibodies detected were antithyroglobulin antibody (15.2%), ANA (12.0%), antimicrosomal antibody (8.7%), anti-dsDNA (5.4%), and rheumatoid factor (4.4%), APA (2.2%) and LAC (1.3%). The prevalence of autoantibodies did not differ by the AFS stage. Conclusion : Autoantibodies were detected in about one-third of patients with endometriosis. The prevalence of autoantibodies did not differ by the stage.

Since the introduction of laparoscopic surgery in 1950`s, pelviscopic surgery has been established as a popularly used surgical modality in many areas due to the wide spread of minimally invasive surgery in 1980`s. In spite of its many advantages compared to laparotomy, pelviscopic surgery is accompanied by many complications caused by equipments or anesthesia as various methods have been developed. Common complications include the injury of major and abdominal wall vessels, intestines, bladder and ureter, incisional hernia, heat injury by electricity, gas embolism, shoulder pain, pneumothorax, subcutaneous emphysema, and venous thrombosis. The frequency of complications is known to be 0.2-10.3%, and the half happens during the process of gas insufflation needle or trocar insertion. Order to prevent the complications, the operators should observe the safety regulations and perform the surgical procedures according the detailed operation principles. The patients with contraindications such as intestinal obstruction, hernia, severe cardiovascular diseases or peritonitis and extreme ages, should be avoided. The operators should be prepared for the expected complications by preoperatively evaluating the risk factors such as history of abdominal surgery, obesity, and heart and lung diseases. In addition, they should be well aware of biophysics of instruments, be experienced, and follow the basic safety rules such as the precautions about unnoticeable electrical damage of neighboring tissues.

Objective : The effects of superovulation on the expression of mRNA and protein of TIMP-3 and MMP-9 in murine endometrium were assessed.Methods : Using murine pregnant uteri of gestation day (g.d.) 4, 5 and 6 after injection of PMSG 5 and 10 IU, the effects of superovulation were assessed and compared with those of natural pregnancy and pseudopregnancy groups using quantitative competitive RT-PCR and immunohistochemical staining.Results : Expression of TIMP-3 mRNA and protein showed an increase in PMSG groups and pseudopregnancy group, while there was no difference in MMP-9 expression between natural pregnancy and PMSG, pseudopregnancy groups on g.d. 4 through g.d. 6. Conclusions : This study suggests that ovarian hyperstimulation by gonadotropin, which produces many oocytes and embryos, may have a detrimental effect on embryonic implantation and its relevant endometrial remodeling process by increase in expression of TIMP-3 in murine endometrium.
