Investigation of Olive Accessions using Nuclear Magnetic Resonance

In this study we used NMR to assess oil content in the 201 olive accessions in the Davis- National Clonal Germplasm Repository(NCGR) collection. Characterization of the olive collection will greatly facilitate stakeholder utilization of this important collection. Since oil content varies widely in olive accessions and oil production is the most important use of olives worldwide, characterization of oil content is critical data for the NCGR olive collection. Oil and moisture content were measured using a proton spectra and integrating this to obtain total signal intensity from the oil and moisture. Oil content is one of the most commercially important attributes of olive genotypes. NMR has been established as an accurate and rapid method for detecting oil content in intact olives. These data were collected at three dates, at monthly intervals, targeting the date of 90% color development for each accession. CPMG pulse sequence was used to acquire signal in time domain and T2 was calculated using a bi-exponential curve fitting method. Inversion recovery pulse sequence was used to acquire spectroscopic data and T1 were calculated for water and oil absorption from proton spectra. The relaxation time constants are useful indexes for examining the state of olive accessions.

The fruit of Styphnolobium japonicum L. also known as the scholar tree, has been studied for its benefits in addressing various diseases since it was shown to contain a variety of bioactive compounds. However, there has not been any investigation into its effect on photo-aging. Thus, we prepared the extract of S. japonicum fruit through a salt-out method (SJFSOE) since this method was known to demonstrate a high efficiency in extracting active ingredients from plants and evaluated the effect of SJFSOE on photo-aging induced on dorsal skin of the hairless mice by ultraviolet B (UVB) radiation (0.18 J/cm2, three times a week for 15 weeks). We found that UVB irradiation induced severe skin damages that were evidenced by significantly increased oxidative stress, inflammatory reaction, apoptosis of keratocytes, expression of genes of matrix metalloproteinases (MMPs) and NADPH oxidase 2 (Nox2), and wrinkle formation (P < 0.001). However, all these detrimental reactions were significantly reduced in the mice treated with oral administration of SJFSOE (200 mg/kg/day for 15 week) (P < 0.001). In contrast to the genes of MMPs and Nox2, the expressions of the genes of hyaluronic acid synthase (Has) 1, Has2, Has3, and collagen (COL) 1A1, COL1A2 were not affected by UVB irradiation but enhanced significantly in the mice treated with both UVB and SJFSOE. L-ascorbic acid (100 mg/kg/day for 15 weeks) showed the beneficial effects like SJFSOE. These results suggest that SJFSOE has the potential as an anti-photo-aging agent by preventing and repairing the damages induced by UV through its antioxidant activity.

In this study, AAHT was estimated using machine learning algorithms of ensemble techniques to improve the limitations of existing studies. Among the machine learning algorithms, a random forest algorithm was used, and the traffic volume estimation accuracy was analyzed as MAPE 20.7%. It was analyzed that the higher the traffic volume level, the higher the traffic volume estimation accuracy, and the traffic volume level of 3,000 vehicles/hour or more was analyzed to be 8% or less of MAPE. It was analyzed that the accuracy secured at the current level was very high compared to the existing model.

Introduction: There are no prior reports on the development of melanocytic nevus at the rectal mucosa, and therefore, it is unclear whether resection procedures must be performed or if close observation will be adequate. In the following sections, a case study on the discovery of a melanocytic nevus at the rectal mucosa and the removal process through endoscopic mucosal resection will be given. We believe that this study can contribute to future treatments of the formation of melanocytic nevus at the rectal mucosa. Case Report: A 70-year-old male with a medical history of no remarkable signs of illness has visited the hospital in 2014.01.06 for colonoscopy. During the procedure, a 1.5 cm by 1.3 cm hyperpigmented lesion was observed at the distal rectum. Biopsy was performed to the polyp at the lesion, and the acquired specimen was tested under Hematoxylin-eosin-stain and immunohistochemical stain for tumor proliferation using the Ki-67 labelling index. The Ki-67 labelling index was less than 1%, and from the results of the Hematoxylin-eosin-stain the lesion was identified as a melanocytic nevus. The patient decided to receive endoscopic resection and therefore, on 2014.11.27 prior to the surgery, colonoscopy was performed. Discussions: Melanocytic nevus may occur at the rectal mucosa which is generally not exposed to ultraviolet light, and such melanocytic nevus must be tested for malignant melanoma. Even if the lesion was concluded benign, either periodic monitoring or the removal of the lesion seems necessary.

Drug-eluting stents (DESs) have reduced restenosis and revascularization rates after percutaneous coronary intervention (PCI) compared with bare-metal stents. Despite these benefits, DESs are associated with increased risk of late thrombotic events. Bioabsorbable vascular scaffolds (BVSs) have been developed to overcome the limitations of DESs. These devices provide short-term scaffolding of the vessel and then dissolve, which should overcome the side effects of DES. The vessel recovers pulsatility and becomes responsive to shear stress and physiological cyclic strain with BVS use. BVSs are currently available in clinical practice, but their safety and feasibility have not been validated for acute myocardial infarction in Korea. We describe a patient with acute myocardial infarction who was safely and successfully treated with PCI using BVS. A 70-year-old man was referred to our hospital for acute chest pain. ECG showed ST-segment elevation in V1-4 precordial leads, coronary angiography (CAG) was performed immediately. CAG revealed total occlusion in middle portion of the LAD. We performed PCI successfully with 3.5x28mm BVS. IVUS was performed after BVS implantation identified good apposition of the scaffold to the vessel wall. After primary PCI, the patient was hemodynamically stable and did not complain of chest pain. The patient was free of symptoms when assessed 7 month later. This case suggests that PCI using BVSs can be used effectively and safely for the treatment of Korean patients with acute myocardial infarction.

Styphnolobium japonicum belongs to the Fabaceae family. This plant, particularly its fruit, is widely used in traditional medicine to treat various disorders. Lately, there has been an increased interest in using the salt-out approach for extracting active substances from plants, leading to improved yields. The purpose of this study was to prepare an extract from the S. japonicum fruit using the salt-out method (SJFSOE) and explore its benefits by examining its effects on the secretion of nitic oxide (NO) from lipopolysaccharide (LPS)-treated Raw264.7 cells (murine macrophage cells) and release of matrix metalloproteases (MMPs) from ultraviolet B (UVB)-irradiated CCD-986sk cells (normal human skin fibroblast cells). Our findings show that SJFSOE contains mainly sophoricoside (877.3 mg/g of the extract), that is over twenty times that obtained by a method using ethanol, and significantly suppressed both LPS-induced secretion of NO and UVB-stimulated release of the pro-MMP-1 and MMP-2 from the respective cells. The results imply that SJFSOE has the potential to offer benefits, especially in conditions characterized by inflammation.

Equilume light masks had no impact on hastening the resumption of estrous cyclicity in mares maintained in outdoor pastures on the mainland of Korea due to the cold weather conditions. Jeju Island is a major horse-breeding site in Korea and is warmer than the mainland during the winter season. Therefore, the primary objective of this study was to explore the efficiency of the Equilume light mask on the resumption of seasonal estrous cycles in Thoroughbred mares on Jeju Island. A total of 20 nonpregnant mares were randomly divided into the Equilume light mask (n = 9) and stable lighting (n =11) groups. The experiment was performed at seven different horse-breeding farms located on Jeju Island from November 15, 2020, to February 15, 2021. The mares were exposed to the respective lights from 16:00 to 23:00. Follicle size and uterine edema were measured by ultrasound scanning. Body condition scores (BCS) were also monitored during the experiment. Statistical analysis was conducted using the SAS and SPSS software, and p-values of < 0.05 were considered statistically significant. Two of the nine (22.2%) mares in the Equilume light mask group and three of the 11 (27.28%) mares in the stable lighting group were still cycling in December and January, which were considered as all-year-round cycling mares. On February 15, there was no difference between groups in the resumption of early seasonal estrus cycle, which was determined by follicles > 25 mm in addition to uterine edema. All mares in the Equilume light mask group and five of the eight mares (62.5%) in the stable lighting group had resumed cycling. Interestingly, six of the seven mares (87.5%) in the Equilume light mask and four of eight mares (50%) in the stable lighting group had already ovulated on February 15 (p > 0.05), as determined by the presence of a recent corpus luteum. No difference was observed in BCS and uterine edema between groups (p > 0.05). In conclusion, the Equilume light mask can be an effective approach to induce early seasonal estrus cycles of mares in Jeju Island, and it also enhances the efficiency of farm management by reducing labor.
