      • 한우에 있어서 GnRH와 PGF_2α에 의한 발정동기화 연구

        This study was conducted to get some information for the timed insemination program development of Korean native cows. Korean native cows that didn't show estrus behavior up to 60days after calving were used. Estrus synchtonization was induced by injection of PGF2α and GnRH. The estrus synchronization rate was 40.0% in treatment of PGF2α- PGF2α and 93.3% in treatment of GnRH- PGF2α -GnRH. All synchronization cows ovulated between 26 and 32hrs after the 2nd injection of GnRH and the highest ovulation rate was shown at 28hrs after the 2nd injection of GnRH. The highest pregnancy rate was shown in case of artificial insemination at 24hrs after the 2nd injection of GnRH. The pregnancy rate was 50.7% in treatment of PGF2α - PGF2α and 76.9% in treatment of GnRH- PGF2α -GnRH.

        미디어의 사회적 죽음에 대한 재현과 맥락적 의미 -고(故) 노무현 대통령 서거 TV 보도에 대한 기호네트워크 분석(Semiotic Network Analysis)-

        The purpose of this study was to investigate patterns of media coverage on the death of former president Moo-Hyun Roh, and its social, political and ideological meanings in the context to Korean society. He had been a central figure who would turn the Korean society into very severe conflicts politically as well as ideologically. Nonetheless, after this death, the Korean society changed rapidly from the atmosphere of criticizing him into the climate for cherishing his great contribution. The authors raised three research questions; that is, `Were there any differences in covering his death among TV stations? If there were differences, what were they?`, `What were differences in types of representation and discourses among TV coverages?` and `What were ideological meanings in such kinds of different types of representation and discourse?`. In order to answer these research questions, they collected news items of three TV stations-that is, KBS 1-TV, MBC TV and SBS TV-from May 23, 2009 to June 10, 2009, and analyzed them through semiotic research methods, especially, Seon-Gi Baek`s `Semitoic Network Analysis(SNA)` and `Discoursive Structure Analysis(DSN)`. As a result of this study, first of all, it was found that there were differences in covering his death among three TV stations on the basis of its own ideological position and favor for or disfavor against him. Second, Korean TV media tended to pay attention to causes of his commit suicide as if it would be a normal incident of commit suicide in order to depoliticize it. Third, they tended not to focus various meanings of his death personally but to emphasize on political and ideological meanings of his death socially in the context to Korean society. In conclusion, the Korean media had a tendency of covering his death differently on the basis of its own political and ideological position: that is, the one tried to put more ideological and political meanings on his death, while the others were inclined to put more personal meanings on his death through de-politicizing meanings of his death.

      • 폐경기 여성에서 호르몬 대치 요법시 병행 투여된 프로제스토젠 투여 방법에 따른 골대사의 생화학적 지표에 미치는 영향

        Purpose : To evaluate the impact of added progestogen on biochemical markers of the bone turnover undergoing hormone replacement therapy in the postmenopausal women. Materials and Methods : There are 71 postmentopausal women who are divided into three groups (Group I, Group II, Group III). Group I(continuous combined)is given conjugated equine estrogen (CEE, 0.625mg, day 1-30) plus medroxyprogesterone acetate (MPA, 2.5mg, day 1-30), Group II(unopposed estrogen) is given conjugated equine estrogen (CEE, 0.625mg, day 1-30) plus medroxyprogesterone acetate (MPA, 5mg, day 1-12). And there were measured serum osteocalcin, serum total alkaline phosphatase and urinary deoxypyridinoline excretion before starting hormone replacement therapy and 12 months later. Results : The serum osteocalcin, serum total alkaline phosphatase and urinary deoxypyridinoline excretion significantly decreased after hormone replacement therapy (Group, I, II, III). And there was no significance in decreases in biochemical markers of the bone turnover among three groups. Conclusions : It was concluded that the impact of added progestogen on the biochemical markers of the bone turnover undergoing hormone replaced therapy in postmenopausal women was not significant than nothing added group. Thus we concluded that progestogen was not impact on decreasing on the biochemical markers of the bone turnover in postmenopausal women undergoing hormone replacement therapy.

      • 제주재래마 특이 표지인자 개발

        본 연구는 제주재래마의 유전적 특성파악과 이를 통한 타품종과의 유전적 특이성을 가지는 DNA 표지인자를 개발하고자 PCR-RAPD기법을 이용하여 제주재래마와 더러브렛, 가고시마의 3품종에서의 다형 band에 대한 특성파악을 실시한 결과이다. 각 품종별로 10두의 혈액을 혼합한 후 분석하였고 200개의 random primer들 중 PCR증폭양상의 재현성을 기준으로 70개의 primer를 선별하였다. 또한 선별된 primer들중 다형성을 9개의 primer를 이용하여 개체별 DNA분석을 실시하였으며 9개의 prkmer에 의해 나타난 15개의 band는 제주재래마 특이 band, 4개는 더러브렛종에 그리고 4개는 가지고시마 재래마에 품종특이 RAPD band가 나타났다. 따라서 RAPD기법에 의해 품종간의 차이를 보이는 표지인자의 개발이 가능할 뿐 아니라 품종간의 특성차이를 보이는 유전인자 개발과 특성파악이 용이한 것으로 평가되었다. This study was carried out using PCR-RAPD technique to estimate genetic characteristics of Cheju Horse (Jorangmal) and to develop the DNA marker for bread identification in horse. Each three horse breeds was represented by pooled DNA from 10 individuals and tested for 200 different random primers for their reproducible PCR pattern. From them 70 primer were selected and 9 primers were used to detect the difference between breeds at the individual level. Fifteen highly polymorphic bands were observed between breeds using 9 primers . Additionally, the 15 most polymorphic bands showed great difference between breeds in their appearing frequencies, implicating their usefulness in the breed discrimination and in the characterization of genetic structure. Therefore, the polymorphism of RAPD could be used in characterizing genomic difference between breeds and some genetic mechanism that give rise to breed disctimination in horse.

        Clinical significance of nontuberculous mycobacteria from respiratory specimens in stem cell transplantation recipients.

        <P>The clinical importance of the isolation of nontuberculous mycobacteria (NTM) from respiratory specimens of stem cell transplant (SCT) recipients is not clear. We investigated the characteristics and clinical impact of NTM isolation in this population. Medical records of adult patients who underwent SCT at the blood and marrow transplantation center at a University Hospital in South Korea between January 2004 and December 2013 were retrospectively reviewed. Twenty-five of 2,658 patients were enrolled, an incidence of 0.94%. The most common NTM species isolated were Mycobacterium avium complex (n = 15) and Mycobacterium abscessus (n = 5). Sixteen patients were co-infected with other pathogens, including Aspergillus species (10 cases). Nonspecific pneumonia and cavitary pneumonia were the most common radiologic patterns. Sixteen patients had underlying lung graft-versus-host disease, including bronchiolitis obliterans (10 cases) with moderate obstructive lung defects. Whereas two of 10 patients who were treated for NTM died as a result of NTM progression, one of 15 patients who were not treated died of the same cause. This study shows that the incidence of respiratory NTM detection in SCT recipients was low and its virulence was not higher than expected. As patients frequently had concomitant infections and underlying lung diseases that made radiologic evaluation more difficult, we suggest that it would be advantageous to perform repeated microbiologic studies when NTM infection is clinically suspected.</P>

        Cks1 regulates human hepatocellular carcinoma cell progression through osteopontin expression

        <P><B>Abstract</B></P> <P>Precise cell cycle regulation is critical to prevent aberrant cell proliferation and cancer progression. Cks1 was reported to be an essential accessory factor for SCF<SUP>Skp2</SUP>, the ubiquitin ligase that targets p27<SUP>Kip1</SUP> for proteasomal degradation; these actions drive mammalian cell transition from G1 to S phase. In this study, we investigated the role played by Cks1 in the growth and progression of human hepatocellular carcinoma (HCC) cells. Silencing Cks1 expression abrogated osteopontin (OPN) expression in a p27<SUP>Kip1</SUP>-dependent manner in Huh7 HCC cells. OPN increased the proliferation, migration and invasion of Huh7 cells. Pharmacological inhibitor studies demonstrated that ERK1/2 signaling is responsible mainly for Cks1-mediated OPN expression. Cks1 appears to regulate ERK1/2 signaling through the expression of dual-specificity phosphatase 16 (DUSP16) because both Cks1 knockdown, which leads to DUSP16 upregulation, and DUSP16 overexpression decreased ERK1/2 phosphorylation and the resulting OPN expression. The same is true for the Cks1-mediated increases in p27<SUP>Kip1</SUP>, suggesting that Cks1 regulates OPN expression through activating ERK1/2 signaling either by suppressing DUSP16 expression or by a p27<SUP>Kip1</SUP>-dependent mechanism. Cks1 and OPN expression levels were significantly higher, but DUSP16 expression levels were significantly lower in HCC tissues than in normal liver tissues. Both Cks1 and OPN expression were negatively correlated with DUSP16 expression, whereas Cks1 expression was positively correlated with OPN expression. Moreover, combined panels for the expression levels of Cks1, DUSP16 and OPN showed significant prognostic power for the risk assessment of HCC patient overall survival. In conclusion, our data propose a novel function for Cks1 as a tumor promoter through the expression of the strongly oncogenic protein OPN in HCC.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Cks1 promotes OPN expression in human HCC cells. </LI> <LI> The Cks1-mediated upregulation of DUSP16 or p27<SUP>Kip1</SUP> decreased OPN expression. </LI> <LI> DUSP16- or p27<SUP>Kip1</SUP>-mediated ERK1/2 inactivation is responsible for OPN abrogation. </LI> <LI> Cks1, DUSP16 and OPN may be a useful prognostic biomarker panel for HCC patients. </LI> </UL> </P>

        DUSP1 induces paclitaxel resistance through the regulation of p-glycoprotein expression in human ovarian cancer cells

        <P><B>Abstract</B></P> <P>The heterogeneity and genetic instability of ovarian cancer cells often lead to the development of drug resistance, closely related with the increased cancer-related mortality. In this study, we investigated the role of dual-specificity phosphatase 1 (DUSP1) in the development of the resistance in human ovarian cancer cells against paclitaxel. Overexpression of DUSP1 in HeyA8 human ovarian cancer cells (HeyA8-DUSP1) up-regulated the expression of the drug efflux pump, p-glycoprotein. Consequently, HeyA8-DUSP1 cells are highly resistant to paclitaxel, with the resistance comparable to that of a multi-drug resistance cell line (HeyA8-MDR). Moreover, over expression of DUSP1 significantly increased the activation of p38 MAPK, leaving the activation of ERK1/2 and JNK1/2 unaffected. Pharmacological suppression of p38 MAPK activity prevents the up-regulation of p-glycoprotein expression and the consequent resistance against paclitaxel in HeyA8-DUSP1 cells. By contrast, HeyA8-MDR cells expressed a significantly higher level of DUSP1, but treatment with small interference RNA against DUSP1 significantly suppressed the expression of p-glycoprotein and the resistance against paclitaxel in HeyA8-MDR cells. Ectopic expression of MKK3, an upstream activator of p38 MAPK, significantly up-regulated the expression of p-glycoprotein and increased the consequent resistance against paclitaxel in HeyA8 cells. Collectively, these data indicated that DUSP1 may induce the resistance against paclitaxel through the p38 MAPK-mediated overexpression of p-glycoprotein in human ovarian cancer cells.</P> <P><B>Highlights</B></P> <P> <UL> <LI> DUSP1 induces Pgp expression and paclitaxel resistance in ovarian cancer cells. </LI> <LI> DUSP1 mediates p38 MAPK activation leading to Pgp-mediated paclitaxel resistance. </LI> <LI> Inhibition of DUSP1 or p38 MAPK reverses Pgp-mediated paclitaxel resistance. </LI> <LI> DUSP1-p38 MAPK-Pgp axis is a novel mechanism for paclitaxel resistance. </LI> </UL> </P>

        New PTS Schemes With Adaptive Selection Methods of Dominant Time-Domain Samples in OFDM Systems

        <P>In orthogonal frequency division multiplexing (OFDM) systems, high peak-to-average power ratio (PAPR) of OFDM signals is one of the most important problems. As a solution to the PAPR problem in OFDM systems, the partial transmit sequence (PTS) is a fairly suitable scheme due to its PAPR reduction performance and distortionless characteristic. However, high computational complexity is a serious problem in the PTS scheme. In this paper, in an effort to reduce its computational complexity, new PTS schemes are proposed using dominant time-domain samples of OFDM signals. Although the proposed PTS schemes use dominant time-domain samples in a manner similar to several existing low-complexity PTS schemes, we propose more efficient selection methods for dominant time-domain samples. The proposed PTS schemes lower the computational complexity compared to the conventional PTS schemes while achieving the optimal PAPR reduction performance.</P>
