      • NMR study of hydrogen exchange during the B-Z transition of a DNA duplex induced by the Zα domains of yatapoxvirus E3L

        The Yaba-like disease viruses (YLDV) are members of the Yatapoxvirus family and have double-stranded DNA genomes. The E3L protein, which is essential for pathogenesis in the vaccinia virus, consists of two domains: an N-terminal Z-DNA binding domain and a C-terminal RNA binding domain. The crystal structure of the E3L orthologue of YLDV (yabZα<SUB>E3L</SUB>) bound to Z-DNA revealed that the overall structure of yabZα<SUB>E3L</SUB> and its interaction with Z-DNA are very similar to those of hZα<SUB>ADAR1</SUB>. Here we have performed NMR hydrogen exchange experiments on the complexes between yabZα<SUB>E3L</SUB> and d(CGCGCG)<SUB>2</SUB> with a variety of protein-to-DNA molar ratios. This study revealed that yabZα<SUB>E3L</SUB> could efficiently change the B-form helix of the d(CGCGCG)<SUB>2</SUB> to left-handed Z-DNA via the active-mono B-Z transition pathway like hZα<SUB>ADAR1</SUB>1.

        Purposes :The purposes of this study was to investigate the self-managing status according to the life styles and the diabetic knowledge of early adulthood. The subjects in this study were 240people, the early adult aged twenties to thirties living in the metropolitan area. Data were collected by using self-administered questionnaires that measure about the knowledge of diabetes; the dietetic behavior; the knowledge of diet; the exercise behavior; the knowledge of exercise. Descriptive statistics, t-test, ANOVA, and correlation were used for data analysis. Result :1. At the knowledge of diabetes degree based on the general character, the subjects who had experiences of diabetic education have higher degree of the diabetic knowledge. 2. At the knowledge of diabetes degree according to the life style, the subjects who have regular diet are appeared highly. Also the subjects who don't take much food of animal origin have higher degree. 3. At the knowledge of diet based on the life style, the subjects who don't keep three times meals a day are appeared highly. Also the subjects who have regular diet have higher degree. 4. At the knowledge of exercise according to the life style, the subjects who take extra nutritions 3-4 times a week are appeared highly. Although the subjects tend to know about dietary, exercise and health management, it showed that they actually don't perform what they know in their everyday of life. The reason for this could be the lack of perceived seriousness about diabetes mellitus. Thus as nursing professionals, we should provide them proper health education and support. Moreover, advanced research on the empowerment is warranted.

        Production of 10-hydroxystearic acid from oleic acid by whole cells of recombinant Escherichia coli containing oleate hydratase from Stenotrophomonas maltophilia

        A putative fatty acid hydratase from Stenotrophomonas maltophilia was cloned and expressed in Escherichia coli. The recombinant enzyme showed the highest hydration activity for oleic acid among the fatty acids tested, indicating that the enzyme is an oleate hydratase. The optimal conditions for the production of 10-hydroxystearic acid from oleic acid using whole cells of recombinant E. coli containing the oleate hydratase were pH 6.5, 35<SUP>o</SUP>C, 0.05% (w/v) Tween 40, 10gl<SUP>-1</SUP> cells, and 50gl<SUP>-1</SUP> oleic acid. Under these conditions, whole recombinant cells produced 49gl<SUP>-1</SUP> 10-hydroxystearic acid for 4h, with a conversion yield of 98% (w/w), a volumetric productivity of 12.3gl<SUP>-1</SUP>h<SUP>-1</SUP>, and a specific productivity of 1.23gg-cells<SUP>-1</SUP>h<SUP>-1</SUP>, which were 18%, 2.5-, and 2.5-fold higher than those of whole wild-type S. maltophilia cells, respectively. This is the first report of 10-hydroxystearic acid production using recombinant cells and the concentration and productivity are the highest reported thus far among cells.

      • Overexpression of angiotensin II type 1 receptor in breast cancer cells induces epithelial-mesenchymal transition and promotes tumor growth and angiogenesis

        The angiotensin II type I receptor (AGTR1) has been implicated in diverse aspects of human disease, from the regulation of blood pressure and cardiovascular homeostasis to cancer progression. We sought to investigate the role of AGTR1 in cell proliferation, epithelial-mesenchymal transition (EMT), migration, invasion, angiogenesis and tumor growth in the breast cancer cell line MCF7. Stable overexpression of AGTR1 was associated with accelerated cell proliferation, concomitant with increased expression of survival factors including poly(ADP-ribose) polymerase (PARP) and X-linked inhibitor of apoptosis (XIAP), as well as extracellular signal-regulated kinase (ERK) activation. AGTR1-overexpressing MCF7 cells were more aggressive than their parent line, with significantly increased activity in migration and invasion assays. These observations were associated with changes in EMT markers, including reduced E-cadherin expression and increased p-Smad3, Smad4 and Snail levels. Treatment with the AGTR1 antagonist losartan attenuated these effects. AGTR1 overexpression also accelerated tumor growth and increased Ki-67 expression in a xenograft model. This was associated with increased tumor angiogenesis, as evidenced by a significant increase in microvessels in the intratumoral and peritumoral areas, and enhanced tumor invasion, with the latter response associated with increased EMT marker expression and matrix metallopeptidase 9 (MMP-9) upregulation. In vivo administration of losartan significantly reduced both tumor growth and angiogenesis. Our findings suggest that AGTR1 plays a significant role in tumor aggressiveness, and its inhibition may have therapeutic implications.

        Purpose: The purpose of this study was to provide basic data for use in the upgrade of healthcare for the elderly and to provide guidance toward educating the elderly in the proper use of medications by determining the level of drug knowledge, current use, and adherence by the elderly. Method: The participants of this study were selected using the convenient sampling method. Participants of this study comprised of 160 elderly who were aged 65 or older, who were participants to the elderly municipal welfare center in Seoul. The instrument used for this study was a structured questionnaire (Han,2011). Collected data were analyzed using the SPSS20.0 program with descriptive statistics, t-test, ANOVA, and Pearson’s correlation coefficient. Result: The mean score for drug knowledge was 7.6±2.9 and drug misuse was 4.7±2.8. There were significant differences about drug knowledge according to the following: level of education (F=8.053 p=.000), and spouse (t=2.14 p=.034). There were significant differences between drug misuse and monthly income (F=2.167 p=.040). There was no statistically significant relationship between drug knowledge and drug misuse (r=-.087, p=.271). Conclusion: A replication study based on broader region and in-depth research is needed. The development of the customized education protocol considering various levels of education, income, spouse and physical condition are needed. Further research is suggested to find out the status of medication education for the elderly by nurses.

        Detailed analysis of the density change on chest CT of COPD using non-rigid registration of inspiration/expiration CT scans

        <P>Objectives One objective was to evaluate the air trapping index (ATI), measured by inspiration/expiration CT, in COPD patients and nonsmokers. Another objective was to assess the association between the pulmonary function test (PFT) and CT parameters such as ATI or other indices, separately in the whole lung, in emphysema, and in hyperinflated and normal lung areas. Methods One hundred and thirty-eight COPD patients and 29 nonsmokers were included in our study. The ATI, the emphysema index (EI), the gas trapping index (Exp -856) and expiration/inspiration ratio of mean lung density (E/Iratio of MLD) were measured on CT. The values of the whole lung, of emphysema, and of hyperinflated and normal lung areas were compared and then correlated with various PFT parameters. Results Compared with nonsmokers, COPD patients showed a higher ATI in the whole lung and in each lung lesion (all P<0.05). The ATI showed a higher correlation than EI with FEF25-75%, RVand RV/TLC, and was comparable to Exp -856 and the E/I ratio of MLD. The ATI of emphysema and hyperinflated areas on CT showed better correlation than the normal lung area with PFT parameters. Conclusions Detailed analysis of density change at inspiration and expiration CT of COPD can provide new insights into pulmonary functional impairment in each lung area.</P>

      • Yongdamsagan-tang, a traditional herbal formula, inhibits cell growth through the suppression of proliferation and inflammation in benign prostatic hyperplasia epithelial-1 cells

        Ethnopharmacological relevance: Benign prostatic hyperplasia (BPH), also called benign enlargement of the prostate, is a progressive disease that is observed in most elderly men. Yongdamsagan-tang, a traditional herbal formula, is used commonly for the treatment of inflammation-related diseases. Although the therapeutic efficacy of Yongdamsagan-tang against BPH in vivo was reported previously, its underlying mechanisms are not clearly understood. Aim of the study: In this study, we investigated the effect of Yongdamsagan-tang water extract (YSTE) and its mechanism on the growth of human BPH epithelial BPH-1 cells. Materials and methods: YSTE was extracted from 11 herbaceous plants and its chemical composition was analyzed by High-performance liquid chromatography (HPLC). YSTE was treated in the epithelial BPH-1 cell line and then cell lysates or supernant were used to evaluate cell viability, cell cycle, proliferation and cytokine production. Results: HPLC revealed that Baicalin and gentiopicroside were involved as the major compounds of YSTE. YSTE treatment in BPH-1 cells repressed cell viability in a dose-dependent manner. Regarding the inhibitory mechanisms of YSTE on cell growth, YSTE inhibited cell proliferation via a decrease in endogenous cyclin D1 protein levels and arrest at the S phase during cell-cycle progression. Furthermore, YSTE treatment in BPH-1 cells suppressed prostaglandin E<SUB>2</SUB> production and cyclooxygenase-2 (COX-2) protein levels. The secretion of the proinflammatory cytokines, interleukin-8 and interleukin-6, was also reduced by YSTE treatment. Conclusions: YSTE in BPH-1 cells showed antiproliferative and anti-inflammatory activities via cell-cycle arrest and downregulation of COX-2 expression, respectively. Taken together, the results of the present study will enhance our understanding of the mechanisms underlying the effect of YSTE in BPH.

      • 중년 남성의 폐경 지식, 폐경 태도가 배우자 지지에 미치는 영향

        Purpose: The purpose was to investigate the relationship between middle-aged men’s menopausal knowledge, menopausal attitude and their support for spouses with menopause. Methods: A correlational descriptive design was used. A convenience sampling was used. The participants were middle-aged men living in Korea(n=150). The instruments of the Knowledge of Menopause Scale, the Attitude of Menopause Measurement and the Spouse Support Scale were used. Result: Multiple regression analysis was performed. The middle-aged men’s menopausal knowledge have positive relationship with their attitude and support for their spouses with menopause. Also, the middle-aged men’s menopausal attitude have positive relationship with support for their spouses with menopause. Conclusion: The study concluded that the higher middle-aged men’s menopausal knowledge and the more positive middle-aged men’s menopausal attitude lead to the more their support on spouses with menopause. Therefore middle-aged men’s spouse support is necessary to improve quality of middle-aged woman’s life with menopause and their mental health, and further studies are also required to confirm.
