<P><B>Abstract</B></P> <P>It is urgently necessary to develop reliable non-invasive stem cell imaging technology for tracking the <I>in vivo</I> fate of transplanted stem cells in living subjects. Herein, we developed a simple and well controlled stem cell imaging method through a combination of metabolic glycoengineering and bioorthogonal copper-free click chemistry. Firstly, the exogenous chemical receptors containing azide (-N<SUB>3</SUB>) groups were generated on the surfaces of stem cells through metabolic glycoengineering using metabolic precursor, tetra-acetylated <I>N</I>-azidoacetyl-<SMALL>D</SMALL>-mannosamine(Ac<SUB>4</SUB>ManNAz). Next, bicyclo[6.1.0]nonyne-modified glycol chitosan nanoparticles (BCN-CNPs) were prepared as imageable nanoparticles to deliver different imaging agents. Cy5.5, iron oxide nanoparticles and gold nanoparticles were conjugated or encapsulated to BCN-CNPs for optical, MR and CT imaging, respectively. These imageable nanoparticles bound chemical receptors on the Ac<SUB>4</SUB>ManNAz-treated stem cell surface specifically via bioorthogonal copper-free click chemistry. Then they were rapidly taken up by the cell membrane turn-over mechanism resulting in higher endocytic capacity compared non-specific uptake of nanoparticles. During <I>in vivo</I> animal test, BCN-CNP-Cy5.5-labeled stem cells could be continuously tracked by non-invasive optical imaging over 15 days. Furthermore, BCN-CNP-IRON- and BCN-CNP-GOLD-labeled stem cells could be efficiently visualized using <I>in vivo</I> MR and CT imaging demonstrating utility of our stem cell labeling method using chemical receptors. These results conclude that our method based on metabolic glycoengineering and bioorthogonal copper-free click chemistry can stably label stem cells with diverse imageable nanoparticles representing great potential as new stem cell imaging technology.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

최근 들어 ‘데이트 폭력’(Dating Abuse, 연인간 폭력)이 사회적 이슈로 떠오르고 있다. 하지만 우리나라는 데이트 폭력을 처벌하는 규정을 따로 두고 있지 않기 때문에 데이트 폭력에 대한 효과적 대책으로 관련 특별법이 제정되어야 한다는 주장이 제기되고 있다. 특히 경찰은 영국에서 시행하고 있는 일명 「클레어법」의 도입을 추진하고 있다. 다른 한편에서는 데이트 폭력이 스토킹범죄와 관련성이 많기 때문에 「스토킹방지법」의 제정을 통해 데이트 폭력에 대처하자는 주장도 있고, 「가정폭력처벌법」상 대상범위를 확대하여 가정폭력범죄에 데이트 폭력범죄를 포함시켜 데이트 폭력피해자를 가정폭력피해자와 같이 보호하자는 주장도 제기되고 있다. 그러나 이러한 입법적 대처방안들이 그 타당성이나 실효성에 대한 검증이 안 된 상태에서 도입되었을 경우 입법의 낭비가 되어 오히려 데이트 폭력을 예방하는데 저해요인이 될 수 있다는 점이 간과되어서는 안 될 것이다. 특히 우리 입법현실을 충분히 고려하지 않고 새로운 제도가 도입될 경우 운영상 많은 문제점이 발생할 우려가 있다. 기존의 입법으로도 충분히 대처할 수 있거나 관련 법률의 개정을 통해 대처할 수 있다면 굳이 새로운 입법을 할 필요성이 없을 것이다. 예컨대 스토킹 유형의 데이트 폭력범죄에 대처하는데 현행 「경범죄처벌법」이 적절하지 않다면 스토킹 범죄를 「형법」으로 입법화하면 될 것이고, 경찰권의 발동이 데이트 폭력에 소극적이라면 「경찰관직무집행법」의 개정을 통해 경찰권이 적극적으로 발동되도록 하면 될 것이다. Recently, dating abuse has become a social issue. For the reason, an argument to legislate a special law for effective measures against dating abuse has been raised. Especially, the police is pushing ahead with introduction of so-called 「Clares Law」 that England is currently enforcing. Others say it is necessary to handle through legislation of the 「Anti-stalking Law」 as dating abuse has high relevance to stalking, and an assertion to protect victims of dating abuse with victims of domestic violence by expanding objects of the 「Laws on Domestic Violence」 and including dating abuse in the domestic violence. In the event that such legislative measures are introduced without verification of validity or effectiveness, however, it may be a waste of legislation. It may be rather an inhibiting element in preventing dating abuse as well. In case a new system is introduced without careful consideration of the reality of our legislation, it is apprehended that many problems will occur. If we can deal with dating abuse sufficiently with existing legislation or through revision of relevant laws, there would be no need to legislate anew.

Sediments from Ocean Driling Program (ODP)Sites 980 and 983 in the North Atlantic Ocean were analyzed toobtain evidence of long-range Milankovitch cycles and to examinethe cycles’ effect on the paleoceanographic evolution of the NorthAtlantic Ocean. Wide cyclic variations in total organic carbon andbiogenic carbonate occur throughout the columns at both sites andprovide distinctive characteristics of both sediment groups. Spec-tral modeling of these variations shows typical 100-ka cyclic vari-ations in both the total organic carbon (TOC) and carbonate recordsat Site 980, although this 10-ka Milankovitch frequency occursonly in the uper, ~472.5 ka, section of the core. In Site 983, only400-ka cycle in carbonate is observed but the 10-ka cycle in TOCand carbonate is absent. The terrigenous content, expressed interms of K, Al, Ti, and Th, also shows strong 100-ka and 400-kacyclic variations at Site 983. The earth’s eccentricity as expressedand precession (23-ka) are important characteristics of North Atlan-tic Ocean sediments. Milankovitch pulse differences in carbonate,TOC at the two sites likely arise from the evolution of paleocean-ography. The dilution of carbonate fractions by terrigenous mate-rials (indicated by the cyclical behavior of trace elements) at Site 983is one of plausible explanation. Climatic warming over the last 600ka probably caused the differences in the sedimentary cycles at thetwo sites; induced meltwater discharge is recorded in the terrige-nous record, and changes in the oceanic circulation system arerelated to major glacialinterglacial climatic episodes that probablyunderlie the differences in the cyclical records.

<P>Biological ligands such as aptamer, antibody, glucose, and peptide have been widely used to bind specific surface molecules or receptors in tumor cells or subcellular structures to improve tumor-targeting efficiency of nanoparticles. However, this active-targeting strategy has limitations for tumor targeting due to inter- and intraheterogeneity of tumors. In this study, we demonstrated an alternative active-targeting strategy using metabolic engineering and bioorthogonal click reaction to improve tumor-targeting efficiency of nanoparticles. We observed that azide-containing chemical reporters were successfully generated onto surface glycans of various tumor cells such as lung cancer (A549), brain cancer (U87), and breast cancer (BT-474, MDA-MB231, MCF-7) via metabolic engineering in vitro. In addition, we compared tumor targeting-of artificial azide reporter with bicyclononyne (BCN)-conjugated glycol chitosan nanoparticles (BCN-CNPs) and integrin alpha(v)beta(3) with cyclic RGD-conjugated CNPs (cRGD-CNPs) in vitro and in vivo. Fluorescence intensity of azide-reporter-targeted BCN-CNPs in tumor tissues was 1.6-fold higher and with a more uniform distribution compared to that of cRGD-CNPs. Moreover, even in the isolated heterogeneous U87 cells, BCN-CNPs could bind artificial azide reporters on tumor cells more unifornaly (similar to 92.9%) compared to cRGD-CNPs. Therefore, the artificial azide-reporter-targeting strategy can be utilized for targeting heterogeneous tumor cells via bioorthogonal click reaction and may provide an alternative method of tumor targeting for further investigation in cancer therapy.</P>

<P>The CO-assisted thermal decomposition of a new, surfactant-ligated Pt precursor, [Pt(acac)(NHR)]<SUB><I>n</I></SUB> (<I>n</I> = 2, 3; R = C<SUB>18</SUB>H<SUB>37</SUB>), gives structurally uniform five-fold twinned Pt nanorods. Objective: The present study aimed to evaluate the efficacy of repetitive transcranial magnetic stimulation (rTMS) at a high frequency using a miniaturized device compared to standard rTMS and sham rTMS for the treatment of depression. Methods: Fifty-four patients with depression were randomly assigned to either 15 days of miniaturized, standard, or sham rTMS. The stimulation consisted of 60 trains of 5 seconds at 10 Hz for 30 minutes. Clinical measures were assessed at baseline and on the final day of the stimulation. Results: A repeated-measures analysis of variance revealed a significant main effect of time and a time by group interaction on Hamilton Rating Scale for Depression scores. There were no significant correlations between individual motor thresholds and changes of clinical outcomes. Our results revealed a significant reduction in the Hamilton Rating Scale for Depression in the miniaturized and standard groups compared to the sham group. Conclusion: The antidepressant utility of miniaturized rTMS using subthreshold stimulation was comparable to that of standard stimulation.
