Partial Polyhedrin Fusion Expression Enhance the Production of Recombinant Protein in Baculovirus

Polyhedrin is the major component of the nuclear viral occlusions produced during replication of the baculovirus Autographa californica multicapsid nuclear polyhedrosis virus (AcMNPV). To enhance the expression level of baculovirus vector system, we constructed several fusion vectors using various fragments of the polyhedrin. The polyhedrin fragments were genetically fused to the enhanced green fluorescent protein (eGFP) under the control of polyhedrin promoter, and their expressions were analyzed in Sf21 insect cells. Expression of the fusion protein was identified by SDS-PAGE and Western blot analysis using anti-GFP and anti-Polyhedrin. The expression level of eGFP was markedly increased by the fusion of partial polyhedrin. Also, the fluorescence intensity of fusion proteins was higher than that of non-fusion protein. Confocal laser scanning microscopy demonstrated that fusion proteins were localized to the cytosol or nucleus of insect cells. In additional, the glycoprotein E2 (gE2) of classical swine fever virus (CSFV) expressed by the these vectors was dramatically increased and its immunogenicity was proofed using experimental animal guinea pigs that were immunized with the partial polyhedrin containing gE2. This study provides a new option for the higher expression of useful foreign recombinant protein by using the partial polyhedrin in BEVS.

이분 주상골은 드문 기형이며 아직도 그 선천성 혹은 외상성의 논란의 여지는 있으나 대부분의 보고가 선천성의 가능성을 시사 하고 있다. 현재 까지 이분 주상골에 대한 국내 증례보고는 없었으며 이에 저자들은 38세 남자의 이분 주상골의 증례를 경험하였기에 문헌 고찰과 함께 보고하고자 한다. Bipartite scaphoid is a rare anomaly and it has been questioned as whether it is of congenital origin or traumatic. However, many authors have reported that congenital origin is more probable. There was no case report in Korea about bipartite scaphoid, but this article reports a case of a 38-year-old male with bilateral bipartite scaphoid.

Purpose: The safety, efficiency, and versatility of novel surgical energy devices have been proved by recent studies. This study aims to investigate the impact of surgical energy devices on operative and oncologic outcomes of minimally invasive colorectal cancer surgery. Methods: The study group included 80 patients who underwent minimally invasive colorectal cancer surgery with a conventional monopolar device and 217 patients with advanced surgical energy devices between August 2015 and December 2017. The propensity score matching for tumor lesion, preoperative level of CEA, and operation technique produced 63 matched pairs. Results: In patient characteristics, there was no significant difference between the groups after the propensity score matching. The amount of blood loss (72 mL . 54 mL, P = 0.123) and conversion cases to another surgery (11.1% . 4.8%, P = 0.187) tended to be higher in monopolar group, while operation time and intraoperative complications were not significantly different. The short-term clinical outcomes including time to soft diet, the length of hospital stays, and the morbidity within 30 days after surgery or pathologic outcomes were comparable between the groups. During the median follow-up of 52.9 and 51.1 months in each study group, the 5-year overall survival rates of the monopolar and advanced energy groups were 84.6% and 91.6% (P = 0.276), and the 5-year disease-free survival rates were 78.0% and 84.6% (P = 0.328), respectively. Conclusion: The use of surgical energy devices based on surgeons’ preference did not show significant impact on operative and long-term outcomes compared with conventional monopolar devices in minimally invasive colorectal cancer surgery.
Processing and Pigment Stability of Cooked and Frozen Cockle, Fulvia mutica

Processing condition and pigment maintenance of cockle, Fulvia mutica were studied. Proximate composition of whole meat was $82.3\%$ moisture, $10.8\%$ crude protein, $0.8\%$ crude lipid, $2.5\%$ carbohydrate and $3.1\%$ crude ash, and that of foot muscle was $80.6\%,\;12.3\%,\;0.3\%,\;2.9\%\;and\;3.3\%$ respectively. When the living cockle was soaked in $2\%$ NaCl solutions, about $90\%$ of silt and mud was removed after 10 hours soaking, and over $92\%$ was removed when the pH was adjusted to 7.5. When the pigment destruction was tested by 40 seconds at $75^{\circ}C,\;80^{\circ}C,\;85^{\circ}C,\;90^{\circ}C\;and\;95^{\circ}C$, retention ratios of pigment in cockle were above $95\%$ at all temperature. Soaking in ethanol for 5 minutes resulted in strong adhesion of pigment to meat. Soaking in seasoning liquid containing $10\%$ soy sauce, $5\%$ wasabi, $5\%$ sugar, $2\%$ vinegar, $2\%$ powdered garlic for 3 minutes was effective for instant processing of cooked and frozen cockle after thawing. After 60 days storage at $-20^{\circ}C$, the contents of moisture, crude lipid, carbohydrate, ash and salinity were not changed so much, and pH and TBA values increased to 6.6 and 0.3 compared with 6.2 and 0.2, respectively, while pigment absorbance at 226 nm was decreased from 2.7 to 2.3. However, in case of 60 days storage at $-45^{\circ}C$, there was no change in these compositions.

<P>An effective combination of host and guest molecules in a framework type of architecture can enhance the structural stability and physical properties of clathrate compounds. We report here that an organic clathrate compound consisting of a fullerene (C<SUB>60</SUB>) guest and a hydroquinone (HQ) host framework shows enhanced hydrogen-storage capacity and good structural stability under pressures and temperatures up to 10 GPa and 438 K, respectively. This combined structure is formed in the extended β-type HQ clathrate and admits 16 hydrogen molecules per cage, leading to a volumetric hydrogen uptake of 49.5 g L<SUP>-1</SUP> at 77 K and 8 MPa, a value enhanced by 130% compared to that associated with the β-type HQ clathrate. A close examination according to density functional theory calculations and grand canonical Monte Carlo simulations confirms the synergistic combination effect of the guest-host molecules tailored for enhanced hydrogen storage. Moreover, the model simulations demonstrate that the lithium-doped HQ clathrates with C<SUB>60</SUB> guests reveal exceptionally high hydrogen-storage capacities. These results provide a new playground for additional fundamental studies of the structure-property relationships and migration characteristics of small molecules in nanostructured materials.</P> [FIG OMISSION]</BR>
