대두와 유사한 ureide대사를 갖는 분열효모 S.pombe에 뿌리혹 특이성 uricase를 만드는 Nodulin-35 유전자의 1.1kb 의 작동역을 pART2 벡터에 크로닝하였다. 이 재조합 프라스미드의 S.pombe 형질전환체로부터 CAT 합성을 측정한 결과 뿌리혹에서만 발현되는 Nodulin-35 유전자가 유사한 대사를 갖고 있는 효모에서 항상성(constitutive) 발현을 하였다. Soybean nodulin-35(N-35) promoter region was cloned in Schizosac-charomyces vector, pART2. The chimaeric gene consisted of 1.1 kb of the N-35 promoter region, chloramphenicol acetyl transferase (CAT) and N-23 polyA. After transformation into S. pombe SP223 transformants were selected with auxortophic marker.CAT activity of the transformant was showed. These data indicate that N-35 gene can express in S.pombe.

Background: Transverse facial clefts are Tessier's number 7 facial cleft among numbers 1-15 in Tessier's classification of craniofacial malformations, which varies from a simple widening oral commissure to a complete fissure extending towards the external ear. Case presentation: In a patient with a transverse facial cleft, to functionally arrange the orbicularis oris muscle and form the oral commissure naturally, we performed a surgical procedure including orbicularis oris muscle reconstruction and cheiloplasty with Z-plasty. Conclusion: We achieved good results functionally and esthetically by orbicularis oris muscle reconstruction and cheiloplasty with Z-plasty. The surgical modality of our anatomical repair and 3 months follow-up results are presented.
Transformation and Expression of a Chimaeric Soybean Nodulin-35 Gene in Transgenic Nicotiana tabacum

대두의 뿌리혹(nodule)에서만 발현되는 뿌리혹 특이성 uricase인 nodulin-35의 유전자로부터 1.1kb의 작동역(promoter region)을 크로닝하였다. 이 작동역 바로 뒤에 CAT 구조유전자를 붙이고, 이 뒤에 N-23유전자의 3' polyA를 붙여 혼성유전자(chimaeric gene)를 만들고, 또한 이 작동역의 제한효소지도를 작성하여 5' 가 일부 삭제(deletion)된 여러 혼성유전자를 만들었다. 이들을 이원운반체(binary vector system)를 사용하여 담배에 형질전환시켜 완전히 재생된 식물체를 얻었으며, 이들 형질전환된 담배에서 이 작동역이 기능을 하는지 조사하기 위해 크로람펜콜의 ?C동위원소를 사용하여 CAT역가를 측정하였으나 이것의 역가는 나타나지 않았다. Nodulin-35(N-35) is a subunit of nodule specific uricase of soy bean(Glycine max), N-35 is preferentially synthesized on free polysomes in nodule tissue and transport to peroxisones of uninfected cells where it was directly localize. A chimaeric gene was constructed, containing the N-35 promoter, chloramphenicol acetyl transferase(CAT) and the termination region from N-23 gene. The different chimaeric genes were also constructed, which the entire 5' upstream sequences from -1100 to -50 was deleted. This gene was introduced into tobacco plants using the binary vector system. However, regenerated transformed plants showed no detectable CAT activity.

Tyrophagus putrescentiae (Tp) as a storage mite inhabitats in stored grains, hay, and straw at agricultural areas. T. putrescentiae stimulates an immune response and triggers inflammatory cytokines release, and thus it is a source of allergen that sensitize and induce allergic reactions. Also, T. putrescentiae has been reported to cause asthma and atopic disease by cross-reactivity with Dermatophagoides pteronyssinus (Dp). The study on T. putrescentiae in human monocytic THP-1 cells is not enough to understand cytokine expression and pathological mechanisms. The aim of this study is to investigate the effect of T. putrescentiae extract (TpE) on production of inflammatory cytokines and expression of mRNA level in THP-1 cells. THP-1 cells are treated with TpE and supernatants were analyzed for the production of cytokines using enzyme-linked immunosorbent assay (ELISA). mRNA level in the culture cells was measured by a reverse transcriptase-polymerase chain reaction (RT-PCR). As a result of this study, TpE significantly induced secretion of interleukin-6, interleukin-8, and monocyte chemotactic protein-1 (MCP-1) in THP-1 cells in time- and dose-dependent manner. These results suggest that TpE may play a role in contributing to inflammatory disease through stimulation of immune cell. Further research of T. putrescentiae is needed to understand the elucidation of the pathogenic mechanism.
Scavenging Effect of Bee Venom for Acua-acupuncture against Nitric Oxide

목적 : 본 연구는 최근 임상에서 많이 사용하는 봉독약침액의 nitric oxide (NO)에 대한 소거효과를 분석하기 위하여 실행되었다. 방법 : 양성대조군으로 비타민과 실험군으로 국산 봉독과 미국산 봉독에 NO를 분비하는 S-nitroso-N-acetylpenicillarnine (SNAP)을 투여한 후 NO의 농도를 540 nm 파장의 자외선 흡수량을 측정하여 평가하였다. 결과 : 국산 봉독은 100㎍/㎖에서 SNAP 처리 1.5, 3, 6 및 12시간에 대조군 (100%)과 비교하여 각각 86.7±1.5, 103.8±1.9, 88.3±2.9 및 85.6±3.2%의 잔류 NO 농도를 보였고, 1.5, 6 및 12시간에서 유의한 NO 소거 효과를 나타냈다. 미국산 봉독에서는 각각 74.0±2.2, 66.5±10.9, 86.1±2.8 및 59.1±3.8%로 모든 시간에서 유의한 결과를 보였다. 결론 : 이상의 봉독의 NO 소거효과는 봉독 약침액이 임상에서 동통, 종창, 등에 치료에 사용하는 작용기전의 하나로 사료된다.
