Identification of Glutathione Conjugates of 2, 3-Dibromopropene in Male ICR Mice

Hepatotoxic potential of 2, 3-dibromopropene (2, 3-DBPE) and its conjugation with glutathione (GSH) were investigated in male ICR mice. Treatment of mice with 20, 50, and 100 mg/kg of 2, 3-DBPE for 24 h caused elevation of serum alanine aminotransferase and aspartate aminotransferase activities. The hepatic content of GSH was not changed by 2, 3-DBPE. Meanwhile, the GSH content was slightly reduced when mice were treated with 2, 3-DBPE for 6 h and significantly increased 12 h after the treatment. Subsequently, a possible formation of GSH conjugate of 2, 3-DBPE was investigated in vivo. After the animals were treated orally with 20, 50, and 100 mg/kg of 2, 3-DBPE, the animals were subjected to necropsy 6, 12, and 24 h later. A conjugate of S-2-bromopropenyl GSH was identified in liver and serum treated with 100 mg/kg of 2, 3-DBPE by using liquid chromatography-electrospray ionization tandem mass spectrometry. The protonated molecular ions $[M+H]^+$ of S-2-bromopropenyl GSH were observed at m/z 425.9 and 428.1 in the positive ESI spectrum with a retention time of 6.35 and 6.39 min, respectively. In a time-course study in livers following an oral treatment of mice with 100 mg/kg of 2, 3-DBPE for 6, 12, and 24 h, the 2, 3-DBPE GSH conjugate was detected maximally 6 h after the treatment. The present results suggested that 2, 3-DBPE-induced hepatotoxicity might be related with the production of its GSH conjugate.

Background: Acne is a inflammatory skin disorders caused by inflammatory biomarkers. Magnesium ascorbyl phosphate (MAP) is a stable precursor of vitamin C. It achieves a constant delivery of vitamin C into the skin and has antioxidative effects. Objectives: To evaluate changes in the expression of inflammatory biomarkers after treatment of cultured sebocytes with MAP Methods: RT-PCR, ELISA and western blotting were performed for inflammatory cytokines, matrix metalloproteinases (MMPs), antimicrobial peptides (AMPs) and keratinocyte differentiation markers before and after treatment of cultured sebocytes with MAP (10-2 mol), lipopolysaccharide (LPS) and a combination of MAP and LPS. RT-PCR and western blotting were also done for Toll-like receptor-4 (TLR-4) and NF-ĸB before and after treatment of cultured sebocytes with MAP, LPS and a combination of MAP and LPS. Results: The expressions of inflammatory cytokines (e.g., IL-1 β and IL-8) and MMPs (e.g., MMP-9) were decreased after treatment with MAP and a combination of MAP and LPS, and increased after treatment with LPS. AMPs (e.g., LL-37) were decreased after treatment with MAP and a combination of MAP and LPS, and increased after treatment with LPS. The expression of TLR-4 and NF-ĸB was decreased after treatment with MAP and a combination of MAP and LPS, and increased after treatment with LPS. Conclusion: MAP, which is a stable precursor of vitamin C, may be a good effective and alternative agent to improve inflammatory reaction in acne.

인체의 혈액생화학 성분은 생리적인 상태나, 유전적 소인, 질병이나 기타 여러 가지 환경적 소인에 따라 달라진다. 따라서 임상에서는 참조치를 설정할 때에는 성별, 연령별 또는 인종별 등에 의해 분류하기를 원하고 있다. Glycated hemoglobin은 당뇨병의 진단, 치료관리에 이미 측정되어 사용된지가 오래이다. 또한 근년에 glycated serum protein 측정법이 개발 보급되어 또다른 장접을 갖는 방법으로서 각광을 받고 있다. 저자들은 참조치로서의 신빙성을 갖을 만한 일부 건강한 한국인들의 후락토사민 농도를 새로운 방법으로 정도관리 상태가 양호한 가운데 측정하였다. 그리고 그 결과를 여러 가지 통계학적인 방법으로 분석하여 유용한 참조치로서의 최적화를 시도하여 보았다. The chemical components of the human body are subjected to vary by physiological processes, genetic differences, diseases and various environmental factors. Therefore, separate reference values of the chemincal components for sex, age groups and other criteria are needed. Glycated hemoglobin is generally measured for the assessment of glycemic control in diabetes mellitus. Recently, however, the method for the measurement of glycated serum protein has drawn increasing attention. This study was udertaken to establish reference ranges for concentration of fructosamine among some healthy Korean and to compare this result with other reports.

Background: Incidence of cutaneous malignant tumor is increasing recently and the changes in pattern may be suggested. Nevertheless, there has been little research into the distribution, incidence of cutaneous malignant tumors in Daegu and Kyungpook Province. Objectives: To analyze changes of during recent two decade in incidence, clinical patterns of cutaneous malignant tumors in Kyungpook national university hospital(KNUH). Methods: We reviewed 1,527 cases of cutaneous malignant tumors from 1994 to 2013 at the Department of Dermatology in KNUH. We also devided subgroup into first (1994-2003) and second decade (2004-2014) to see changed over the years. Results: The average incidence of cutaneous malignant tumors among the total number of outpatients was 1.57% through whole study period. The incidence of cutaneous malignant tumors in the first half of the study period was 0.37% and in the second half was 2.05%. The highest average incidence was basal cell carcinoma (BCC) (first vs. second decade: 0.15% vs. 0.66%), followed by squamous cell carcinoma (SCC) (0.11% vs. 0.49%) and malignant melanoma (MM) (0.05% vs. 0.23). The most common site was the face in each decade. The overall recurrence was diagnosed in 55 of the 1,527 patients (3.60%). In each disease, the recurrence rate was 1.79% vs. 0.58% (BCC), 4.17% vs. 0.81% (SCC), and 0.60% vs. 1.47% (MM). Conclusion: This epidemiological study provided useful information for the distribution, incidence and prognosis of skin tumors.
Quantitative Assay for the Binding of Jun-Fos Dimer and Activator Protein-1 Site

The Jun and Fos families of eukaryotic transcription factors form heterodimers capable of binding to their cognate DNA enhancer elements. We are interested in searching for inhibitors or antagonists of the binding of the Jun-Fos heterodimer to the activator protein-1 (AP-1) site. The basic-region leucine zipper (bZIP) domain of c-Fos was expressed as a fusion protein with glutathione S-transferase, and allowed to form a heterodimer with the bZIP domain of c-Jun. The heterodimer was bound to glutathione-agarose, to which were added radiolabeled AP-1 nucleotides. After thorough washing, the gel-bound radioactivity was counted. The assay is faster than the coventional electrophoretic mobility shift assay because the gel electrophoresis step and the autoradiography step are eliminated. Moreover, the assay is very sensitive, allowing the detection of picomolar quantities of nucleotides, and is not affected by up to 50% dimethylsulfoxide, a solvent for hydrophobic inhibitors. Curcumin and dihydroguaiaretic acid, recently known inhibitors of Jun-Fos-DNA complex formation, were applied to this Jun-GST-fused Fos system and revealed to decrease the dimer-DNA binding.
