The gas chromatographic behavior fo three antkhistamines Chloropheniramine, Diphenhydramine , Tripelennamine and their salts were investigated using alkaline precolumn packed with 20% KOH on 60-80 mesh activated alumina. Symmetrical peaks were achieved under the condition, inlet temperature 260℃, cooumn temerature 180℃, carrier gas flow rate 30㎖/min. The peaks of antihistamine salts coincided with those of bases in alcohol solution. Chromatographic peaks of many drugs of organic bases, vitamines and antipyretics were Substracted by the precolumn.

The methanol extract of Nelumbo seed (Nelumbo nucifera GAERTN) was treated with 10% alcoholic KOH solution and three components were isolated from the unsaponificated matters. The sterols were identified as ergosterol and β-Sitosterol by T.L.C., G.L.C. and chemical tests.

The carpophores of Hirshipporus fusco-violaceus(Schard.ex Fr.) which grows widly in Korea were collected in the Gyeong Gi Province and extracted with chloroform and methanol. Two compounds were isolated from the extract and one of these compounds were identified as ergosterol by T.L.C., G.L.C. and chemical tests.

A method for the rapid determination of paeonol in Moutan Root-bark produced in Korea was established by high speed liquid chromatography. Paeonol was separated in a 8㎜ i.d. ×115㎜ column of μ-Bondapak C??, using a Waters associate Model 244 liquid chormatograph, with CH₃OH:H₂O:CH₃COOH(50:50:1) as the desorption solution. Separation was completed within 9min. Paeonol in Moutan Root-bark was extracted with methanol and the extract was injected directly into the column. Paeonol content was calculate from previously prepared calibration curves based on standard paeonol. At 250nm, the precision of the determination was about ±1.2%. A rapid method for determing p-methoxycinnamic acid(p-MCA) in Scrophulariae Radix(Scrophularia Oldhami Oliver) produced in Korea was established by high speed liquid chromatography. p-MCA was separated in a 8mm i.d×115mm column of μ-Bondapak C??, using a waters associate Model 244, with CH₃CN:H₂O:CH₃COOH(40:60:1) as the desorption solution, Separation was completed within 6min. p-MCA in Scrophulariae Radix was extracted with water and the extract solution was injected directly into the column. p-MCA content was estimated from previously prepared calibration curves based on standard p-MCA. At 310nm, the precision of the determination was about ±1.5%. <P>Mutations in the coactivator CREB-binding protein (CBP) are a major cause of the human skeletal dysplasia Rubinstein-Taybi syndrome (RTS); however, the mechanism by which these mutations affect skeletal mineralization and patterning is unknown. Here, we report the identification of 3-phosphoinositide-dependent kinase 1 (PDK1) as a key regulator of CBP activity and demonstrate that its functions map to both osteoprogenitor cells and mature osteoblasts. In osteoblasts, PDK1 activated the CREB/CBP complex, which in turn controlled runt-related transcription factor 2 (RUNX2) activation and expression of bone morphogenetic protein 2 (BMP2). These pathways also operated in vivo, as evidenced by recapitulation of RTS spectrum phenotypes with osteoblast-specific Pdk1 deletion in mice (Pdk1osx mice) and by the genetic interactions observed in mice heterozygous for both osteoblast-specific Pdk1 deletion and either Runx2 or Creb deletion. Finally, treatment of Pdk1osx and Cbp+/- embryos with BMPs in utero partially reversed their skeletal anomalies at birth. These findings illustrate the in vivo function of the PDK1-AKT-CREB/CBP pathway in bone formation and provide proof of principle for in utero growth factor supplementation as a potential therapy for skeletal dysplasias.</P>
