        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1. Our results suggest that DN is an effective inducer of HO-1 in the pancreas, and that HO-1 regulates neutrophil infiltration in AP via CXCL2 inhibition.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Desoxo-narchinol-A (DN) is a natural compound of HO-1 inducer in pancreas. </LI> <LI> Mechanism of DN-induced HO-1 is mediated by MAPK/Activator Protein-1/HO-1 signaling. </LI> <LI> DN-induced HO-1 blocks neutrophil infiltration into pancreas via inhibition of CXCL2. </LI> <LI> DN inhibits cerulein-induced acute pancreatitis (AP) and AP-associated lung injury. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        사례보고 : 하수오 복용 후 발생한 재발성 독성 간염 1예

        Toxic hepatitis has been reported as a major cause of acute hepatitis, but its potential induction by herbal remedies and/or health foods is usually neglected. We experienced a case of toxic hepatitis associated with Polygoni multiflori, a Chinese herb commonly known as Ho-Shou-Wu. A 54-year-old woman consumed Ho-Shou-Wu for 1 month, after which she experienced fatigue and overall weakness. A diagnosis of toxic hepatitis was made based on her clinical history, the findings for viral markers and other laboratory data, and ultrasonography. Her condition improved considerably after she stopped taking Ho-Shou-Wu. However, she resumed taking Ho-Shou-Wu immediately after discharge from hospital, which aggravated her symptoms and liver function. She was immediately readmitted and stopped taking Ho-Shou-Wu. Her relapse into hepatitis immediate after resuming consumption of the herb is strongly indicative of the validity of Koch`s postulate in this case.

        Protective Effect of Heme Oxygenase-1 on High Glucose-Induced Pancreatic β-Cell Injury

        <P><B>Background</B></P><P>Glucose toxicity that is caused by chronic exposure to a high glucose concentration leads to islet dysfunction and induces apoptosis in pancreatic β-cells. Heme oxygenase-1 (HO-1) has been identified as an anti-apoptotic and cytoprotective gene. The purpose of this study is to investigate whether HO-1 up-regulation when using metalloprotophyrin (cobalt protoporphyrin, CoPP) could protect pancreatic β-cells from high glucose-induced apoptosis.</P><P><B>Methods</B></P><P>Reverse transcription-polymerase chain reaction was performed to analyze the CoPP-induced mRNA expression of HO-1. Cell viability of INS-1 cells cultured in the presence of CoPP was examined by acridine orange/propidium iodide staining. The generation of intracellular reactive oxygen species (ROS) was measured using flow cytometry. Glucose stimulated insulin secretion (GSIS) was determined following incubation with CoPP in different glucose concentrations.</P><P><B>Results</B></P><P>CoPP increased HO-1 mRNA expression in both a dose- and time-dependent manner. Overexpression of HO-1 inhibited caspase-3, and the number of dead cells in the presence of CoPP was significantly decreased when exposed to high glucose conditions (HG). CoPP also decreased the generation of intracellular ROS by 50% during 72 hours of culture with HG. However, decreased GSIS was not recovered even in the presence of CoPP.</P><P><B>Conclusion</B></P><P>Our data suggest that CoPP-induced HO-1 up-regulation results in protection from high glucose-induced apoptosis in INS-1 cells; however, glucose stimulated insulin secretion is not restored.</P>

        Load distribution in group piles is an important matter of consequence for the safe design and the economical design. In the group piles, the model tests were to investigate the effects on spacing-to-diameter ratio of pile, number of piles and arrangement of the pile. The tops of pile were all set to rotate freely. This paper considered the load distribution by the location of the pile as compared the behavior of group piles with single pile. For a three row pile group at 3D spacing, the measured individual row distributions were 35~38%(lead), 32∼34%(middle) and 27∼30%(tail). This results were compared very favorably to those by McVay et al(1995). The shadowing effect for the parallel direction of lateral loading appears to be more significant than the one for the perpendicular direction of lateral loading.

      • Listeria spp.의 분리와 특성에 관한 연구

        냉동상태로 유통되고 있는 식육점 우육과 수입육전문점 우육 및 도축장 유래 생우육에서 Listeriosis spp.를 분리하여 시료에 따른 균종별 검출율과 생화학적 성상을 확인하고 특히 listeriosis를 유발하는 L. monocytogenes에 대해서는 serotype과 항생제 감수성 시험을 실시하여 아래와 같은 결과를 얻었다. 1. 분리된 46주의 Listeria spp.는 모두 Catalase, Motility 양성이었다. 그리고, β -Hemolysis, CAMP test, 생화학시험 결과, 4균주는 L. monocytogenes, 40균주는 L. innocua로, 그리고 2균주는 L. welshimeri였으며, 시료 60건 中 46시료에서 Listeria spp.가 분리되어 68.3%의 검출율을 나타내었다. 2. Listeria spp.의 균종별 분리율은 L. innocua(8.7%), L. monocytogenes(86.95%), L. welishmeri(4.3%)의 순으로 나타났다. 시료에 따른 Listeria spp.의 균종별 분포는 수입 우육 25건 중 L. monocytogenes가 1주(4%), L. innocua가 13주(52%)로 분리되었고, 시판되고 있는 한우 25건 중 L. monocytogenes가 3주(12%), L. innocua가 20주(80%)분리되었고, 도축장 한우 10건 중 L. innocua가 7주(70%), L. welishmeri가 2주(20%)분리되었다. 3. 분리된 L. monocytogenes 4균주 모두 Stapylococcus aureus ATCC 25923와 Rhodococcus equi KCTC 9082에 양성반응을 나타내었고, L. monocytogenes 로 분리된 4균주의 혈청형은 serotype 1이 1주, Serotype 4가 3주로 나타났으나, 정확한 serovars는 알 수 없었다. 4. 분리된 L. monocytogenes 4주에 대한 항생제 감수성 시험에서 전 균주가 Ampicillin(AM), Cephalothin(CF), Chloramphenicol(C), Erythromycin(E), Gentamycin(GM), Kanamycin(K), Methicillin(Dp), Vancomycin(VA)에 감수성을 나타냈고, Clindamycin(CC) Polymycin(PB)에 대해서는 내성을 나타냈으며, Streptomycin(S)은 L. monocytogenes ATCC 19111 균주와 L. monocytogenes serotype 1에는 감수성을, 4에는 중정도감수성을 나타내었으며, Tetracycline(Te)는 L. monocytogenes ATCC 19111 균주에는 중정도감수성을, L. monocytogenes serotype 1과 4에는 감수성을 나타내었다. Listeria species isolated from specimens are 46 strains which positive to catalase and molility. After β-Hemolysis, CAMP test and boichemical tests, 4 strains are L.monocytogenes, 40 strains are L. innocua and 2 strains are L. welshimeri. Isolation rate of Listeria spp. was 68.3%. Isolation rate of Listeria spp. on samples are L. monocytogenes, 40 strains are L. monocytogenes(86.95%), L. innocua(8.7%) and L. welshimeri(4.3%). 4 L. monocytogenes strains isolated are positive on Stapylococcus aureus ATCC 25923 and Rhodococcus equi KCTC 9082. Serotype of isolated strains were serotype 1 and serotype 4. Antibiotics sensitivity of all L. monocytogenes isolated are sensitivity to Ampicillin(AM), Cephalothin(CF), Chloramphenicol(C), Erythromycin(E). Gentamycin(GM), Kanamycin(K), Methicillin(Dp), Vancomycin(VA) and of those are resistant to Clindamycin(CC) and Polymycin(PB). Streptomycin(S) is sensitivity to L. monocytogenes ATCC 19111, L. monocytogenes serotype 1 and is intermediate sensitive to L. monocytogenes serotype 4. Tetracycline(Te) is intermediate sensitive to L. monocytogenes ATCC 19111 and is sensitivity to L. monocytogenes serotype 1 and 4.

        The purpose of this study, the local restaurants in gimcheon ceo and chef, traditional food to target residents identify overall against the recognition. This can represent an area based on regional food ways that can be commercialized. To this end the following measures are presented. First, gimcheon should enable the local restaurants of gimcheon ways in. The most important element of the restaurant just because the food tastes of tourists coming to the area a special flavor with local restaurants, and local attractions that can represent the area restaurants. Second, the grapes and plum Festival, gimcheon related practice enabled the food. Many people are new to the topic of food. Food festival, cultural experience and to provide a place for locals to be able to inform the food culture festival. Third, representing agricultural and speciality by developing water-assisted traditional processed food merchandising. Gimcheon is being produced in a prime agricultural and specialities and distinctive flavors and seasonings in the sauce can do the products sales outlook is very bright with a judgment. Fourth, the flavor are willing provide food through the map and tourist information. On a map, the taste itself makes cooperating and of food-related at the level of the Association can represent the different forms and areas of gimcheon theme will yield vigorously. These effects in gimcheon of tourists and local economic development and tourist visits are expected to have increased.

      • FGR 환경설비 시스템 보일러의 성능 및 배기 배출물에 미치는 재순환 배기의 영향

        The effect of recirculated exhaust gas on performance and exhaust emissions with FGR rate are investigated by using a natural circulation, pressurized draft and water tube boiler with FGR system operating at several boiler loads and over fire air(OFA) damper openings. The purpose of this study is to apply the FGR system to a power plant boiler for reducing NO_(x) emissions. To activate the combustion, the suction damper of two stage combustion system installed in the upper side of wind box is opened by handling the lever between 0˚ and 90˚, and the OFA with 0 to 20% into the flame is supplied, as the combustion air supplied to burner is reduced. It is found that the fuel consumption rate divided by evaporation rate does not show an obvious tendency to increase or decrease with rising the FGR rate, and NO_(x) emissions are decreased, at the same OFA damper opening, as FGR rates are elevated and boiler loads are dropped. While a small quantity of soot is emitted without the operational conditions of boiler load, OFA damper opening and FGR rate, because soot emissions are eliminated by the electrostatic precipitator with a collecting efficiency of 86.7%.
