Clinical molecular subtyping reveals intrinsic mesenchymal reprogramming in gastric cancer cells

The mesenchymal cancer phenotype is known to be clinically related to treatment resistance and a poor prognosis. We identified gene signature-based molecular subtypes of gastric cancer (GC, n = 547) based on transcriptome data and validated their prognostic and predictive utility in multiple external cohorts. We subsequently examined their associations with tumor microenvironment (TME) features by employing cellular deconvolution methods and sequencing isolated GC populations. We further performed spatial transcriptomics analysis and immunohistochemistry, demonstrating the presence of GC cells in a partial epithelial-mesenchymal transition state. We performed network and pharmacogenomic database analyses to identify TGF-β signaling as a driver pathway and, thus, a therapeutic target. We further validated its expression in tumor cells in preclinical models and a single-cell dataset. Finally, we demonstrated that inhibition of TGF-β signaling negated mesenchymal/stem-like behavior and therapy resistance in GC cell lines and mouse xenograft models. In summary, we show that the mesenchymal GC phenotype could be driven by epithelial cancer cell-intrinsic TGF-β signaling and propose therapeutic strategies based on targeting the tumor-intrinsic mesenchymal reprogramming of medically intractable GC.

Antithrombin (AT) plays a pivotal role as the primary inhibitor of blood coagulation proteases. Hereditary AT deficiency, predominantly attributed to serpin family C member 1 (SERPINC1) gene mutations, is an autosomal-dominant thrombophilic disorder associated with an elevated risk of thrombotic diseases. Due to this mode of inheritence, most cases are reported to be cased by heterozygous mutations, we report a unique case of congenital AT deficiency with a homozygous SERPINC1 that resulted in neonatal cardiac thrombosis. This case underscores the complexity and varied clinical presentations of AT deficiency, emphasizing the importance of early diagnosis and intervention in congenital thrombophilic conditions.

The goal of the current study is to assess the antitumor mechanism by combination of Angelica gigas and Torilis japonica Ethanol Extracts (AT) in prostate cancers. Here, AT reduced the viability and the number of colonies in LNCaP prostate cancer cells. Furthermore, AT induced G1 phase arrest and decreased the protein expression of Cyclin D1, Cyclin E, cdk2 and increased p27 in LNCaP cells. Also, AT significantly decreased expression of PSA level and AR at mRNA and protein levels in LNCaP cells. Interestingly, AT attenuated the expression of AR, PSA and Wnt-3a and also reduced AR and PSA stability in LNCaP cells. Conversely, dihydrotestosterone (DHT) reversed the ability of AT to reduce AR and PSA, since AR and PSA were upregulated by DHT in a time-dependent manner. Notably, AT disrupted the protein-protein interaction as well as reduced nuclear translocation and fluorescent expression of β-catenin and AR in LNCaP cells. Taken together, our findings suggest that combination of Angelica gigas and Torilis japonica suppresses proliferation of LNCaP cells via G1 arrest and inhibition of β-catenin and androgen receptor as a potent anticancer agent.
Building Calibration Curve for Py-GC/MS Analysis of SBR/BR Blend Rubber Compounds

A calibration curve is needed to determine the SBR and BR blend ratio of SBR/BR blend rubber compounds using pyrolysis-gas chromatography/mass chromatography (Py-GC/MS) or Py-GC. In general, a calibration curve is obtained using reference SBR/BR vulcanizates with various blend ratios. In this study, the calibration curves were obtained using reference samples made of rubber solutions and were compared to those plotted using the reference SBR/BR vulcanizates. Calibration curves using variations of 1,3-butadiene/styrene, 4-vinylcyclohexene (VCH)/styrene, 2-phenylpropene (PhP)/butadiene, PhP/VCH, 4-phenylcyclohexene (PhCH)/butadiene, and PhCH/VCH ratios with the BR content were examined for the suitability. We found that the calibration curves obtained using the mixed rubber solution references (1,3- butadiene/styrene and PhP/butadiene) could replace those constructed using the reference SBR/BR vulcanizates. The calibration curves of 1,3-butadiene/styrene and PhP/butadiene obtained using the raw references can be used for the determination of the SBR/BR blend ratios by applying some correction factors.

Wear particles of the model tread compounds for bus and truck tires were made using a laboratory abrasion tester and characterized based on their size distributions, shapes, and crosslink densities. The influence of the carbon black contents and rubber compositions (NR= 100 and NR/BR= 80/20) on the production of wear particles was investigated. The wear particles were separated according to size using a sieve shaker. The shape properties of the wear particles were analyzed using an image analyzer and scanning electron microscopy (SEM). Their shapes were observed as tiny stick cookies or sausages with bumpy surfaces. The particle size distribution tended to be smaller with increasing carbon black content. Moreover, the particle size distributions of the NR = 100 samples were larger than that of the NR/BR blend samples. There were different filaments in the wear particles. The filament diameters tended to be thinner with increasing carbon black content. The crosslink density increased with increasing carbon black content, and the crosslink densities of the NR= 100 samples were lower than those of the NR/BR blend ones. The particle size distribution tended to be smaller with increasing crosslink density. Based on the experimental results, the wear particles can be produced by detaching debris from the main body through repetitive strain and recovery.

Abrasion tests of an SBR compound were conducted using four different types of abrasion testers (cut and chip, Lambourn, DIN, and LAT100). The abrasion test results were analyzed in terms of size distributions and morphologies of the wear particles. Most wear particles were larger than 1000 μm. The wear particle size distributions tended to decrease as the particle size decreased. Except for the Lambourn abrasion test, the wear particles smaller than 212 μm were rarely generated by the other three abrasion tests, implying that small wear particles were produced through friction by introducing talc powder. Shapes of the wear particles varied depending on the abrasion testers. The wear particles generated from the Lambourn abrasion tester had stick-like shapes. The cut and chip abrasion test showed a clear abrasion pattern, but the DIN abrasion test did not show any specific abrasion pattern. The Lambourn and LAT100 abrasion tests showed irregular abrasion patterns.
The Neural Correlates of Positive Versus Negative Thought-action Fusion in Healthy Young Adults

Objective: Thought-action fusion (TAF), one of the most-studied dysfunctional beliefs in obsessive-compulsive disorder, represents an individual’s belief that his/her thoughts directly influence events. TAF belief types are divided into personal thoughts relating to positive (positive TAF) and negative outcomes (negative TAF). However, the neural mechanisms underlying both aspects of the TAF response remain elusive. Methods: This functional magnetic resonance imaging study aimed to investigate the neural circuits related to positive and negative TAF and their relationships with psychological measures. Thirty-one healthy male volunteers participated in a modified TAF task wherein they were asked to read the name of a close person embedded in positive statements (PS) or negative statements (NS). Results: Conjunction analysis revealed activation of the fusiform and lingual gyri, midcingulate and superior medial frontal gyri, inferior orbitofrontal gyrus, and temporoparietal junction. The NS > PS comparison showed additional activation in the precuneus and medial prefrontal cortex, superior frontal gyrus, insula, globus pallidus, thalamus, and midbrain. Precuneus activity was associated with the TAF score among these areas. Moreover, activity in the inferior orbitofrontal gyrus, insula, superior, middle and medial frontal gyri, globus pallidus, inferior parietal lobule, and precuneus was associated with dimensional obsessive-compulsive scores. In contrast, the PS > NS comparison revealed no significant activation. Conclusion: These results suggest that negative TAF, relative to positive TAF, recruits additional regions for self-referential processing, salience, and habitual responding, which may contribute to the activation of the belief that a negative thought increases the probability of that negative outcome.

Objective: Recent studies highlighted the triple-network model which illustrated the interactions among three large-scale networks including salience network (SN). The functional magnetic resonance imaging used in this study was designed to investigate the characteristics of three large-scale networks associated with the thought-action fusion (TAF) in patients with obsessive-compulsive disorder (OCD) using power spectral density (PSD) analysis. Methods: This study included 32 OCD patients and 38 age-matched healthy controls (HC). The TAF task was modified from the experiment of Rassin. PSD from time courses in large-scale networks of each subject was measured to compare between the groups for both TAF and resting state. Results: In SN, OCD reported lower power in the low-frequency domain of SN compared to HC using the two-sample t test during the TAF task (t = −2.395, p = 0.019) but not in the resting state. The PSD in the low-frequency domain of the SN had a significant negative correlation with state score in the guilty inventory (r = −0.361, p = 0.042) in OCD patients. Conclusion: This study suggests that OCD patients showed reduced SN power which can be prominent in a certain situation, such as TAF. In addition, the PSD alterations in SN cause difficulty in processing ambiguous emotional cues in social situations, and the difficulty can be connected with a negative feeling (e.g., guilt).
Analysis of Peroxide Curing Agents in EVA Compounds and Vulcanizates

Poly(ethylene-co-vinyl acetate) compounds and vulcanizates containing dicumyl peroxide (DCP) (HD) were prepared, and the curing agent and reaction products were analyzed using gas chromatography/mass spectrometry (GC/MS). Samples containing trially cyanurate (TAC) and DCP (HDT) were also prepared and analyzed. Some raw DCP were decomposed in the injector region of GC to produce acetophenone. DCP was detected in the HD compound but was not observed in the HD vulcanizate, and instead acetophenone and 2-phenyl-2-propanol were detected. Both DCP and TAC were observed in the HDT compound but not in the HDT vulcanizate, where acetophenone and 2-phenyl-2-propanol were detected. Thus, some of the DCP decomposed during the compounding process. The formation mechanism for acetophenone and 2-phenyl-2-propanol during the crosslinking reaction was identified, and differences in the crosslinking reactions of HD and HDT compounds were discussed.
