ZNF509S1 downregulates PUMA by inhibiting p53K382 acetylation and p53-DNA binding

Expression of the POK family protein ZNF509L, and -its S1 isoform, is induced by p53 upon exposure to genotoxic stress. Due to alternative splicing of the ZNF509 primary transcript, ZNF509S1 lacks the 6 zinc-fingers and C-terminus of ZNF509L, resulting in only one zinc-finger. ZNF509L and -S1 inhibit cell proliferation by activating p21/CDKN1A and RB transcription, respectively. When cells are exposed to severe DNA damage, p53 activates PUMA (p53-upregulated modulator of apoptosis) transcription. Interestingly, apoptosis due to transcriptional activation of PUMA by p53 is attenuated by ZNF509S1. Thus we investigated the molecular mechanism(s) underlying the transcriptional attenuation and anti-apoptotic effects of ZNF509S1. We show that ZNF509S1 modulation of p53 activity is important in PUMA gene transcription by modulating post-translational modification of p53 by p300. ZNF509S1 directly interacts with p53 and inhibits p300-mediated acetylation of p53 lysine K382, with deacetylation of p53 K382 leading to decreased DNA binding at the p53 response element 1 of the PUMA promoter. ZNF509S1 may play a role not only in cell cycle arrest, by activating RB expression, but also in rescuing cells from apoptotic death by repressing PUMA expression in cells exposed to severe DNA damage.

Five hundred and forty crossbred (Korean native black pig${\times}$Landrace) F2 were selected at a commercial pig farm and then divided into six different coat color groups: (A: Black, B: White, C: Red, D: White spot in black, E: Black spot in white, F: Black spot in red). Birth weight, 21st d weight, 140th d weight and carcass weight varied among the different coat color groups. D group (white spot in black coat) showed a significantly higher body weight at each weigh (birth weight, 140th d weight and carcass weight) than did the other groups, whereas the C group (red coat color) showed a significantly lower body weight at finishing stage (140th d weight and carcass weight) compared to other groups. Meat quality characteristics, shear force, cooking loss and meat color were not significantly different among the different coat color groups, whereas drip loss was significantly higher in F than in other groups. Most blood characteristics were not significantly different among the different groups, except for the red blood cells.

O-ethyl S-methyl ethylphosphonothioate [LD_(50) (rat, oral) 4.6㎎/㎏ : K_i(bovine erythrocyte acetylcholinesterase) 303 M^(-1min-1)] was selected as a model compound to study the mode of action of O, S-dialkyl alkylphosphonothioates which have been hypothesized to be toxic via a bioactivation process. Two chemical oxidants, meta-chloroperoxybenzoic acid and monoperoxyphthalic acid, and rat liver microsomal oxidases were used to mimic the action of mixed function oxidases on the model compound. The formation of S-oxide, a very unstable active intermediate, was proposed based on the identification of metabolic products. Furthermore, a trapping experiment with ethanol showed that the unstable intermediate S-oxide had the ability to phosphorylate acetylcholinesterase which is an important enzyme in nerve systems. The S-oxide intermediates are presumed to be responsible for the toxicity of O,S-dialkyl alkylphosphonothioates.
RPS27a enhances EBV-encoded LMP1-mediated proliferation and invasion by stabilizing of LMP1

Epstein-Barr virus (EBV)-encoded latent membrane protein 1 (LMP1) is an oncoviral protein that plays a pivotal role in EBV-induced oncogenic transformation. The function of LMP1 in EBV-induced oncogenesis has been well studied. However, the molecular mechanisms underlying LMP1 protein stability remain poorly understood. In this study, we found that ribosomal protein s27a (RPS27a) regulates LMP1 stability by a tandem affinity purification analysis. RPS27a interacts directly with LMP1 in vitro and in vivo. Furthermore, overexpression of RPS27a increases the half-life of LMP1 in 293T cells, whereas downregulation of RPS27a using lentiviral shRNA technology accelerates the decrease in LMP1 protein level in EBV-transformed B cells. We show that LMP1 ubiquitination via the proteasome is completely inhibited by overexpression of RPS27a. RPS27a also enhances LMP1-mediated proliferation and invasion, suggesting that RPS27a interacts with LMP1 and stabilizes it by suppressing proteasome-mediated ubiquitination. These results suggest that RSP27a could be a potential target in EBV-infected LMP1-positive cancer cells.

시중에서 5종의 생 막걸리를 구입하여 이들의 안지오텐신 전환효소 저해 활성을 측정한 후, 막걸리에 들어 있는 유산균의 수를 확인하였다. 순수배양된 유산균들을 16S rRNA 유전자를 증폭하여 염기서열을 결정함으로써 동정하였다. 그 결과 5종의 제품들 중 Da와 Ku 막걸리가 안지오텐신 전환효소 저해 활성이 타 제품들보다 우수하였다. 이들 제품에서 Uncultured Lactobacillus sp., Lactobacillus sp., Lactobacillus paracasei subsp. tolerans, Lactobacillus crustorum 등의 유산균이 동정되었으며, 다른 제품과는 다른 유산균총을 갖고 있음을 확인하였다. The goal of this study was to isolate lactic acid bacteria which has superior functionalities from commercially available raw Makgeolli. To accomplish this, we measured the angiotensin converting enzyme(ACE) inhibitory activity in 5 different brands of Makgeolli obtained from market. Also we isolated pure cultures from Makgeolli and identified them. Makgeolli brands called Da and Ku showed a superior ACE inhibitory activity and had a different microbial flora with others.

In this study, we have developed the static and dynamic simulation program of a high speed current collection system. The catenary wire is modeled to discrete masses connected by massless strings and the pantograph is replaced with 3 d.o.f equivalent models that are composed of masses, springs and dampers. We derived partial differential equations of motion from the equivalent model and developed the simulation program. Then, we calculated the static equilibrium state of the overhead catenary and the dynamic behaviors of the high speed current collection system. The analysis results were compared with the results of GASENDO software developed at RTRI in Japan.

light rail vehicle is modeled as a 2 d. o. f linear system for the design of vertical suspension characteristics and a 4 d.o. f linear system for the design of lateral suspension characteristics. FRA"s class-5-track irregularity is used for the exciting disturbance on track. Suspension stiffness and damping is selected on the basis of the ride quality and suspension stroke trade-off for the bogie of light rail vehicle. The optimum value of primary and secondary suspension characteristics is determined. And the stability of full vehicle model for the LRV is analyzed using the VAMPIRE program and critical speed is determined.

In this study, we developed the simulation program of an overhead catenary-pantograph system. The overhead catenary is modeled with point mass and the pantograph is replaced wi th 3 d.o.f model which is composed of mass, spring and damper. Using the developed program, we analyzed the static structure of the overhead catenary and the dynamic characteristics of an overhead catenary-pantograph system such as uplift displacement of contact wire and contact force. we compared the analysis resul ts wi th the results of GASENDO software developed at RTRI in Japan. The behaviors of uplift displacement of contact wire and contact force were similar with the results of GASENDO software.
